Crude ligninolytic enzyme extracts from Phanerochaete chrysosporium fungi were applied to sugarcane bagasse, prior to thermomechanical (TMP) and chemithermomechanical pulping (CTMP), and their properties were compared with the normal TMP and CTMP and also with TMP and CTMP pretreated with Ceriporiopsis subvermispora and P. chrysosporium fungi. The sugarcane bagasse was impregnated with the crude enzyme extract containing lignin peroxidase (LiP), manganese peroxidase (MnP), and laccase (Lac). The results show that pretreatment with enzyme crude extract is an advantageous way to produce TMP and CTMP from sugarcane bagasse, as compared with only fungal pretreatment. Enzymatic pretreatments need only hours to enhance pulping and paper properties, compared with the weeks necessary for fungal treatments. Higher pulp yields were obtained compared with the fungal pretreatments. Enzymatic pretreatment reduced the energy consumption in a proportion similar to that of C. subvermispora fungal pretreatment and increased the pulp tensile index compared with the normal TMP and CTMP pulps, although the tensile strength was somewhat lower than that for pulps from C. subvermispora fungal pretreatment before CTMP processing. An advantage of enzymatic pretreatment is that brightness is increased compared with normal TMP and CTMP processes, whereas fungal pretreatments reduce the brightness.  相似文献

A simple method for simultaneously determining cell-associated Ca and Zn in Caco-2 cells is described. Calcium and zinc uptake was measured via radioisotopes (45)Ca and (65)Zn. Preliminary studies revealed that (65)Zn, a positron (beta(+)) and gamma emitter, contributed to (45)Ca counts in a liquid scintillation counter (LSC). However, (45)Ca, being a true beta emitter, did not contribute to the counts in a gamma counter (gammaC). To differentiate the counts of (45)Ca from those of (65)Zn, first a (65)Zn-labeled cell suspension was read in a gammaC and an LSC, thus obtaining the relationship between the radioactive counts obtained from the gammaC and LSC. This information defined the linear relationship between gammaC (65)Zn counts per minute (CPM) and LSC (65)Zn CPM. Because the (45)Ca and (65)Zn counts obtained in the LSC are additive, giving total LSC CPM, the value of LSC (45)Ca CPM was obtained by subtracting LSC (65)Zn CPM from total LSC CPM for the dual-labeled cell sample, obtaining then LSC (45)Ca CPM. To determine the absolute activity or disintegrations per minute (DPM) of each isotope in the dual-labeled sample, the linear relationship between DPM and CPM was determined for each isotope. The method is simple and straightforward for the determination of (45)Ca counts from a sample also containing (65)Zn, using gamma and liquid scintillation counters.  相似文献

Changes in the volatile composition during maturation and artificial ripening of yellow-fleshed nectarines were investigated by means of GC-FID and GC-MS over three years (1999-2001). Unripe and commercially ripe fruits were placed in ripening chambers until complete ripening and compared to tree-ripe nectarines. Firmness, weight, soluble solids (SS), titratable acidity (TA), SS/TA ratio, organic acids, and sugars were also determined. Furthermore, the tree-ripe and artificially ripened yellow-fleshed nectarines were subjected to sensory analysis by a taste panel. In 2001, a similar experiment on white-fleshed nectarines (cv. Vermeil) was performed to compare with the results obtained on yellow-fleshed nectarines. Levels of volatiles compounds, in particular, lactones and C(13) norisoprenoids, were found to be the same or higher in the artificially ripened nectarines compared with the tree-ripe nectarines. In addition, no significant difference was observed for sweetness, sourness, and the intensities of "peach odors" or "peach aroma" in the sensory analysis between the tree-ripe samples and the artificially ripened nectarines. However, in the latter the levels of SS and the SS/TA ratio were very close to those observed in the initial unripe samples and significantly lower than those observed in tree-ripe nectarines.  相似文献

Summary Six wild barley (Hordeum spontaneum) accessions, from a diverse range of habitats, and two spring-cultivated barleys, were examined for variation in durations of development phases. The durations of the leaf initiation and spikelet initiation phases were longer and spikelet growth phases shorter, in wild than in cultivated barley. Across all wild and cultivated barleys the rate and duration of spikelet initiation were negatively correlated, but neither was related to the number of spikelets per spike. The number of spikelets was positively correlated with the number of leaves and the ratio of the number of spikelets to the number of leaves declined with increasing time to anthesis, indicating that each successive leaf was associated with a diminishing increase in the number of spikelets. The duration of culm elongation and final culm length were shorter in accessions of cultivated barley compared with wild barley. Trimethylamine oxide (TMAO), which has previously not been known to occur in significant amounts in the blood of marine teleosts, rose to concentrations of approximately 50 mM in the blood of winter-acclimatized rainbow smelt, Osmerus mordax. Urea also increased in the blood of cold-acclimatized smelt, and, with TMAO, contributed significantly to the winter freezing point depression. TMAO and urea also varied seasonally in muscle and liver tissues. TMAO and urea appeared to be reabsorbed from the urine. Losses of TMAO and urea from the head region of the fish, where most of the losses appeared to occur, were approximately 9 μmol and 8 μmol 100 g−1 h−1, respectively. Despite the effluxes, TMAO and urea levels in both the blood and muscle either increased or were maintained in starved, cold-acclimated fish, suggesting that they were synthesized in response to cold temperature. TMAO was also found in the blood of some other cold-hardy teleosts.  相似文献

The purpose of this work was to determine the degree of oil retention from an oil spill area in view of natural physical weathering, and to evaluate the compositional changes of the natural oil retained in the soil material. The percentage of oil retained over the 1 yr period of weathering was determined at various intervals, and the compositional changes were studied by G.L.C. and I.R. The results of G.L.C. indicated the loss of low molecular weight n-paraffins, and that of I.R. indicated the formation of sulfones and ether groups as a result of oxidation reaction due to natural exposure. It was established that up to 4% oil was retained in the soil material after 6 mo of exposure, and that about 3.5% remained after a year.  相似文献

Dynamic simulations models may enable for farmers the evaluation of crop and soil management strategies, or may trigger crop and soil management strategies if they are used as warning systems, e.g. for drought risks and for nutrients shortage. Predictions by simulation models may differ from field observations for a variety of reasons, and such deviations can be revealed instantly by traditional or by new field monitoring techniques. The objective of this study was to improve simulation results by integrating remote sensing observations during the growing season in the simulation (i.e. run-time calibration). The Rotask 1.0 simulation model was used as it simulates daily interactions between climate (radiation temperature, vapour pressure, wind speed, precipitation), soils (water holding capacities, soil organic matter dynamics, evaporation) and crops (light interception, dry matter production, nitrogen uptake, transpiration). Various run-time calibration scenarios for replacing simulated values by remotely observed values were tested. For a number of times in the growing season, simulated values of leaf area index (LAI) and canopy nitrogen contents were replaced with values estimated from remote sensing. Field experiments were carried out in the Netherlands in 1997 (validation) and 1998 (calibration) with potato variety Bintje. Destructive field samplings were performed to follow LAI and canopy nitrogen development in the growing season. Remote sensing observations at canopy level were taken by CropScan™ equipment, covering the electromagnetic spectrum between 460–810 nm in eight spectral bands. LAI and canopy nitrogen were monitored at various moments throughout the growing season by relating them with vegetation indices (VI) that were calculated from the combination of specific remote sensing bands. The results of this study show that run-time calibration of mechanistic simulation models may enhance simulation accuracy, depending on the method how additional information is integrated. It is advised to synchronize dry matter balances and internal nitrogen balances in accordance with adjustments to observed calibration variables (in this case LAI and canopy nitrogen content). It is shown an integrated approach follows the actual crop–soil system more closely, which is helpful for specific crop management and precision agriculture in general. Run-time calibration with variables that can be estimated from remote sensing observations gives more accurate simulation results of variables that can not be observed directly, e.g. the evolution of soil inorganic nitrogen contents. High frequencies of remote sensing observations and interpolation in between them, allow reconstructing the evolution of LAI and canopy nitrogen contents to be integrated in the simulation, thereby increasing simulation accuracy of other model variables.  相似文献

The particular behavior during wine aging of fermentative branched fatty acid ethyl esters, related to yeast nitrogen metabolism, compared that of their straight-chain analogues, related to yeast lipid metabolism, was first checked in 1-5 year aged Muscadet wines. Quantitative SIDA measurements showed that the levels of the former increased, whereas those of the latter decreased. Then, three hypothetical pathways suggested in the literature to explain these variations of branched esters were investigated. Two Muscadet and Sylvaner wines were spiked with levels of deuterated isobutanoic acid and its ethyl ester, similar to those of their natural analogues, then they were submitted to model aging. Quantitative SIDA measurements on the formation of these natural and labeled ethyl esters from the corresponding acids revealed that the behavior of the natural and labeled compounds were similar. The acid levels were much higher than the ester levels in the initial young wine, and a significant upward trend of their esterification ratios to those of the acid-ester equilibrium was observed with aging. Thus, this equilibrium proved to be the most effective in generating the branched fatty acid ethyl esters during wine aging. In contrast, the formation of these acids by Strecker-type degradation of wine amino acids in the conditions of the model aging or by hydrolysis of their glycoconjugates proved to be ineffective.  相似文献
