In 2011, following severe flooding in Eastern Australia, an unprecedented epidemic of equine encephalitis occurred in South-Eastern Australia, caused by Murray Valley encephalitis virus (MVEV) and a new variant strain of Kunjin virus, a subtype of West Nile virus (WNVKUN). This prompted us to assess whether a delta inulin-adjuvanted, inactivated cell culture-derived Japanese encephalitis virus (JEV) vaccine (JE-ADVAX™) could be used in horses, including pregnant mares and foals, to not only induce immunity to JEV, but also elicit cross-protective antibodies against MVEV and WNVKUN. Foals, 74–152 days old, received two injections of JE-ADVAX™. The vaccine was safe and well-tolerated and induced a strong JEV-neutralizing antibody response in all foals. MVEV and WNVKUN antibody cross-reactivity was seen in 33% and 42% of the immunized foals, respectively. JE-ADVAX™ was also safe and well-tolerated in pregnant mares and induced high JEV-neutralizing titers. The neutralizing activity was passively transferred to their foals via colostrum. Foals that acquired passive immunity to JEV via maternal antibodies then were immunized with JE-ADVAX™ at 36–83 days of age, showed evidence of maternal antibody interference with low peak antibody titers post-immunization when compared to immunized foals of JEV-naïve dams. Nevertheless, when given a single JE-ADVAX™ booster immunization as yearlings, these animals developed a rapid and robust JEV-neutralizing antibody response, indicating that they were successfully primed to JEV when immunized as foals, despite the presence of maternal antibodies. Overall, JE-ADVAX™ appears safe and well-tolerated in pregnant mares and young foals and induces protective levels of JEV neutralizing antibodies with partial cross-neutralization of MVEV and WNVKUN. 相似文献

Perilipins have been reported to limit the interaction of lipases with neutral lipids within the droplets, thereby regulating neutral lipid accumulation and utilization. This study aimed to identify the location and expression of PLIN1 and PLIN2 in porcine oocytes during maturation. Quantitative real‐time polymerase chain reaction (qRT‐PCR), immunostaining and Western blot methods were used to characterize the expression and distribution patterns of PLIN1 and PLIN2 in porcine oocytes. The results showed that PLIN1 was not detectable in porcine oocytes. PLIN2 and BODIPY 493/503‐detected neutral lipid droplets appeared identical distribution patterns and extensive colocalization in both GV and MII porcine oocytes. PLIN2 protein expression was higher in GV oocytes than that in MII oocytes (p < 0.05), although PLIN2 mRNA expression was similar in both groups. These findings suggested that PLIN2 was a major lipid droplet‐associated protein in porcine oocytes. 相似文献

The purpose of this study was to determine the diurnal composition and concentration of volatile fatty acids (VFA) and to determine VFA composition and concentration differences between stomach compartment 1 (C1) and caecum of alpacas fed grass and alfalfa hay. The study was divided into two experiments. In Experiment 1 (EXP 1), 10 male alpacas (3+ years old, 65 kg BW) were divided into two groups, housed in drylot pens, provided ad libitum water and fed alfalfa (AH) or grass hay (GH) for 30 days. The alpacas were slaughtered and the digestive tract collected, divided into sub‐tract sections, weighed and digesta sampled for pH, dry matter (DM) and NDF. Volatile fatty acid composition and concentration were determined on C1 and caecal material. Four adult male (3+ years old, 60 kg BW), C1 fistulated alpacas were housed in metabolism crates and divided into two forage groups for Experiment 2 (EXP 2). Alpacas were fed the forages as in EXP 1. Diurnal C1 VFA samples were drawn at 1, 3, 6, 9, 12, 18 and 24 h post‐feeding. There were no differences between forages for tract weight, C1 and caecum digesta DM or NDF. Differences were noted (p < 0.05) for pH between forages and sub‐tract site. Volatile fatty acids concentrations were different (p < 0.05) for forage and site, and total VFA was higher for AH than GH (110.6 and 79.1 mm ) and C1 than caecum (40.7 and 27.6 mm ). Proportion of VFA was significant (p < 0.05) for forage and site, C1 acetate highest for GH (84.8 vs. 74.0 mm ) and caecum acetate 83.7 and 76.2 mm for GH and AH respectively. These data demonstrate the level of VFA produced in C1 and the caecum of alpacas and the diurnal VFA patterns. Composition of VFA is similar to other ruminant species. 相似文献

Paddlefish are gaining increasing acceptance as an aquaculture species worldwide. Commercial trout feeds, containing high protein and lipid levels, are currently used in intensive culture; however, nutritional requirements of paddlefish are not currently known. A study was conducted examining the effects on growth, survival and fillet composition of juvenile paddlefish when fed commercial feeds differing in protein and lipid levels. Paddlefish larvae were first stocked in 14.0 m3 round tanks and fed trout starter feeds for 43 days until trained to accept a 1.6 mm pellet. Paddlefish juveniles of mean weight (±SE) 20±0.27 g were randomly stocked into six0.02 ha ponds at 12 500 ha?1 and fed floating commercial trout or catfish (lower protein and lipid) feeds, twice daily (08:00 and 15:30 hours) for 92–97 days. At harvest, there were no significant differences in final weight, percent survival, specific growth rate , relative growth and feed conversion ratio between treatments, which averaged 223.6 g, 96.2%, 2.5% day?1, 10.2 and 1.98 respectively. Surface feeding activity index was significantly higher in ponds supplied with catfish feed than in ponds supplied with trout feeds. Relative pellet buoyancy was not a factor in feeding activity. Fulton's condition factor averaged0.238, was not significantly different, and was similar to a reported value for extensively cultured paddlefish (zooplanktivore). There was no significant difference in liver somatic index between treatments, which averaged 1.91%. Percent protein and moisture of fillets averaged 14.9% and 80.9%, respectively, and were not significantly different between treatments. However, lipid content of fillets was significantly higher in paddlefish fed the trout feed (4.45%), compared with paddlefish fed the catfish feed (2.42%). Fillet lipid content for both treatments was higher than reported values for extensively cultured paddlefish. Percent abdominal fat was significantly higher (0.82%) in paddlefish fed the trout feed compared with paddlefish fed the catfish feed (0.52%). Results from this study indicate that paddlefish can be fed a commercial catfish feed labeled to contain 32% protein and 4.5% lipid without adverse effects on growth, survival and fillet composition, lowering production costs. 相似文献

Worldwide, most sea‐urchin populations are in decline. Future market demands will likely be met through aquaculture, which may consist of gonad enhancement of wild‐caught sea urchins. In this context, we examined three land‐based containment systems for suitability in maintaining commercial‐size green sea urchins (Strongylocentrotus droebachiensis). Mortality rate, gonad quantity, gonad quality (colour, brightness, firmness, texture) and cleaning efficiency associated with each containment system were the criteria for comparing the effectiveness of the systems (large raceways, small raceways and washtub tanks). After 6 weeks, urchins maintained in large raceways displayed significantly higher mortality rates than urchins maintained in either the small raceways or washtub tanks (mean±SD: 24.3±10.4%, 8.0±3.5% and 4.3±1.5% respectively). Significant differences in cleaning time were detected among the three systems with washtub tanks requiring significantly more cleaning time (0.11±0.02 min urchin?1) than the other two systems and large raceways requiring significantly less cleaning time (0.06±0.02 min urchin?1) than the other two systems. No significant differences in gonad characteristics were detected among the three containment systems. Because of the higher mortality rate in the large raceways and the increased cleaning time required for the washtub tanks, the small raceways were considered to be the best of the three systems tested. 相似文献

Apple scab, caused by Venturia inaequalis, can lead to large losses of marketable fruit if left uncontrolled. The disease appears in orchards during spring as lesions on leaves. These primary lesions are caused by spores released at bud burst from overwintering sources; these spores can be sexually produced ascospores from the leaf litter or asexual conidia from mycelium in wood scab or within buds. The relative importance of conidia and ascospores as primary inoculum were investigated in an orchard in southeast England, UK. Potted trees not previously exposed to apple scab were placed next to (c. 1 m) orchard trees to trap air‐dispersed ascospores. Number and position of scab lesions were assessed on the leaves of shoots from both the potted trees (infection by airborne ascospores) and neighbouring orchard trees (infection by both ascospores and splash‐dispersed, overwintered conidia). The distribution and population similarity of scab lesions were compared in the two tree types by molecular analysis and through modelling of scab incidence and count data. Molecular analysis was inconclusive. Statistical modelling of results suggested that conidia may have contributed approximately 20–50% of the primary inoculum in early spring within this orchard: incidence was estimated to be reduced by 20% on potted trees, and lesion number by 50%. These results indicate that, although conidia are still a minority contributor to primary inoculum, their contribution in this orchard is sufficient to require current management to be reviewed. The distribution of infectious salmon anaemia (ISA) was examined among 80 cages from three Atlantic salmon grow-out farms in Maine, USA that were stocked with smolts from a single hatchery. Cage-level disease was broadly defined as one or more moribund fish testing positive for infectious salmon anaemia virus (ISAV) by RT-PCR and a second confirmatory test (IFAT, culture or genotype sequence). Spatio-temporal and cage-level risks were explored using logistic regression and survival analysis. Non-spatial risk factors associated with ISA, or shortened survival time to disease, included increased predation, trucking company choice for smolt transfers, a finely-sedimented benthic substrate, and smaller average size of smolts at stocking. Univariable analysis identified the time-dependent spatial factor 'adjacency to newly infected cages' to be predictive of new infection in neighbouring cages 11-12 weeks later. However, none of the spatial factors, or their lags retained relevance in multiple-variable models. The results suggest a diffuse distribution of virus exposure throughout infected sites, with host-susceptibility factors probably influencing disease manifestation in individual cages. The narrow focus of the current study may limit application of the findings to other sites and year-classes. However, these data support the relevance of husbandry efforts to optimize fish health in regions affected by ISAV. 相似文献

This paper focuses on the interaction between genotype and environment, a critical aspect of plant breeding, from a physiological perspective. We present a theoretical framework largely based on Bradshaw's principles of phenotypic plasticity (Adv. Gen. 13: 115) updated to account for recent developments in physiology and genetics. Against this framework we discuss associations between plasticity of yield and plasticity of phenological development. Plasticity was quantified using linear models of phenotype vs environment for 169 wheat lines grown in 6 environments in Mexico, 32 sunflower hybrids grown in at least 15 environments in Argentina and 7 grapevine varieties grown in at least 14 environments in Australia.In wheat, yield ranged from 0.6 to 7.8 t ha−1 and the range of plasticity was 0.74–1.27 for yield and 0.85–1.17 for time to anthesis. The duration of the post-anthesis period as a fraction of the season was the trait with the largest range of plasticity, i.e. 0.47–1.80. High yield plasticity was an undesirable trait as it was associated with low yield in low-yielding environments. Low yield plasticity and high yield in low-yielding environments were associated with three phenological traits: early anthesis, long duration and low plasticity of post-anthesis development.In sunflower, yield ranged from 0.5 to 4.9 t ha−1 and the range of plasticity was 0.72–1.29 for yield and 0.72–1.22 for time to anthesis. High yield plasticity was a desirable trait as it was primarily associated with high yield in high-yielding environments. High yield plasticity and high yield in high-yielding environments were associated with two phenological traits: late anthesis and high plasticity of time to anthesis.In grapevine, yield ranged from 1.2 to 18.7 t ha−1 and the range of plasticity was 0.79–1.29 for yield, 0.86–1.30 for time of budburst, 0.84–1.18 for flowering, and 0.78–1.16 for veraison. High plasticity of yield was a desirable trait as it was primarily associated with high yield in high-yielding environments. High yield plasticity was associated with two phenological traits: plasticity of budburst and plasticity of anthesis.We report for the first time positive associations between plasticities of yield and phenology in crop species. It is concluded that in addition to phenology per se (i.e. mean time to a phenostage), plasticity of phenological development merits consideration as a distinct trait influencing crop adaptation and yield. 相似文献

In a prospective cross-over study, the duration and magnitude of effect on the electrolyte and plasma volume changes of intravenous (IV) hypertonic hydroxyethyl starch (hyperHES) (7.5%/6%) and mannitol (20%) were compared. Eight Beagle dogs received an IV infusion of 4mL/kg hyperHES (group H) and 4mL/kg mannitol 20% (group M) on separate occasions. Urine and blood samples were taken in the first (T(60)) and second (T(120)) hour after infusion. Significant increases in plasma volume at each time point in group H and M were noted when compared to baseline (start of infusion=T(0)) level. There was no significant difference between groups. Both fluids resulted in diuresis, although no significant difference between groups was noted. A significant increase in plasma sodium (Na) was demonstrated in group H between T(0) and T(60) with a significant increase in the Na and chloride (Cl) fractional excretion (FE) between T(0), T(60) and T(120). In group M no changes in plasma electrolyte concentrations were detected, although FE of Na, Cl and K was increased significantly between T(0) and T(60). In conclusion, hyperHES and mannitol appear to have a volume expanding effect lasting at least 120 min. The hypernatraemia induced by hyperHES was minimal compared to previous reports of hypertonic saline use, and no clinical side effects were noted. HyperHES showed comparable effects to mannitol in increasing plasma volume and diuresis and could be considered for these applications. 相似文献
