Complementation studies were carried out, using temperature-sensitive (t-s) mutants of blue-tongue virus (BTV). The results proved to be inconclusive as only low indices of complementation were obtained. No discrepancy was found between the previous classification of these mutants in 6 recombination classes and the complementation data recored. In general, the t-s mutants require a latent growth period of 16-20 h at 28 degrees C and maximum titres can be demonstrated 40-48 h post-infection. One mutant, (F211), however, consistently had a growth lag phase of 32 h. Mutants of the 6 recombination groups were further classified into 2 groups by temperature-shift studies. One calss of mutants expressed their t-s lesion prior to 24 h and the other class only after 24 h post-infection. Mutant F73 was found to be defective in its ability to synthesize ssRNA at a late stage in the replication cycle at the non-permissive temperature.  相似文献

In the Netherlands the total active inland drift-sand area has been declining rapidly during the last 50 years. To preserve the inland drift sands, it is necessary to understand its origin and development and the role of human activity in this semi-natural ecotype. The objective of this literature review is to describe the development of the drift-sand ecotopes, to explain the rapid decline of the active drift sands, and to develop a management strategy for the remaining active drift sands. Inland drift-sand landscapes are relatively young landscapes of Holocene age. They often occur as oval-shaped cells with a length of 1.5 to over 6 km in the direction of the prevailing wind. These cells presumably represent reactivated deposits of Younger Cover Sands. Large-scale erosion events in combination with human activity suppressed the development of vegetation. After the change in land use in the first half of the 20th century in which most of the drift sands were re-afforested, the vegetation succession started to show a progressive development. In this stage inland drift-sand ecotopes developed in most of the remaining drift sands with all forms of the typical succession stages from bare sand to forest. The rate at which this development took place mainly depended on the geomorphological development stage of the area, the area size and human activity. Since the 1960s the increased nitrogen deposition has accelerated the vegetation succession, not only resulting in a further decline of the drift sands, but also in a loss of the fragile balance between the different ecotopes and loss of its typical habitants like the Tree Grayling and Tawny Pipit. Most drift-sand vegetation and fauna need the presence of bare sand nearby and a certain level of erosion activity to survive. To preserve the drift-sand ecotype, it is therefore recommended to keep the area affected by erosion sufficiently large (process management). In the meantime one should also ‘maintain’ or increase the wind force in the drift-sand area by suppressing the growth of high vegetation and removing trees, which form a wind barrier. In areas which are less suitable for reactivation, one could restore the mosaic vegetation by removing the vegetation on a limited scale (pattern management). More research is needed to develop a more balanced management strategy and to develop a management tool for the managers of inland drift sands. Also the role of the increased nitrogen deposition in the regeneration process needs further investigation in order to find an effective way to suppress its effect. The development of management strategies for the Dutch inland drift sands might be of great value to drift-sand areas in Western Europe where nature conservationists start to show more interest in the restoration of former drift-sand areas.  相似文献

ABSTRACT The interaction between the oomycete Pythium oligandrum and various soilborne oomycete and fungal plant pathogens (P. ultimum, P. aphanidermatum, Fusarium oxysporum f. sp. radicis-lycopersici, Verticillium albo-atrum, Rhizoctonia solani, and Phytophthora megasperma) was studied by light and electron microscopy in order to assess the relative contribution of mycoparasitism and antibiosis in the antagonistic process. Scanning electron microscope investigations of the interaction regions showed that structural alterations of all pathogenic fungi and oomycetes (except for Phytophthora megasperma) occurred soon after contact with the antagonist. Light and transmission electron microscope studies of the interaction region between the antagonist and P. ultimum revealed that intimate contact between both partners preceded a sequence of degradation events including aggregation of host cytoplasm and penetration of altered host hyphae. Localization of the host wall cellulose component showed that cellulose was altered at potential penetration sites. A similar scheme of events was observed during the interaction between P. oligandrum and F. oxysporum f. sp. radicis-lycopersici, with the exception that complete loss of host protoplasm was associated with antagonist invasion. The interaction between P. oligandrum and R. solani resulted in an abnormal deposition of a wall-like material at potential penetration sites for the antagonist. However, the antagonist displayed the ability to circumvent this barrier and penetrate host hyphae by locally altering the chitin component of the host hyphal wall. Interestingly, antagonist cells also showed extensive alteration as evidenced by the frequent occurrence of empty hyphal shells. In the case of Phytophthora megasperma, hyphal interactions did not occur, but hyphae of the plant pathogen were damaged severely. At least two distinct mechanisms appear to be involved in the process of oomycete and fungal attack by P. oligandrum: (i) mycoparasitism, mediated by intimate hyphal interactions, and (ii) antibiosis, with alteration of the host hyphae prior to contact with the antagonist. However, the possibility that the antagonistic process may rely on the dual action of antibiotics and hydrolytic enzymes is discussed.  相似文献

We describe three different outbreaks of mastitis caused by M. mycoides subspecies mycoides LC type (Mmm LC) in three goat flocks from the Extremadura Region of south-west Spain. Thirty-two fast-growing isolates were obtained on Hayflick's and Friis's media with inhibitors from different specimens. All were identified as Mmm LC in spite of their cultural, biochemical and serological features.  相似文献

Lambs were infected daily with 1000 Ostertagia circumcincta larvae. At intervals of 4 weeks groups of lambs were each challenged with a dose of 28,000 larvae, and their worm burdens compared with those of unchallenged controls. The measure of their ability to resist a challenge infection, which would have determined their degree of "protection", was obscured by the separate phenomenon of self-cure. Evidence suggested that the different manifestations of self-cure were determined by the state of resistance of the host and followed a set chronological sequence.  相似文献

In the process to the marketing of cultivars, identification of superior test locations within multi-environment variety trial schemes is of critical relevance. It is relevant to breeding organizations as well as to governmental organizations in charge of cultivar registration. Where competition among breeding companies exists, effective and fair multi-environment variety trials are of utmost importance to motivate investment in breeding. The objective of this study was to use genotype main effect plus genotype by environment interaction (GGE) biplot analysis to evaluate test locations in terms of discrimination ability, representativeness and desirability, and to investigate the presence of multiple mega-environments in cotton production in the Yangtze River Valley (YaRV), China. Four traits (cotton lint yield, fiber length, lint breaking tenacity, micronaire) and two composite selection indices were considered. It was found that the assumption of a single mega-environment in the YaRV for cotton production does not hold. The YaRV consists of three cotton mega-environments: a main one represented by 11 locations and two minor ones represented by two test locations each. This demands that the strategy of cotton variety registration or recommendation must be adjusted. GGE biplot analysis has also led to the identification of test location superior for cotton variety evaluation. Although test location desirable for selecting different traits varied greatly, Jinzhou, Hubei Province, China, was found to be desirable for selecting for all traits considered while Jianyang, Sichuan Province, China, was found to be desirable for none.  相似文献

Andean Potato is a major crop for farmers in the Andes and represents an important gene pool for potato improvement. We present the population structure and genetic diversity of 88 Andigena accessions collected in Northwestern Argentina based on functional markers (25 SSR) distributed along 12 chromosomes. Polymorphic information content ranged from 0.40 to 0.87. A Bayesian approach, a Principal Coordinate Analysis and a Cluster Analysis revealed the presence of: I) a major group containing most of the Andean accessions and II) a smaller group including the out-group cv. Spunta and the sequenced genotype DM. Several group specific alleles were detected. AMOVA showed that 81% of the variability was within each group. Eleven of the SSRs analyzed are linked or within genes reported to regulate traits of nutritional and industrial interest. Additionally, the allelic variant of a photoperiod dependent tuberization regulator gene, StCDF1.2, was exclusively detected in accessions clustered in group II.  相似文献
