Apoptosis‐Related Protein Expression During Pre‐ and Post‐Natal Testicular Development After Administration of Glucocorticoid in utero in the Sheep

Pre‐natal glucocorticoids are used in women at risk of preterm delivery to induce foetal lung maturation. However, glucocorticoids can produce negative outcomes for other tissues such as the reproductive system. We therefore tested the effects of pre‐natal betamethasone on testicular morphology and apoptotic protein immune expression during pre‐ and post‐natal development. Pregnant ewes (n = 42) bearing singleton male foetuses were randomly allocated to receive intramuscular injections of saline or betamethasone (0. 5 mg/kg) at 104, 111 and 118 days of gestation (DG). Testes were collected at 121 and 132 DG, and at 45 and 90 post‐natal days (PD) and subjected to morphometric analysis (volume densities of sex cords and interstitial tissues; sex cord diameter). Immunohistochemistry (% stained area) was used to assess active caspase‐3, Bax, Bcl‐2 and cell‐cycle proteins (PCNA). Compared with control values, betamethasone treatment decreased sex cord diameter at 121 DG, 45 and 90 PD, and sex cord volume at 90 PD. Active caspase‐3 was decreased by betamethasone at 121 DG and 90 PD, but Bax was increased in all betamethasone groups. Bcl‐2 and PCNA decreased in the betamethasone groups at 121 DG and 45 PD, but increased at 132 DG and 90 PD. We conclude that high levels of pre‐natally administered glucocorticoid reduce foetal testicular development, perhaps via changes in the balance between pro‐ and anti‐apoptotic proteins and cell‐cycle proteins. These outcomes could compromise the future spermatogenic potential of male offspring.     

化脓棒状杆菌的分离与鉴定

北京动物园的2头斑羚和1头岩羊相继发病死亡。剖检发现肺部均有淤血和化脓灶。经取其肺组织作细菌培养,均分离到纯粹的杆菌。经细菌形态、培养特性和生化特性分析,鉴定为化脓棒状杆菌。     

东北地域农村朝鲜族民居实态调查研究

以位于延边朝鲜族自治州的朝鲜族聚居村龙山村为研究对象进行调查，阐明了朝鲜族的民居特性。农村民居由木结构土草房变为砖瓦房，平面结构中通间与双通屋较多，屋内间数逐年增加，继承了北朝鲜东北部的民居特色，并维持朝鲜民族传统居住习惯（火炕取暖和坐式生活），但在适应延边地理和气候特性后，逐渐转变成汉族、满族等多民族混居的文化形态。     

Absorption and Synthesis of Immunoglobulins G in Newborn Calves

Newborn calves (n=19) got 4.5 liters of pooled colostrum within three feedings in the first 14 hours post natum (p.n.). The immunoglobulin G1 (IgG1) and IgG2 concentrations in the colostrum pool were 54.9 mg/ml and 4.2 mg/ml. The precolostral serum IgG concentrations in calves were 0.15 mg/ml (IgG1; SD 0.24) and 0.06 mg/ml (IgG2; SD 0.14). The highest serum IgG levels p.n. were measured 12 hours after the first colostrum feeding (9.3 mg IgG1/ml (SD 4.0), 0.8 mg IgG1/ml (SD 1.0). Thereafter, the mean IgG1 level was reduced continuously to the significant lowest concentration of 4.9 mg/ml (SD 2.3) at day 28 p.n. and then increased continuously to the significant highest concentration of 9.0 mg/ml (SD 4.8) on day 77 p.n. The mean concentration of IgG2 was lowest on day 11 p.n. (0.5 mg/ml; SD 0.4) and highest on day 77 p.n. (1.2 mg/ml; SD 0.6).
In blood from 198 calves, housed in Germany and sampled between day 4 and 6 p.n., the IgG concentration averaged 4.9 mg/ml serum (SD 3.3). From 93 dams of these calves a sample of the first colostrum could be obtained showing a mean concentration of 22.0 mg IgG/ml (SD 11.0). IgG levels in the colostrum and in the serum showed a correlation of r=0.37.
In Kenia IgG levels of three week old calves from two farms were measured. The calves were always with mother for the first 24 hours. The mean serum IgG concentrations of the calves were 22.5 mg IgG/ml (n=7, SD 6.8) and 15.2 mg IgG/ml (n=15; SD 6.3). Comparing to the serum IgG levels found in calves of our studies in Germany there were significant differences.     

Photopolymerization and Mass-Independent Sulfur Isotope Fractionations in Carbon Disulfide

Irradiation of gaseous carbon disulfide [CS2(g)] at 313 nanometers produces a dark brown aerosol of (CS2)x. Its thermal decomposition products include disulfur (S2), carbon monosulfide (CS), and (CS)x. The photopolymerization process is accompanied by a large mass-independent isotopic fractionation of sulfur (a 5 to 10 per mil sulfur-33 excess and a 61 to 84 per mil sulfur-36 deficit). Excess sulfur-33 has been observed in several classes of meteorites. Photochemical production of (CS2)x may be important in the origin and evolution of cosmochemical environments such as the presolar nebula, meteorites, asteroids, and planetary atmospheres.     

REVEGETATION OF DECOMPOSED GRANITE ROADCUTS IN KOREA: DEVELOPING DIGGER,EVALUATING COST EFFECTIVENESS,AND DETERMINING DIMENSIONS OF DRILLING HOLES,REVEGETATION SPECIES,AND MULCHING TREATMENT

Decomposed granite roadcuts are difficult to revegetate after losing the topsoil and vegetation cover. We developed a new drilling machine, Digger, to efficiently drill six holes simultaneously on decomposed granite roadcuts to facilitate revegetation. The Digger consists of a base machine (0.7 m³‐level excavator) and a mounting body with six hydraulic motors instead of a bucket. We tested its performance on two roadcuts in southwest Korea using time‐motion studies, which showed that the Digger can drill 240 m² of decomposed granite roadcuts daily. The unit cost of the Digger was less than a half of other roadcut stabilization and revegetation techniques in Korea, making the Digger a cost‐effective revegetation technology. Field germination and growth tests were also conducted to identify appropriate diameter and depth of drilling holes, suitable revegetation species, and mulching treatment. We drilled holes with three different diameters and depths, filled the holes with a mixture of plant seeds and cultivated soil, applied mulching treatments (coir geotextile, shade net, and no mulching), and measured the germination and growth results at two field plots after 1 month and 1 year. The results showed that drilling diameter 10 cm and depth 10 cm were large enough to result in better plant germination and growth. Erosion control species, Poa pratensis L. and Eragrostis curvula (Schrad.) Nees, survived and grew better than native woody species. Coir geotextile improved the plant germination and growth. The time‐motion and revegetation results show that the Digger can be a promising technology to restore decomposed granite roadcuts. Copyright © 2013 John Wiley & Sons, Ltd.     

Evaluation of recombinant 28 kDa outer membrane protein of Brucella abortus for the clinical diagnosis of bovine brucellosis in Korea

Brucella spp. are Gram-negative, facultative, intracellular coccobacilli that are pathogenic to a variety of mammals, including ruminants and humans. The conventional serological test for diagnosing brucellosis in cattle in Korea is the standard tube agglutination test. However, agglutination tests sometimes give false-positive results due to cross-reactions with other pathogens. The outer membrane proteins of Brucella species have been extensively studied for their immunogenicity and serodiagnostic applications. However, an application of B. abortus OMPs for serodiagnosis has not been successfully established. In this study, cloning and expression of B. abortus Omp28, a group 3 antigen, were accomplished by PCR amplification cloning into a pMAL expression system, and purification of a recombinant Omp28 (rOmp28). The immunogenicity of rOmp28 was confirmed by Western blot analysis with Brucella-positive bovine serum. To determine whether rOmp2 has a potential benefit for use in the serodiagnosis of bovine brucellosis, rOmp28-based ELISA and latex bead agglutination test were performed. B. abortus positive (n=122) or negative (n=88) from TAT were positive (118/122, 96.7%) or negative (84/88, 95.4%) in ELISA and were positive (94/122, 77%) or negative (71/88, 81.7%) in that the latex bead agglutination test, respectively. The sensitivity, specificity and accuracy were 96.7, 95.4, 96.2% in ELISA and 77, 80.6, 78.5% in latex bead agglutination test, respectively. These findings suggest that the rOmp28 of B. abortus might be a good candidate for developing serological diagnostic tools for bovine brucellosis.
