Several water-soluble pectic polysaccharides were isolated from the pumpkin fruit biomass and characterized by composition, structural features and molecular properties. The pectic polysaccharides were tested for antitussive activity by studying the effects of citric acid-induced cough reflex in guinea pigs and reactivity of the airway smooth muscle in vivo conditions in comparison to the narcotic drug codeine. Oral administration of all pectic polysaccharides from pumpkin inhibited the number of coughs induced by citric acid in guinea pigs, but to various extents. The results indicated that the antitussive activity of the pectic polysaccharides is affected by their molecular and structural properties, whereby a synergistic action between the polysaccharide and non-carbohydrate components on the biological response has been suggested as well. The cough depressive efficacy of most of the tested polysaccharides was comparable and even higher than that of codeine. Moreover, the application of these polysaccharides provoked any side effects what is their advantage towards the conventional opioid-derived antitussive agents. 相似文献

Steadily increasing damage to Norway spruce forests in Europe has caused researchers and managers to consider whether these forests can be converted to more stable ecosystems. In a central European mountain region, we investigated whether management systems (MSs) specified by regional stakeholders provide sound alternatives to the currently applied management. We used the forest model Sibyla to explore whether the tested MSs differ in their sensitivity to climate change in terms of altered biomass production, stand structure, forest damage, and financial outcome. The tested MSs were no-management (NM), currently applied management (BAU), and management based on the preferences of forest managers (FM) or on the preferences of other stakeholders (OSH). With NM, spruce remained dominant during the simulation period 2010–2100, and the rate of damage significantly increased. Spruce also remained dominant with FM, while the abundance of non-spruce species significantly increased with BAU and OSH. The rate of salvage logging converged at 50% of the total harvest for all MSs up to 2050. Climate change reduced biomass production (?15%) with all MSs but had a negligible effect on biodiversity indicators. The average initial value of the simulated stands was 20,000 € ha?1 and the nominal value in 2100 was between 1900 and 10,900 € ha?1. The Net Present Value calculated with the 2% interest rate was negative during the whole simulation period (?5600 to ?18,500 € ha?1 in 2100). Effect of climate change on all financial indicators was negative. Our findings indicate that secondary spruce forests are highly vulnerable and that the systems proposed by both forest managers and other regional stakeholders failed to significantly reduce forest damage and stabilize forest production. 相似文献

Forest soil is susceptible to changes in its top layers. These changes occur during ground-based forest harvesting and the rate of soil regeneration depends on the environmental conditions and the extent of the disturbance. This paper was focused on analyzing the changes of soil characteristics such as the depth of the erosion profile, bulk density of soil, its penetration resistance, and the subsurface concentration of CO2 in soils five years after forest harvesting. The study took place in four forest stands harvested by a skidder and a harvester/forwarder combination. Statistical analyses did not prove significant changes of the characteristics of the disturbed soil after the five-year period without machine traffic: the profile depth did not change significantly, except for one stand, where the ruts became deeper. Other characteristics, such as the bulk density of soil also did not show any significant regeneration (1.29–1.36 g cm?3 in the rut; initial measurements versus 1.34–1.38 g cm?3 in the rut; repeated measurements). The penetration resistance, as well as the subsurface CO2 concentration, were variable, and the results inconclusive. Our results suggest that five years was not enough time for soil to regenerate significantly after being disturbed by ground-based machinery. 相似文献

The free radical scavenging activity of the water infusions, different organic solvent extracts and some constituents from Ligustrum vulgare and Ligustrum delavayanum leaves was assessed with the aid of DPPH radical. Among the samples screened, water infusions had the strongest free radical scavenging capacity. Limiting surface soil disturbance caused by forest harvesting machines is an important task and is influenced by the selection of efficient and reliable predictors of such disturbance. Our objective was to determine whether soil moisture content affects soil load bearing capacity and the formation of ruts. Measurements were conducted in six forest stands where various machines operated. We measured the formation of ruts along skid trails in connection with varying soil moisture content. Soil moisture content was determined through the gravimetric sampling method. Our results showed that severe(rut depth16–25 cm) to very severe disturbance(rut depth [26 cm)occurred in forest stands where the instantaneous soil moisture exceeded its plasticity limits defined through Atterberg limits. Atterberg limits of soil plasticity ranged from 26 to 32 % in individual stands. Regression and correlation analysis confirmed a moderately strong relationship(R = 0.52; p \ 0.05) between soil moisture content and average rut depth. This confirmed that soil moisture is a suitable and effective predictor of soil disturbance. 相似文献

Abandoned fields are perceived as potential habitats for species of threatened semi-natural dry grasslands. However, information
is lacking regarding how the spontaneous colonization of abandoned fields depends on the broader spatial context. We recorded
the occurrence of 87 target species in 46 abandoned fields and 339 dry grasslands. We tested the effect of the isolation of
abandoned fields from source grasslands on the number of dry grassland species occurring in abandoned fields either with or
without habitat characteristics being used as covariates. The isolation of the fields was calculated using the distance and

always explained the number of grassland species in the abandoned fields better than I

. The effect of isolation became smaller or even non-significant with the inclusion of covariates; it also changed with the
method used for measuring distance (edge-to-edge or center-to-center), and it was lower when other abandoned fields were considered
as additional source habitats. The different performance of the two isolation measures can be explained by the weak species–area
relationship in the grasslands, indicating differences in their habitat quality. Species richness is a better proxy of habitat
importance in terms of propagule source than habitat area, and the new isolation measure is therefore suitable for studying
the effects of landscape structure on species richness in landscapes presenting a weak species–area relationship, such as
areas exhibiting pronounced effects of land-use history. Inclusion of habitat characteristics as covariates may considerably

ABSTRACT: Molecular epidemiology has proven to be an essential tool in the control of classical swine fever (CSF) and its use has significantly increased during the past two decades. Phylogenetic analysis is a prerequisite for virus tracing and thus allows implementing more effective control measures. So far, fragments of the 5′NTR (150 nucleotides, nt) and the E2 gene (190 nt) have frequently been used for phylogenetic analyses. The short sequence lengths represent a limiting factor for differentiation of closely related isolates and also for confidence levels of proposed CSFV groups and subgroups. In this study, we used a set of 33 CSFV isolates in order to determine the nucleotide sequences of a 3508-3510 nt region within the 5′ terminal third of the viral genome. Including 22 additional sequences from GenBank database different regions of the genome, comprising the formerly used short 5′NTR and E2 fragments as well as the genomic regions encoding the individual viral proteins Npro, C, Erns, E1, and E2, were compared with respect to variability and suitability for phylogenetic analysis. Full-length E2 encoding sequences (1119 nt) proved to be most suitable for reliable and statistically significant phylogeny and analyses revealed results as good as obtained with the much longer entire 5′NTR-E2 sequences. This strategy is therefore recommended by the EU and OIE Reference Laboratory for CSF as it provides a solid and improved basis for CSFV molecular epidemiology. Finally, the power of this method is illustrated by the phylogenetic analysis of closely related CSFV isolates from a recent outbreak in Lithuania. 相似文献

The effect of application of omega-3 polyunsaturated fatty acids (omega-3 PUFA) on intestinal colonization by Lactobacillus paracasei and on cellular immunity has been investigated in gnotobiotic pigs. The administration of polyunsaturated fatty acids positively affected the adhesion of Lactobacillus paracasei to the jejunal mucosa of gnotobiotic piglets. When compared to the control group, the number of Lactobacillus paracasei adhering to the jejunal mucosa was by 12% higher in piglets of the experimental group (5.10 log 10/cm2 vs. 4.55 log 10/cm2). The respective counts of Lactobacillus paracasei adhering to the ileal and colonic mucosa of 28 day old gnotobiotic piglets reached 4.45 and 5.05 log 10/cm2 in group C and 4.44 and 4.95 log 10/cm2 in group E. Omega-3 PUFA supplementation increased the phagocytic activity of neutrophils by almost 100% on day 28 of life as well as the subpopulations of lymphocytes (CD8) in the peripheral blood of germ-free piglets on day 21 of life. Our results indicate that the action of probiotics in the gut may be modulated by dietary PUFA. The stimulatory effect of PUFA upon adhesion of lactobacilli could be used for enhancing the effectiveness of probiotics in inhibiting digestive tract pathogens. 相似文献

The aim of this study was to investigate the effect of neonatal iron deficiency on immune functions in young piglets. While control piglets were not given any iron preparation until the age of 21 days, another group of piglets was given 200 mg of Fe(3+)-dextran i.m. on day 3. Red blood cell parameters in the former, iron-deficient group were characteristic of hypochromic anaemia. In addition, the total leucocyte count (P < 0.01), relative and absolute neutrophil count (P < 0.01) and absolute lymphocyte count (P < 0.05) in peripheral blood were found significantly lower in iron-deficient piglets than in their iron-supplemented counterparts. Lymphocyte activity as measured by in vitro lymphocyte transformation test was impaired in iron-deficient piglets. A statistically significant decrease in circulating B-lymphocyte numbers was found in non-supplemented animals. Iron deficiency apparently negatively influenced the immunocompetence in piglets. 相似文献

The existence of two types of the immunoglobulin (Ig) light chain in pigs was documented>30 years ago and has been confirmed by the cloning of porcine light chain genes homologous to human and murine Ig kappa (Igkappa) and Ig lambda (Iglambda). However, immunochemical reagents defining these two light chain isotypes have not been characterized. Here, we show that rabbit antisera specific for human Igkappa and Iglambda and certain anti-porcine light chain monoclonal antibodies (mAb) are useful in distinguishing light chain isotypes by flow cytometry (FCM). Porcine B cell lines L23 and L35 stained positive only with anti-human Iglambda antiserum and were negative when tested using anti-human Igkappa antiserum. While mAbs K139.3E1, 1G6 and 27.7.1 also tested positive on these cell lines, mAb 27.2.1 did not. Therefore, FCM was used to examine the hypothesis that K139.3E1, 1G6 and 27.7.1 are Iglambda-specific whereas mAb 27.2.1 recognizes the Igkappa chain in pigs. Double staining of peripheral blood mononuclear cells (PBMC) with pairs of anti-light chain mAbs and using cocktails of anti-light chain mAbs and anti-human polyclonal antiserum, confirmed this hypothesis with the exception that mAb K139.3E1 appears to recognize only a subset of Iglambda(+) B cells in most pigs. In summary, we identified two pan-specific anti-pig Iglambda mAbs, one anti-lambda mAb that recognizes a lambda-light chain subset and one anti-pig Igkappa mAb. 相似文献
