Backfilling a self-assembled monolayer (SAM) of long poly (ethylene glycol) (PEG) with short PEG is a well-known strategy to improve its potential to resist fouling. Here it is shown, using x-ray photoelectron spectroscopy, contact angle, and atomic force microscopy, that backfilling PEG thiol with oligo (ethylene glycol) (OEG) terminated alkane thiol molecules results in underbrush formation. The authors also confirm the absence of phase separated arrangement, which is commonly observed with backfilling experiments involving SAMs of short chain alkane thiol with long chain alkane thiol. Furthermore, it was found that OEG addition caused less PEG desorption when compared to alkane thiol. The ability of surface to resist fouling was tested through serum adsorption and bacterial adhesion studies. The authors demonstrate that the mixed monolayer with PEG and OEG is better than PEG at resisting protein adsorption and bacterial adhesion, and conclude that backfilling PEG with OEG resulting in the underbrush formation enhances the ability of PEG to resist fouling.  相似文献

ObjectivesCardiac repolarization, measured as QT and Tpeak to Tend (TpTe) intervals on the ECG, is important, as irregularities caused by diseases, ventricular hypertrophy, drugs and genetic defects can trigger arrhythmias which predispose human patients to syncope and sudden cardiac death. In horses, repolarization is not well described and therefore QT analysis cannot yet be used diagnostically. Therefore, we sought to describe reference values for the normal QT and TpTe intervals in Standardbreds and to determine the best method for heart rate (HR) correction.Animals30 Standardbreds.MethodsQT and TpTe intervals were measured during rest and exercise and plotted against HR converted to Rpeak to Rpeak interval (RR). Data were fitted with relevant regression models. Intra- and inter-observer agreement was assessed using Bland–Altman analyses.ResultsData were best described by a piecewise linear model (r2 > 0.97). Average prediction error of this model was smaller than for both Bazett and Fridericia corrections. Coefficient of repeatability of intra- and inter-observer variability was 8.76 ms and 5.64 ms respectively and coefficient of variation was 1.77% and 2.76% respectively. TpTe increased with RR in stallions.ConclusionsThe QT interval in Standardbred horses shortens with decreasing RR interval (increasing HR) as in humans, but in a markedly different order as it clearly follows a piecewise linear model. The equine QT interval can be measured easily and there is small intra- and inter-observer variability. This model of the equine QT interval provides clinicians with a method that could support a diagnosis of repolarization disturbances in horses.  相似文献

The ability of lactating mink dams to control glucose homoeostasis, when fed diets containing different ratios of metabolizable energy (ME) from protein, fat and carbohydrates, was studied by measuring plasma concentrations of glucose, insulin, glucagon, urea and free fatty acids (FFA), in the fasted and absorptive state 4 weeks postpartum, in two consecutive years. A total of 36 yearling female mink, fitted with jugular vein catheters and raising litters of six or seven kits, was fed ad libitum lactation diets with different amounts of ME derived from protein, fat and carbohydrates (year 1:61:37:2, 46:37:17 and 31:37:32; year 2:61:38:1, 47:52:1 and 33:66:1). After 3 h fasting the dams were fed 210 kJ ME of the experimental diets. Blood samples were drawn 10 and 5 min before feeding and 30, 60, 90, 120, 150 and 180 min postprandially. The glucose concentration was increased 30 to 150 min postprandially in dams fed the carbohydrate-containing diets (46:37:17 and 31:37:32), whereas the glucose concentration showed no postprandial response in dams fed the carbohydrate-free diets (61:38:1, 47:52:1 and 33:66:1). Plasma insulin concentrations were increased 30 to 120 min postprandially in all dams, irrespective of dietary treatment. Plasma concentrations of glucagon were higher ( P <0.005) in dams fed the low-protein diets (31:37:32 and 33:66:1) than in dams fed the high-protein diets (61:37:2 and 61:38:1). Postprandially, the glucagon : insulin ratios decreased in dams fed the carbohydrate-containing diets, whereas the glucagon:insulin ratios tended to increase in dams fed the carbohydrate-free diets. Plasma concentrations of urea were significantly higher in dams fed the high-protein diets. Plasma concentrations of FFA, measured in the second experiment (year 2) only, showed increased concentrations postprandially, the responses being significant in dams fed the 33:66:1 and 61:38:1 diets. In conclusion, the mink is able to regulate the concentrations of blood constituents involved in maintaining glucose homoeostasis, and thereby to adapt to a wide range of dietary protein and carbohydrate supply.  相似文献

Background

The objective of this study was to investigate the association between average daily gain and the number of Lawsonia intracellularis bacteria in faeces of growing pigs with different levels of diarrhoea.

Methods

A longitudinal field study (n = 150 pigs) was performed in a Danish herd from day 29 to 47 post weaning. Every third day all pigs were weighed, subjected to a clinical examination and faecal samples were obtained. Faecal samples were subjected to dry matter determination and absolute quantification by PCR for L. intracellularis and porcine circovirus type 2 (PCV2). Association between average daily gain, faecal dry matter content, numbers of L. intracellularis bacteria and PCV2 genome copies in faeces was investigated in a multilevel mixed-effects linear model.

Results

Increasing numbers of L. intracellularis log10 bacteria/g faeces were significantly associated with decreasing average daily gain (P < 0.001). The association was decreasing with increasing faecal dry matter content (P < 0.01). The number of PCV2 log10 copies/g faeces was not significantly associated with average daily gain of the pigs (P > 0.5).

Conclusion

The results suggest a potential application of a PCR quantifying L. intracellularis in growing pigs. Faecal dry matter content must be taken into consideration in interpretation of such test results.  相似文献

  • Dredging blue mussels (Mytilus edulis) and thus removing structural elements, inducing resuspension of sediment as well as reducing filtration capacity, will inevitably affect the ecosystem. The study demonstrates that the impacts of fishing can be reduced through gear developments.
  • A new light dredge was tested on commercial vessels using two different experimental setups. First, a twin haul experiment tested the standard gear (i.e., a Dutch dredge) against the light dredge by fishing the two gears side by side onboard the same vessel. Second, a single dredge experiment tested the absolute performance of the two gears by fishing in areas with a known blue mussel density.
  • Results from the twin haul experiment demonstrate that the weight of sediment retained in the gear per square metre fished is 49% less in the light dredge compared with the Dutch dredge which will reduce resuspension of sediment at the surface. Also, the drag resistance of the light dredge was significantly less (177.1 vs. 202.7 kg m‐1). In the twin haul experiment no significant difference was found in the catch per unit effort (CPUE) of the two gears. The single dredge experiment, on the other hand, demonstrated a significant increase in CPUE exceeding 200% when using the light dredge.
  • Seafloor tracks made by the two dredges could not be distinguished by use of side‐scan sonar and the tracks were still detectable 2 months after fishing.
  • It was concluded that replacement of the Dutch dredge with the light dredge would reduce the impact of the fishery on the ecosystem by (i) reducing resuspension of sediment, (ii) reducing fuel consumption, and (iii) potentially reducing energy transfer to the sediment through a reduced gear drag resistance. A potential increase in catch efficiency may reduce the area affected.

An electron spin resonance (ESR)-based method was used for evaluating the levels of radical formation during mashing and in sweet wort. The method included the addition of 5% (v/v) ethanol together with the spin trap alpha-4-pyridyl(1-oxide)- N- tert-butylnitrone (POBN) to wort, followed by monitoring the rate of formation of POBN spin adducts during aerobic heating of the wort. The presence of ethanol makes the spin trapping method more selective and sensitive for the detection of highly reactive radicals such as hydroxyl and alkoxyl radicals. Samples of wort that were collected during the early stages of the mashing process gave higher rates of spin adduct formation than wort samples collected during the later stages. The lower oxidative stability of the early wort samples was confirmed by measuring the rate of oxygen consumption during heating of the wort. The addition of Fe(II) to the wort samples increased the rate of spin adduct formation, whereas the addition of Fe(II) during the mashing had no effect on the oxidative stability of the wort samples. Analysis of the iron content in the sweet wort samples demonstrated that iron added during the mashing had no effect on the iron level in the wort. The moderate temperatures during the early steps of mashing allow the endogenous malt enzymes to be active. The potential antioxidative effects of different redox-active enzymes during mashing were tested by measuring the rate of spin adduct formation in samples of wort. Surprisingly, a high catalase dosage caused a significant, 20% reduction of the initial rate of radical formation, whereas superoxide dismutase had no effect on the oxidation rates. This suggests that hydrogen peroxide and superoxide are not the only intermediates that play a role in the oxidative reactions occurring during aerobic oxidation of sweet wort.  相似文献

Nine stream sites in the Blackfoot River watershed in southeastern Idaho were sampled in September 2000 for water, surficial sediment, aquatic plants, aquatic invertebrates, and fish. Selenium was measured in these aquatic ecosystem components, and a hazard assessment was performed on the data. Water quality characteristics such as pH, hardness, and specific conductance were relatively uniform among the nine sites examined. Selenium was elevated in water, sediment, aquatic plants, aquatic invertebrates, and fish from several sites suggesting deposition in sediments and food web cycling through plants and invertebrates. Selenium was elevated to concentrations of concern in water at eight sites (>5 g/L), sediment at three sites (>2 g/g), aquatic plants at four sites (>4 g/g), aquatic invertebrates at five sites (>3 g/g), and fish at seven sites (>4 g/g in whole body). The hazard assessment of selenium in the aquatic environment suggested low hazard at Sheep Creek, moderate hazard at Trail Creek, upper Slug Creek, lower Slug Creek, and lower Blackfoot River, and high hazard at Angus Creek, upper East Mill Creek, lower East Mill Creek, and Dry Valley Creek. The results of this study are consistent with results of a previous investigation and indicate that selenium concentrations from the phosphate mining area of southeastern Idaho were sufficiently elevated in several ecosystem components to cause adverse effects to aquatic resources in the Blackfoot River watershed.  相似文献

Water characteristics and meat microstructure of NaHCO3-enhanced pork were compared with NaCl- and Na4O7P2-enhanced pork using low-field proton NMR relaxometry, advanced microscopy techniques, and traditional meat quality measurements. Porcine samples were enhanced at 4 degrees C for 48 h with sodium salts individually and in the following combinations: (i) 5% NaCl, (ii) 5% Na4O7P2, (iii) 3% NaHCO3, (iv) 5% NaCl and 5% Na4O7P2, (v) 5% NaCl and 3% NaHCO3, (vi) 5% Na4O7P2 and 3% NaHCO3, and (vii) 5% NaCl, 5% Na4O7P2, and 3% NaHCO3. Independently of the marinade used, the water-binding capacity was improved, cooking loss was reduced, and the yield was enhanced compared with nonmarinated pork samples. This was also reflected in the water mobility within the samples measured by proton NMR relaxometry. Visualization of samples by confocal laser scanning microscopy (CLSM) revealed salt-dependent microstructural changes in the green pork samples treated with NaHCO3, giving rise to nearly complete disintegration of overall structures. High-resolution visualization by atomic force microscopy (AFM) further suggested that a higher cooking loss in sodium chloride-enhanced samples could be ascribed to less solubilization and higher heat-induced protein denaturation compared with phosphate- and bicarbonate-enhanced samples.  相似文献
