This study presents the validation of a high-performance liquid chromatography diode array detector (HPLC-DAD) method for the determination of different mycosporine-like amino acids (MAAs) in the red alga Bostrychia scorpioides. The investigated MAAs, named bostrychines, have only been found in this specific species so far. The developed HPLC-DAD method was successfully applied for the quantification of the major MAAs in Bostrychia scorpioides extracts, collected from four different countries in Europe showing only minor differences between the investigated samples. In the past, several Bostrychia spp. have been reported to include cryptic species, and in some cases such as B. calliptera, B. simpliciuscula, and B. moritziana, the polyphyly was supported by differences in their MAA composition. The uniformity in the MAA composition of the investigated B. scorpioides samples is in agreement with the reported monophyly of this Bostrychia sp.  相似文献

Plant–microorganism associations have long been studied, but their exploitation in agriculture partially or fully replacing chemical fertilizers is still modest. In this study, we evaluated the combined action of rhizobial and plant growth-promoting rhizobacteria inoculants on the yields of soybean and common bean. Seed inoculation with rhizobia (1.2?×?106 cells seed?1) was compared to co-inoculation with Azospirillum brasilense in-furrow (different doses) or on seeds (1.2?×?105 cells seed?1) in nine field experiments. The best in-furrow inoculant dose was 2.5?×?105 cells of A. brasilense seed?1 for both crops. Inoculation with Bradyrhizobium japonicum increased soybean yield by an average 222 kg?ha?1 (8.4 %), and co-inoculation with A. brasilense in-furrow by an average 427 kg?ha?1 (16.1 %); inoculation always improved nodulation. Seed co-inoculation with both microorganisms resulted in a mean yield increase of 420 kg?ha?1 (14.1 %) in soybean relative to the non-inoculated control. For common bean, seed inoculation with Rhizobium tropici increased yield by 98 kg?ha?1 (8.3 %), while co-inoculation with A. brasilense in-furrow resulted in the impressive increase of 285 kg?ha?1 (19.6 %). The cheaper, more sustainable inoculated treatment produced yields equivalent to the more expensive non-inoculated + N-fertilizer treatment. The results confirm the feasibility of using rhizobia and azospirilla as inoculants in a broad range of agricultural systems, replacing expensive and environmentally unfriendly N-fertilizers.  相似文献

Little is known about the adaptability of rambutan (Nephelium lappaceum) to highly acidic soils rich in aluminum (Al). A 2-yr field study was conducted to determine the effects of various levels of soil Al on dry matter production, plant growth, and nutrient concentration in the leaves of four cultivars of rambutan. Cultivars and the cultivar x year interaction were not statistically significant for most variables measured in the study. Total, leaf, petiole, stem and root dry weights significantly increased at soil Al concentrations ranging from 0.67 cmol kg?1 to 11.0 cmol kg?1. At this range of soil Al, the concentrations of Al and manganese (Mn) in leaf tissue declined sharply. The results of this study demonstrate that rambutan is highly tolerant to acid soils and that tolerance may involve an Al- and Mn- exclusion mechanism.  相似文献

This paper presents the effect that the traditional cooking process of black beans (Phaseolus vulgaris, Tamazulapa variety) has on the quantity and composition of soluble (SDF) and insoluble (IDF) dietary fiber of beans, as well as on its protein digestibility and protein quality. There was an increase of IDF from 18.1% in cooked beans to 22.4% in fried beans, and a decrease in SDF from 8.4% to 6.6%, respectively. Starch content decreased from 34.5% to 31.3%. No change was found in lignin. The xylose content was higher in IDF than in SDF and decreased to some extent from cooked to fried beans. Arabinose content was similar in IDF and SDF with no change caused by processing. The fraction containing glucose, mannose and galactose in IDF was higher than in SDF, the content increasing in IDF and decreasing in SDF, with processing. Protein content in IDF was higher than in SDF, with no major change when processing. About 29.5% of the total protein of beans was bound in DF. Protein digestibility and protein quality decreased from cooked to fried beans and was positively related to IDF.  相似文献

Foliar spots caused by Pseudomonas coronafaciens pv. garcae (Pcg), Pseudomonas amygdali pv. tabaci (Pat) and Pseudomonas cichorii (Pch) are major bacterial diseases that can reduce coffee production. However, little is known about the genetic diversity and molecular mechanisms underlying the pathogenicity to coffee plants of these bacteria. In this study, genome sequences of Pcg, Pat and Pch strains isolated from coffee plants in Minas Gerais state, Brazil, were used to assess their variability and plasticity, and compare their type III secretion system (T3SS) and apoplastic effector repertoires as well as tabtoxin biosynthetic/detoxification genes. Genomic diversity was found for all three phytopathogens, among which Pch possesses the highest number of exclusive proteins. The Pcg genome is the most stable whereas that of Pch is the most plastic, which is related to their host ranges. When compared with those of Pseudomonas syringae pv. tomato DC3000, hrp/hrc gene sequences are more conserved in Pcg and Pat than in Pch, which also possesses the smallest T3SS and the largest apoplastic effector repertoires. The only T3SS effector family common to all three pathogens is AvrE, suggesting that, as for other plant–Pseudomonas interactions, it may play a crucial role for pathogenicity towards coffee plants. Apoplastic proteins associated with maintaining the redox balance and degrading proteins/peptides not previously described as important in plant–bacteria interactions were found. Gain/loss of the tabtoxin biosynthetic cluster with retention of the antitoxin gene was observed, indicating that tabtoxin production is not a limiting factor for the occurrence of mixed infections.  相似文献

Eleven experimental and three commercial white quality protein maize (QPM) hybrids and two regular endosperm controls were planted at Celaya, Guanajuato, Mexico with the aim of comparing grain physical characteristics, protein quality, lime-cooking and tortilla making properties. All genotypes were planted under irrigation using a density of 80,000 plants/ha and fertilized with 250 kg N-60 P-60 K per hectare. When compared with the controls these QPM genotypes had lower test (77.4 vs. 76.5 kg/hL) and 1,000 kernel weights (327 vs. 307 g), softer endosperm texture (2.5 vs. 1.8 where 1 = soft, 2 intermediate and 3 hard endosperm), lower protein (10.0 vs. 8.0%), higher nixtamal water uptake after 30 min lime-cooking (50.0 vs. 53.1% moisture) and lower pericarp removal scores. The lower thousand-kernel weight and softer endosperm texture observed in the QPM genotypes lowered the optimum lime-cooking time as estimated with regression equations. Most QPM genotypes had higher amounts of lysine, tryptophan and albumins/globulins when compared with the controls. QPMs HEC 424973, HEC 774986 and HEC 734286 had the best grain traits for nixtamalization and therefore the best potential for industrial utilization. The commercial use of these QPM hybrids should benefit Mexicans who depend on tortillas as the main staple.  相似文献
