Magnetic resonance imaging (MRI) is commonly used to diagnose degenerative lumbosacral stenosis; however, studies show limited correlation between imaging and clinical signs. The purpose of this prospective observer agreement study was to use dynamic MRI of the lumbosacral (LS) spine of healthy dogs to determine reliable reference ranges. Twenty‐two healthy large breed dogs were prospectively enrolled. MRI of the LS spine was performed in T2‐weighted, T1‐weighted, and T2‐weighted SPACE sequences in neutral, flexed, and extended positions. Four observers performed image analyses. Measurements included LS angle, vertebral canal height and area, and LS foraminal areas. Ordinal categorical assessment of loss of fat signal in the foramina, LS compression, intervertebral disc (IVD) degeneration, spondylosis, and IVD protrusion was also performed. The majority of values were significantly larger in flexion versus neutral position, and significantly smaller in extension versus neutral position (P < .05). Subclinical compression and IVD protrusion was noted in a neutral position in 45% and 55% of dogs and in an extended position in 85% and 73% of dogs, respectively. Interobserver agreement was strong (intracluster correlation coefficient [ICC] > .5) except for the L7:LS vertebral canal area ratio (ICC ≤ .03). Intraobserver agreement was high (rho > .5) for all measurements except for the mid‐L6:LS vertebral canal height ratio (rho = .38). There was poor interobserver agreement for loss of fat signal in the foramina and evidence of compression. This study provides the groundwork for future studies using dynamic MRI to evaluate dogs with signs of clinical LS disease. 相似文献

In August 2018, Public Health England (PHE) was made aware of five probable cases of Shiga toxin‐producing Escherichia coli (STEC) O157:H7 among individuals reporting participation in a mud‐based obstacle race. An additional four cases, identified via routine whole‐genome sequencing, were subsequently linked to the same event. Two of the nine cases were due to secondary household transmission. Despite an agreement between the event organizers and the local authority, to ensure that all livestock were removed from the site 28 days before the event, sheep were observed grazing on some of the routes taken by the runners 2 days prior to the race taking place. A retrospective review of incidents reported to PHE between 2015 and 2018 identified 41 cases of gastroenteritis associated with muddy assault course events. Of these, 25 cases were due to infection with STEC O157:H7, of which all but one were associated with outbreaks. Due to the environment in which such events take place, it is impossible to entirely remove the risk of exposure to potentially pathogenic zoonoses. However, race organizers should ensure that livestock are removed from the course 28 days before the event. They should also ensure that participants are made aware of the risk of contracting gastrointestinal disease from the environment, and to stress the importance of hand hygiene post‐event and the risk of secondary transmission, particularly to children who are at risk of developing haemolytic uraemic syndrome. 相似文献

Schoolyards and suburban parks are two environments where active tick surveillance may inform local management approaches. Even in a state such as New York with a robust active tick surveillance programme operated by the state Department of Health, these settings are not routinely covered. The goal of this study was to highlight the importance of active surveillance for tick‐borne pathogens by describing their prevalence in ticks collected from schoolyards and suburban parks and to guide the use of integrated pest management in these settings. Tick dragging was performed in three regions of New York State: Long Island, the Lower Hudson Valley and the Capital Region. A total of 19 schoolyards and 32 parks were sampled. The location, habitat and weather at the time of tick collection were recorded. Ticks were speciated and tested for the presence of 17 pathogens with a novel application of nanoscale real‐time PCR. The causative agents of Lyme disease, anaplasmosis, babesiosis and Powassan virus disease were all detected from Ixodes scapularis in various sites throughout the capital region and south‐eastern counties of New York state. The most common agent detected was Borrelia burgdorferi, and coinfection rates were as high as 36%. This surveillance study also captured the first of the invasive Asian longhorned tick species, Haemaphysalis longicornis, in New York state (collected 2 June 2017). Results from this study highlight the importance of collaborative efforts and data sharing for improvement of surveillance for tick‐borne disease agents. 相似文献

Consumption of anthocyanins has been related with beneficial health effects. However, bioavailability studies have shown low concentration of anthocyanins in plasma and urine. In this study, we have investigated the bacterial-dependent metabolism of malvidin-3-glucoside, gallic acid and a mixture of anthocyanins using a pH-controlled, stirred, batch-culture fermentation system reflective of the distal human large intestine conditions. Most anthocyanins have disappeared after 5 h incubation while gallic acid remained constant through the first 5 h and was almost completely degraded following 24 h of fermentation. Incubation of malvidin-3-glucoside with fecal bacteria mainly resulted in the formation of syringic acid, while the mixture of anthocyanins resulted in formation of gallic, syringic and p-coumaric acids. All the anthocyanins tested enhanced significantly the growth of Bifidobacterium spp. and Lactobacillus-Enterococcus spp. These results suggest that anthocyanins and their metabolites may exert a positive modulation of the intestinal bacterial population. 相似文献

Consumer demand for cleaned squid generates a substantial amount of waste that must be properly disposed of, creating an economic
burden on processors. A potential solution to this problem involves converting squid by-products into an organic fertilizer,
for which there is growing demand. Because fertilizer application to lawns can increase the risk of nutrient contamination
of groundwater, we quantified leaching of NO3–N and PO4–P from perennial ryegrass turf (Lolium perenne L.) amended with two types of fertilizer: squid-based (SQ) and synthetic (SY). Field plots were established on an Enfield
silt loam, and liquid (L) and granular (G) fertilizer formulations of squid and synthetic fertilizers were applied at 0, 48,
146, and 292 kg N ha−1 year−1. Levels of NO3–N and PO4–P in soil pore water from a depth of 60 cm were determined periodically during the growing season in 2008 and 2009. Pore
water NO3–N levels were not significantly different among fertilizer type or formulation within an application rate throughout the
course of the study. The concentration of NO3–N remained below the maximum contaminant level (MCL) of 10 mg L−1 until midSeptember 2009, when values above the MCL were observed for SQG at all application rates, and for SYL at the high
application rate. Annual mass losses of NO3–N were below the estimated inputs (10 kg N ha−1 year−1) from atmospheric deposition except for the SQG and SYL treatments applied at 292 kg N ha−1 year−1, which had losses of 13.2 and 14.9 kg N ha−1 year−1, respectively. Pore water PO4–P levels ranged from 0 to 1.5 mg P L−1 and were not significantly different among fertilizer type or formulation within an application rate. Our results indicate
that N and P losses from turf amended with squid-based fertilizer do not differ from those amended with synthetic fertilizers
or unfertilized turf. Although organic in nature, squid-based fertilizer does not appear to be more—or less—environmentally

Faecal contamination of drinking water extracted from alluvial aquifers can lead to severe problems. River water infiltration can be a hazard for extraction wells located nearby, especially during high discharge events. The high dimensionality of river?Cgroundwater interaction and the many factors affecting bacterial survival and transport in groundwater make a simple assessment of actual water quality difficult. The identification of proxy indicators for river water infiltration and bacterial contamination is an important step in managing groundwater resources and hazard assessment. The time resolution of microbial monitoring studies is often too low to establish this relationship. A proxy-based approach in such highly dynamic systems requires in-depth knowledge of the relationship between the variable of interest, e.g. river water infiltration, and its proxy indicator. In this study, continuously recorded physico-chemical parameters (temperature, electrical conductivity, turbidity, spectral absorption coefficient, particle density) were compared to the counts for faecal indicator bacteria, Escherichia coli and Enterococcus sp. obtained from intermittent sampling. Sampling for faecal indicator bacteria was conducted on two temporal scales: (a) routine bi-weekly monitoring over a month and (b) intense (bi-hourly) event-based sampling over 3 days triggered by a high discharge event. Both sampling set-ups showed that the highest bacterial concentrations occurred in the river. E. coli and Enterococcus sp. concentrations decreased with time and length of flow path in the aquifer. The event-based sampling was able to demonstrate differences in bacterial removal between clusters of observation wells linked to aquifer composition. Although no individual proxy indicator for bacterial contamination could be established, it was shown that a combined approach based on time-series of physico-chemical parameters could be used to assess river water infiltration as a hazard for drinking water quality management. 相似文献

Laboratory confirmation methods are important in bovine cysticerosis diagnosis as other pathologies can result in morphologically similar lesions resulting in false identifications. We developed a probe-based real-time PCR assay to identify Taenia saginata in suspect cysts encountered at meat inspection and compared its use with the traditional method of identification, histology, as well as a published nested PCR. The assay simultaneously detects T. saginata DNA and a bovine internal control using the cytochrome c oxidase subunit 1 gene of each species and shows specificity against parasites causing lesions morphologically similar to those of T. saginata. The assay was sufficiently sensitive to detect 1 fg (Ct 35.09 ± 0.95) of target DNA using serially-diluted plasmid DNA in reactions spiked with bovine DNA as well as in all viable and caseated positive control cysts. A loss in PCR sensitivity was observed with increasing cyst degeneration as seen in other molecular methods. In comparison to histology, the assay offered greater sensitivity and accuracy with 10/19 (53%) T. saginata positives detected by real-time PCR and none by histology. When the results were compared with the reference PCR, the assay was less sensitive but offered advantages of faster turnaround times and reduced contamination risk. Cyclodextrins improve post-thaw viability and motility of semen as well as mediate cholesterol efflux and subsequent acrosome reaction in spermatozoa from several species. The objectives of this study were: (a) to assess the effect of prefreeze addition of 60 mM hydroxypropyl-β-cyclodextrin (β-CD) on post-thaw viability and motility of jack and stallion semen cryopreserved in ethylene glycol-based freezing extenders containing 5% or 20% (v/v) egg yolk (LEY and HEY, respectively), and (b) to evaluate the ability of 1 μM calcium ionophore A23187 and/or 60 mM β-CD to induce acrosome reaction in thawed jack and stallion spermatozoa. Post-thaw motility of spermatozoa cryopreserved in HEY was higher (P < .05) for jack but lower (P < .05) for stallion spermatozoa when compared with LEY. Jack and stallion spermatozoa both exhibited higher (P < .05) motility when cryopreserved in 60 mM β-CD than without β-CD. Curvilinear velocity was faster (P < .05) for jack and stallion spermatozoa cryopreserved in LEY than in HEY. A treatment × time interaction affected (P < .05) the proportion of spermatozoa that underwent acrosome reaction. Post-thaw incubation of jack and stallion spermatozoa with β-CD for 90 minutes induced acrosome reaction in 85% and 22% of viable sperm cells, respectively; however, only 32% of jack and 8% of stallion spermatozoa incubated with calcium ionophore underwent acrosome reaction. This study is the first to evaluate the effect of β-CD (not loaded with cholesterol) on jack semen cryopreservation, and results reveal that β-CD may be a useful tool to enhance semen cryopreservation and to induce post-thaw acrosome reaction in jack spermatozoa. 相似文献

The digestible energy and apparent nutrient digestibility coefficients of common diet ingredients were determined for pacu Piaractus brachypomus (370.21 ± 17.56 g). Fish were fed with pelleted practical diets to apparent satiation and the feces were collected by siphoning. The digestibility value for each ingredient was determined by comparison of the digestibility of a test diet with a reference diet (24.5% crude protein and 1% chromic oxide). The digestible energy values of soybean meal (SBM), fish meal (FM), corn (CN), and wheat bran (WB) were 2382. 3826, 3353, and 1784 kcal/kg, respectively. The apparent dry matter digestibility coefficients were 83.72, 90.14, 89.13, and 82.05% for SBM, FM, CN, and WB, respectively. The apparent crude protein digestibility coefficients were 75.88, 90.49, 85.06, and 61.62% for SBM. FM, CN, and WB, respectively. The apparent lipid digestibility coefficients were 63.03, 77.00, 83.01, and 82.45% for SBM, FM, CN, and WB, respectively. The digestibility of protein, lipid, and energy from SBM were somewhat low compared to values for other warmwater omnivorous fishes, but similar to values reported for pacu-caranha P. mesopotamicus. Otherwise, the nutrient and energy availability of the ingredients to P. brachypomus was similar to that of other fish. This information will be useful in formulating nutritious, economical diets for pacus. 相似文献
