The aim of this work was to verify the potential of infrared (IR) spectroscopy in near and mid regions to detect the beginning of the malolactic fermentation (MLF) occurring in a model-wine and the further cells autolysis. MLF in wine is a secondary biotransformation due to lactic acid bacteria that usually occurs spontaneou,;ly or after starter inoculation at the end of alcoholic fermentation. Nowadays, it is desirable to supply winemakers with a new rapid and non-destructive approach to monitor MLF progress and 1R spectroscopy technology appears to be suitable for this purpose. The transformation of L-malic acid into L-lactic acid was carried out by inoculating a synthetic wine with an Oenococcus oeni culture and it was monitored through microbiological and chemical methods. At the same time, Fourier transform near infrared (FT-NIR) spectral data, in diffusive transflection mode using an optic probe, and FT-IR spectra, using an germanium crystal attenuated total reflectance (ATR) cell, were collected. Principal component analysis of the spectra was able to identify absorption bands related to the key molecular modifications that took place during the L-malic acid transformation. Thus, the samples were discriminated according to the fermentation phase. Although this study is a preliminary approach, results confirm that near infrared (NIR) and mid infrared (MIR) spectroscopy could be successfully applied to detect the start of MLF and the autolysis of the lactic acid bacteria (LAB) cells. 相似文献

Two bacterial strains Lactobacillus fructivorans (AS17B), isolated from adult sea bream (Sparus aurata) gut, and Lactobacillus plantarum (906), isolated from human faeces, were administered contemporaneously, during sea bream development using Brachionus plicatilis and/or Artemia salina and dry feed as vectors. The probiotic treatment influenced gut colonization: at 35 days post-hatching (p.h.) L. fructivorans was not present in the gut, but the treatment induced colonization by L. plantarum. At 66 days p.h., L. fructivorans was evident also in the control; moreover, when suitable environmental conditions appeared in the post-metamorphosis gastro-intestinal tract, competition between L. plantarum and L. fructivorans occurred. At 90 days p.h., L. plantarum was displaced by L. fructivorans that became significantly higher with respect to the control. Litopenaeus vannamei (Boone) grown in ponds are exposed to salinities of less than 5 g L?1 during inland shrimp culture or to more than 40 g L?1 from evaporation and reduced water exchange in dry, hot climates. However, dietary requirements for shrimp grown in low or high salinities are not well defined, particularly for fatty acids. Feeding shrimp postlarvae with highly unsaturated fatty acids (HUFA) enhances tolerance to acute exposure to low salinity, as a result of better nutritional status, or/and specific effects of HUFA on membrane function and osmoregulation mechanisms. This study analysed the effect of HUFA supplementation (3% vs. 34%) on L. vannamei juveniles reared for 21 days at low (5 g L?1), medium (30 g L?1) and high salinities (50 g L?1). Juveniles grown at 5 g L?1 had lower survival compared with controls (30 g L?1) or shrimp grown at 50 g L?1, but no significant effect on survival was observed as a result of HUFA enrichment. In contrast, growth was significantly lower for shrimp grown at 50 g L?1, but this effect was compensated by the HUFA‐enriched diet. Osmotic pressure in haemolymph was affected by salinity, but not by HUFA enrichment. Shrimp fed HUFA‐enriched diets had significantly higher levels of eicosapentaenoic acid and docosahexaenoic acid in hepatopancreas and gills. These results demonstrate that growth at high salinities is enhanced with diets containing high HUFA levels, but that HUFA‐enriched diets have no effect on shrimp reared at low salinities. 相似文献

In order to find out a new effective accumulator of polycyclic aromatic hydrocarbons (PAHs) useful for monitoring studies on a large scale and low costs, the accumulation capacity of both biological and artificial matrixes (mosses and polyester fibers, respectively) has been tested. For this purposes, Hypnum cupressiforme and dacron® were exposed to pollution airborne in two sites located nearby an active iron industry and in center of the town of Trieste, where high PAH pollution spots, due to vehicular traffic, are usually detected. The samplers were exposed in six sampling sessions for 21 days. The results obtained were compared with data collected by active PAH samplers, usually employed for official widespread monitoring. The level of correlation between the data sets was calculated. Furthermore, a repeatability study of data was performed. Two feeding trials were conducted to determine the digestibility of a casein-based semi-purified diet and the effects of different protein levels on growth and protein use of spotted sand bass Paralabrax maculatofasciatus juveniles. For trial I, a semipurified diet with vitamin-free casein as the sole source of protein was fed three times a day to apparent satiation, for a period of 20 d. Feces were collected by siphoning each tank. The digestibility of the experimental diet was high: 97% for protein, 89% for lipids, and 84% for gross energy, whereas that of organic matter was 78%. For trial II, seven diets were formulated using vitamin-free casein at graded levels (25, 30, 35, 40, 45, 50, and 55% protein). Triplicate tanks for each dietary treatment were stocked with fish and fed by hand three times a day to apparent satiation for 6 wk. Perfomance of fish fed the different diets was evaluated for survival, percent weight gain, specific growth rate, feed conversion ratio, and protein efficiency ratio. Survival was 100% for all treatments. Growth of spotted sand bass juveniles increased as the dietary protein increased, but no evidence of reaching a plateau was found. The daily feed intake values showed an inverse relation to the protein content of the diets. The feed conversion ratio did not differ among diets containing 40% protein or greater. The results indicate that spotted sand bass juveniles with 2.5-g mean weight need at least 55% dietary protein for best growth when casein is the sole protein source. This paper proposes an evolutionary reading of rural development referred to cases of rapid industrial growth, where a strong concentration process has involved the main urban centers and the successful industrial districts. This territorial development pattern has gradually extinguished rural society and its institutional basis, creating a clear separation between new central and peripheral areas. The consequent effects on local economy and social dynamics reveal the long‐term risks raised in terms of development sustainability. This study was conducted in Brazil in order to assess the potential risk posed by gene escape from transgenic into non-transgenic plants and wild populations. A new methodology was applied to evaluate the gene flow between common bean cultivars, by means of a specially delineated experiment in two stages. The first stage consisted of the planting of one cultivar with violet flowers (BB) as pollen source (‘Diamante Negro’), and a receiver (‘Talismã’) with white flowers (bb), at different distances. The source was sown in the center of the area. The pollen receiver cultivar was sown, in concentrical squares around it. At maturity, the rows were sampled at varied distances from the source in the four cardinal directions. In the second stage, the sampled seeds of the previous stage were sown, and the percentage of outcrossing was evaluated during flowering through the presence of violet flowers (Bb). The highest frequency of natural hybrids, 0.136%, occurred at a distance of 0.5 m between the cultivars. The natural outcrossing rate was practically zero beyond a distance of 3.25 m. 相似文献
