Uracil DNA glycosylase activity is dispensable for immunoglobulin class switch

Activation-induced cytidine deaminase (AID) is required for the DNA cleavage step in immunoglobulin class switch recombination (CSR). AID is proposed to deaminate cytosine to generate uracil (U) in either mRNA or DNA. In the second instance, DNA cleavage depends on uracil DNA glycosylase (UNG) for removal of U. Using phosphorylated histone gamma-H2AX focus formation as a marker of DNA cleavage, we found that the UNG inhibitor Ugi did not inhibit DNA cleavage in immunoglobulin heavy chain (IgH) locus during CSR, even though Ugi blocked UNG binding to DNA and strongly inhibited CSR. Strikingly, UNG mutants that had lost the capability of removing U rescued CSR in UNG-/- B cells. These results indicate that UNG is involved in the repair step of CSR yet by an unknown mechanism. The dispensability of U removal in the DNA cleavage step of CSR requires a reconsideration of the model of DNA deamination by AID.     

Effect of Lherzolite on Chemical Fractions of Cd and Zn and their Uptake by Plants in Contaminated Soil

Immobilizing materials such as lherzolite could reduce metal bioavailability but the effectiveness of lherzolite on the extractability and bioavailability of cadmium (Cd) and zinc (Zn) is rarely investigated. We conducted a greenhouse experiment to investigate the effect of 5% application of lherzolite to a contaminated soil on the chemical fractionation of Cd and Zn and their uptake by radish (Raphanus sativus L.) and Japanese mustard spinach (Brassica rapa L. var. perviridis). Both plants were grown in a highly contaminated (with Cd and Zn) sandy loam soil. Plants were cultivated consecutively three times in the same pots. After the third cultivation, soil samples were collected and analyzed by sequential extraction procedure into five operationally defined fractions (F1—exchangeable, F2—carbonate-bound, F3—oxides-bound, F4—bound with organic matter, and F5—residual). Addition of lherzolite to soil decreased 50% of exchangeable (F1) Cd but it increased the carbonate (F2), oxide (F3), and organic (F4) fraction Cd. For Zn, application of lherzolite resulted into the reduction of both F1 (87%) and F2 (33%) fractions but it increased the F3, F4, and F5 fractions. The reduction in exchangeable fraction of Cd and Zn in the soil resulted in higher plant growth and lower concentrations of both Cd (64% to 92%) and Zn (78% to 99%) in plant tissues of both plant species grown. We may thus conclude that application of lherzolite resulted into lower availability of these metals in the soil leading to lower uptake of Cd and Zn by plant roots, lower toxicity, and ultimately higher plant growth.     

Determination of the Chemical Structures of Tandyukisins B–D,Isolated from a Marine Sponge-Derived Fungus

Tandyukisins B–D (1–3), novel decalin derivatives, have been isolated from a strain of Trichoderma harzianum OUPS-111D-4 originally derived from the marine sponge Halichondria okadai, and their structures have been elucidated on the basis of spectroscopic analyses using 1D and 2D NMR techniques. In addition, their chemical structures were established by chemical transformation. They exhibited weak cytotoxicity, but selective growth inhibition on panel screening using 39 human cancer cell lines.     

A mixed apocrine gland tumor with metastases to the bone and bone marrow in a miniature poodle

A 10-year-old female miniature poodle had a mass in its carpal joint of the left forelimb. The tumor was divided into small multiple lobules by delicate connective tissues, and necroses were found in some of the central lobules. In some connective stromal areas, chondroid and osteoid tissues were formed. The tumor cells were similar to the structure of apocrine gland epithelial cells with apical blebs resembling apocrine secretion and eosinophilic secretary materials within the luminal space, and spindle cells were sometimes found in the basal area of the glandular structure. In some areas, tumor cells invaded in the blood vessels, bone and bone marrow. Immunohistochemically, the tumor cells forming tubulo-acinar to solid structures were intensely positive for cytokeratin and keratin K8/K18, and the spindle cells were positive for vimentin and alpha-smooth muscle actin. This case was diagnosed as a malignant mixed apocrine gland tumor with metastases to the bone and bone marrow.     

Hematology of free-living marsh deer (Blastocerus dichotomus) from southeast Brazil.

This work reports basic hematologic values of a sample of a population of free-living marsh deer (Blastocerus dichotomus) living by the Paraná River in Southeast Brazil. Hematologic values are presented separately for male, female, and young animals as well as for anesthetized and nonanesthetized cervids. Nonanesthetized deer restrained by physical means had significantly higher erythrocyte indices and total leukocyte counts. Comparisons of blood parameters of anesthetized animals of different ages and gender differed slightly, with only two significant differences observed: young animals had significantly higher red blood cell counts than adult males and a lower blood total protein content when compared to adult females. Results indicate that two main reference ranges for blood values should be considered for marsh deer, for blood obtained from anesthetized or physically restrained individuals.
