BACKGROUND: The present study investigated the effect of seven Bacillus‐species plant‐growth‐promoting rhizobacteria (PGPR) seed treatments on the induction of disease resistance in cowpea against mosaic disease caused by the blackeye cowpea mosaic strain of bean common mosaic virus (BCMV). RESULTS: Initially, although all PGPR strains recorded significant enhancement of seed germination and seedling vigour, GBO3 and T4 strains were very promising. In general, all strains gave reduced BCMV incidence compared with the non‐bacterised control, both under screen‐house and under field conditions. Cowpea seeds treated with Bacillus pumilus (T4) and Bacillus subtilis (GBO3) strains offered protection of 42 and 41% against BCMV under screen‐house conditions. Under field conditions, strain GBO3 offered 34% protection against BCMV. The protection offered by PGPR strains against BCMV was evaluated by indirect enzyme‐linked immunosorbent assay (ELISA), with lowest immunoreactive values recorded in cowpea seeds treated with strains GBO3 and T4 in comparison with the non‐bacterised control. In addition, it was observed that strain combination worked better in inducing resistance than individual strains. Cowpea seeds treated with a combination of strains GBO3 + T4 registered the highest protection against BCMV. CONCLUSION: PGPR strains were effective in protecting cowpea plants against BCMV under both screen‐house and field conditions by inducing resistance against the virus. An efficient, in vitro clonal propagation protocol has been established for Gardenia latifolia Ait. using mature nodal explants on Murashige and Skoog (MS) medium fortified with cytokinins (BA/Kn/2-iP) (1.0–5.0 mg l?1) in combination with auxin IAA (0.5 mg l?1). Maximum bud break (87 %) with shoot number (7.2 ± 0.26) observed on MS medium supplemented with BA (4.0 mg l?1) and IAA (0.5 mg l?1). Maximum number of shoots (30 ± 0.46) with shoot length of (0.9 ± 0.03 cm) observed on MS medium supplemented with BA (2.0 mg l?1), Kn (2.0 mg l?1) and IAA (0.5 mg l?1). Further elongation of shoots (3.5 ± 0.06 cm) was achieved on MS medium supplemented with BA (1.0 mg l?1) and IAA (0.1 mg l?1). About 70 % of root induction occurred in half-strength MS medium supplemented with IBA (4.0 mg l?1) in 4–6 weeks. Further elongation of roots with average length (9.0 cm) was achieved in culture bottles containing vermiculite and ¼ strength MS salts. After their partial hardening in these bottles for 30 days they were transferred to pots containing a mixture of soil and vermicompost (1:1) for acclimatization. The acclimatized plantlets were established in the field successfully with 85 % survival rate.  相似文献

  • 1. Giant clams have been a sustainable resource for millennia, but unregulated harvesting has led to local extinctions within the Indo‐Pacific region. Giant clam mariculture can produce large numbers of juveniles for restocking wild populations where natural recruitment is low or absent.
  • 2. Singapore is surrounded by more than 60 small islands, many with fringing reefs. These reefs, however, experience increased turbidity and sedimentation resulting from massive coastal development projects and regular dredging of shipping lanes.
  • 3. Seven reefs off Singapore's southern islands were surveyed (9670 m2) for giant clams. Also, an experiment was conducted to determine the growth of Tridacna squamosa reared in aquaria under three light treatments: ~50% ambient photosynthetically active radiation (PAR); ~25% ambient PAR; and ~12% ambient PAR. Finally, 144 clams (T. squamosa) were transplanted to four reefs around Singapore to study survival and growth in a heavily impacted environment.
  • 4. A total of 23 adult clams from three species were found during the survey, representing a mean density of 0.24 per 100 m2. Most clams were found at Raffles Lighthouse, Singapore's ‘best’ reef. No juvenile clams were encountered. In the aquarium experiment, clam growth was significantly different among the three light treatments, with growth greatest in the ~50% ambient PAR treatment. Of the 144 transplanted clams, 116 (80.6%) were recovered after 7 months. All specimens had increased in size, with growth rates among reefs ranging from 3.3 mm month?1 (SD=1.3 mm) to 4.8 mm month?1 (SD=1.6 mm).
  • 5. Results suggest that, despite high levels of sedimentation and turbidity on Singapore's reefs, giant clams can survive and grow well. Restocking efforts using maricultured clams may be effective in enhancing the dwindling local populations. It is not clear, however, whether a self‐sustaining community can be established as high sedimentation may hinder larval settlement and survival.

Research carried out to assess the impact of open-pollinated Tomato leaf curl virus (ToLCV)-resistant tomatoes and hybrids on the livelihoods of resource-poor farmers in Southern India is described and discussed. Three high-yielding ToLCV-resistant tomato varieties were developed initially using conventional breeding and screening techniques involving inoculation by ToLCV-viruliferous whitefly, Bemisia tabaci. In 2003 and 2004, respectively, these varieties were released officially by the Karnataka State Seed Committee and the Indian Ministry of Agriculture through notification in the Gazette of India. From 2003 to 2005, eleven seed companies bought breeder seed of the ToLCV-resistant varieties and used them to begin breeding F1 hybrids from them. Socio-economic studies carried out to assess the benefits obtained from growing the ToLCV-resistant varieties found that farmers could gain up to 10 times the profit by growing the ToLCV-resistant varieties compared to the pre-existing ToLCV-susceptible varieties. Adoption of ToLCV-resistant tomatoes was also associated with reduced pesticide use. Extra income from tomato sales was prioritised by farmers to pay for children's education, better nutrition and medicines. In a joint effort with the commercial seed sector in India, a promotional field day was organised in 2007. As well as the three ToLCV-resistant varieties, 62 ToLCV-resistant hybrid tomatoes were exhibited during a farmer-field day by 17 commercial seed companies and several public institutes. Tomatoes with ToLCV-resistance are now grown widely in South India and seeds of the three open-pollinated varieties have been distributed to more than 12 countries. In a field experiment with four moisture regimes and eight nitrogen levels, the ratios between evapotranspiration and pan evaporation (EtEo) were low in the initial stages of crop growth and attained maximum values at 70–80% (20 and 40% available soil moisture depletion (ASMD)), 65% (60% ASMD) and 55% (80% ASMD) of the crop growth stage. Amongst nitrogen levels, the evapotranspiration ratio (ETR) was highest (3573) under no nitrogen and lowest (1312) with 180 kg N/ha. The 20% ASMD regime utilised less water (ETR= 1499 to produce a kilogram of grain than did the other moisture regimes. The lowest evapotranspiration ratio (914) was recorded with 20% ASMD and 180 kg N/ha in combination. The highest ETR (3954) was found with 60% ASMD and no nitrogen. An additive effect of nitrogen and moisture was found, in indicating that they can be substituted one for the other, when one of them becomes a constraint.  相似文献

ABSTRACT We evaluated combinations of two strains of plant growth-promoting rhizobacteria (PGPR) formulated with the carrier chitosan for the ability to induce growth promotion of tomato plants and resistance to infection by Cucumber mosaic virus (CMV). Each PGPR combination included GB03 (Bacillus subtilis) and one of the following PGPR strains: SE34 (B. pumilus), IN937a (B. amyloliquefaciens), IN937b (B. subtilis), INR7 (B. pumilus), or T4 (B. pumilus). The PGPR combinations formulated with chitosan are referred to as biopreparations. Tomato plants treated with each of the biopreparations appeared phenotypically and developmentally similar to nonbacterized control plants that were 10 days older (referred to as the older control). When plants were challenged with CMV, all plants in the biopreparation treatments and the older control treatment had significantly greater height, fresh weight, and flower and fruit numbers than that of plants in the CMV-inoculated same age control treatment. CMV disease severity ratings were significantly lower for biopreparation-treated and older control tomato plants than for that of same age control plants at 14 and 28 days postinoculation (dpi). CMV accumulation in young noninoculated leaves was significantly less for all biopreparation-treated plants and those in the older control than for the same age control plants at 14 dpi and for four of the five biopreparation treatments at 28 dpi. In those tomato plants shown to be infected, the amount of CMV in noninoculated leaves was significantly lower for three of the biopreparation treatments and the older control treatment at 14 dpi and biopreparation G/INR7 treatment at 28 dpi when compared with the control treatment. These data show that treatment of tomato plants with biopreparations results in significant enhancement of growth and protection against infection by CMV.  相似文献
