The essential oil of Artemisia annua aerial parts, consisting of camphor (44%), germacrene D (16%), trans-pinocarveol (11%), beta-selinene (9%), beta-caryophyllene (9%) and artemisia ketone (3%), was screened for its antimicrobial activity. The essential oil remarkably inhibited the growth of tested Gram-positive bacteria Enterococcus hirae and both tested fungi. The expression of growth factors was evaluated immunohistochemically in normal and cystic ovaries of sows. The immunohistochemically stained area (IHCSA) was quantified by image analysis to analyse the expression of these proteins in the follicular wall of secondary, tertiary and cystic follicles. IGF‐I immunoreactivity was strong in the granulosa cell layer (GC), moderate in the theca interna (TI) and mild in the theca externa (TE) of the normal follicles. There was severe reduction of the labelling to IGF‐I in the GC of the follicular and luteinized cysts. In the normal follicles, the reactivity for IGF‐II was very similar to pattern noted in IGF‐I. There was reduction of the IHCSAs in the GC of the follicular and luteinized cysts, but the decrease was not significant. The staining of the IGF‐II in the TI and TE of the cysts was increased, in comparison with normal follicles. The IHCSAs for VEGF were higher in the GC and TE of the normal follicles in contrast to TI, but this difference was noted only in the tertiary follicle. The VEGF reactivity increased in the GC of the cysts, in relation to normal follicles. The results of the current study show that the formation of ovarian cysts in sows is associated with alterations in the immunohistochemical expression of some growth factors. 相似文献
Crystallization and melting properties of triacylglycerols (TGs) in anhydrous goat's milk fat (AGMF) are investigated by X-ray diffraction as a function of temperature (XRDT) coupled with high-sensitivity differential scanning calorimetry (DSC), using synchrotron radiation and Microcalix. The polymorphic behavior of AGMF was monitored by varying the cooling rates between 5 and 1 degrees C/min from 45 to -20 degrees C with their subsequent melting at 1 degrees C/min. Quenching of AGMF at -20 degrees C was also examined to determine the metastable polymorphic form of AGMF. At intermediate cooling rates, TGs in AGMF crystallize, from about 18 degrees C in two different lamellar structures with triple chain length 3Lalpha stacking of 72 A and a double chain length 2Lalpha stacking of 48 A, which are correlated to two overlapped exothermic peaks recorded by DSC. A reversible transition sub alpha <--> alpha was observed. Subsequent heating at 1 degrees C/min shows numerous structural rearrangements before final melting. At fast cooling of AGMF (5 degrees C/min), similar unstable crystalline varieties are formed while three endotherms are recorded. Several new unstable lamellar structures are observed after quenching. All of these data are compared to those previously reported at slow cooling (0.1 degrees C/min) showing a relative stability of the structures formed. In spite of general similitude, the thermal and structural behavior of the goat's milk is more complex than that of the cow's milk. 相似文献
Yield mapping technologies can help to increase the quantity and quality of agricultural production. Current systems only focus on the quantification of the harvest, but the quality has equal or greater importance in some perennial crops and impacts directly on the financial profitability. Therefore, a system was developed to quantify and relate the quality obtained in the classification line with the plants of the orchard and for decision-making. The system is comprised of hardware, which obtains the location of the harvester bag during harvesting and unloading at the unloading site, and software that processes the collected data. The cloud of real-time data contributed from the different collectors (bins) allows the construction of yield maps, considering the multi-stage harvesting system. Further, the system enables the creation of a detailed map of the plants and fruits harvested. As the harvest focuses on quality, it takes place in stages, depending on the ripening of the fruits. In addition to the yield maps, the system allows identification of the efficiency of each worker undertaking the harvest by the number of performed discharges and by the time spent. The system was developed in partnership with the Federal Technological University of Paraná and Embrapa Uva & Vinho and was tested in apple orchards in southern Brazil. Although the system was evaluated with only data from apple cultivation, monitoring the quality and quantifying other orchard fruits can positively impact the fruit sector.
The comparison of resistance to natural tick infestation by Rhipicephalus microplus (Canestrini, 1887) of crossbred Senepol?×?Limousin and purebred Limousin cattle was investigated. The Senepol breed, originated from St Croix Island in the Caribbean is considered as a Bos taurus breed adapted to tropical conditions. Despite its B. taurus genetic background, it is believed to have a good tick resistance, but this resistance has never been assessed previously. Tick counts under natural infestation were carried out to investigate the difference of susceptibility between crossbred Senepol?×?Limousin and purebred Limousin cattle. Mixed-effect models were used to assess the effect of the breed on the number of ticks. Results show that Senepol?×?Limousin are five times less infested by ticks than purebred Limousin. These results underline the opportunity to use Senepol cattle for crossing with susceptible B. taurus breeds in tick infested areas, to combine tick resistance with beef production abilities. 相似文献
Brucella spp. are Gram-negative bacteria that behave as facultative intracellular parasites of a variety of mammals. This genus includes smooth (S) and rough (R) species that carry S and R lipopolysaccharides (LPS), respectively. S-LPS is a virulence factor, and mutants affected in the S-LPS O-polysaccharide (R mutants), core oligosaccharide or both show attenuation. However, B. ovis is naturally R and is virulent in sheep. We studied the role of B. ovis LPS in virulence by mutating the orthologues of wadA, wadB and wadC, three genes known to encode LPS core glycosyltransferases in S brucellae. When mapped with antibodies to outer membrane proteins (Omps) and R-LPS, wadB and wadC mutants displayed defects in LPS structure and outer membrane topology but inactivation of wadA had little or no effect. Consistent with these observations, the wadB and wadC but not the wadA mutants were attenuated in mice. When tested as vaccines, the wadB and wadC mutants protected mice against B. ovis challenge. The results demonstrate that the LPS core is a structure essential for survival in vivo not only of S brucellae but also of a naturally R Brucella pathogenic species, and they confirm our previous hypothesis that the Brucella LPS core is a target for vaccine development. Since vaccine B. melitensis Rev 1 is S and thus interferes in serological testing for S brucellae, wadB mutant represents a candidate vaccine to be evaluated against B. ovis infection of sheep suitable for areas free of B. melitensis. 相似文献
A method for the purification of chinook Salmon (Oncorhynchus tshawytscha) GH, which retains its biological activity, is described. The biological activity was investigated with an established radioreceptor
assay using liver membranes from pregnant rabbits and bovine GH as standard and labelled hormone. The enrichment of the preparation
was checked with electrophoresis (SDS-PAGE). Extraction and further steps were carried out using low molarity alkaline buffer
(pH 8–10, M = 100 mM). Three chromatography steps were performed (Concanavalin-A sepharose, Bio-gel P60, DEAE). Ion exchange
chromatography was performed under isocratic conditions (using a 50 cm column). Two isoforms (sGH1 and sGH2) were isolated.
The purification yield is 0.7% compared to lyophilized pituitaries. The molecule is homogeneous in SDS-PAGE. Contamination
by prolactin, gonadotrophin and corticotrophin is negligible (< 0.5%). It could be demonstrated that the biological activity
of the preparation is maintained since this preparation stimulates the growth of juvenile trout (Salmo gairdneri) and binds specifically (35%) to trout liver membranes. 相似文献
Forest management can seriously modify the biodiversity of forest dwelling species, but the consequences are poorly known for certain taxa, particularly soil fauna, for which few studies have been published. We compared the biodiversity of dictyostelids cellular slime moulds in a managed and an unmanaged forest in the French Alps and analysed the influence of environmental factors on species richness and abundance of dictyostelids. To our knowledge, this study is the first one undertaken in the European Alps. We must better understand the influence of various environmental factors on the biodiversity of these organisms if we want to accurately define their functional role in the soil. In our study, dictyostelids showed lower levels of diversity compared to previously published results. The mean species richness of dictyostelids was marginally higher in unmanaged than in managed forests and biodiversity indices were significantly correlated with elevation and pH. This suggests that environmental factors have a predominant effect on the biodiversity of dictyostelids and that the effect of forest management is secondary. 相似文献
