The selective effect that refrigeration exerts on the autochthonous microbiota of the eastern oyster was investigated using culture-independent methods. Oysters from two different locations along the Gulf Coast of Mexico were analyzed concurrently during 2-week refrigeration. Ribosomal intergenic sequence analysis (RISA) generated complex bacterial community fingerprints that provided a strong association between RISA-defined clusters and sample date. Denaturing gradient gel electrophoresis (DGGE) profiles obtained with universal primers did not reflect any association based on date or geographic origin of the samples. During the storage period, Vibrio parahaemolyticus was replaced, at least in overall abundance, with other nonpathogenic Vibrio species better adapted to cold temperatures. This study shows that refrigeration itself does not limit the growth of the bacterial community nor reduces its richness.  相似文献

In Europe, the English yew species (Taxus baccata L.) is endangered. Intensive human land-use, including forest management, has caused a decrease of the yew populations all over Europe. In Austria, gene conservation forests are used for the in situ conservation of populations of this rare tree species by silvicultural treatments. In order to improve the conservation management in these gene conservation forests, this study addresses the relation between competition and viability of yew populations through the use of structural diversity indices. The structural indices, which include mingling, tree–tree distance, diameter, and tree height differentiation, were determined for a structural group of four trees as well as the neighbouring trees of the male and female yews at the monitoring plots on a regular grid in three gene conservation forests. Although the three study sites provided quite different environmental conditions for English yew, the vitality of each individual yew was influenced by the inter-specific competition of the neighbouring tree species at all sites. Low vitality was associated with a small mean distance to neighbours and large tree height differentiation. In conclusion, we suggest that a combination of different structural indicators is needed for an integrative assessment of conservation status in the gene conservation forests. This would help improve the evaluation of the impact management has on yew population viability.  相似文献

Extracts of the Floridian marine cyanobacterium Lyngbya cf. confervoides were found to deter feeding by reef fish and sea urchins (Diadema antillarum). This antifeedant activity may be a reflection of the secondary metabolite content, known to be comprised of many serine protease inhibitors. Further chemical and NMR spectroscopic investigation led us to isolate and structurally characterize a new serine protease inhibitor 1 that is formally derived from an intramolecular condensation of largamide D (2). The cyclization resulted in diminished activity, but to different extents against two serine proteases tested. This finding suggests that cyanobacteria can endogenously modulate the activity of their protease inhibitors.  相似文献

The defence responses of grapefruit and lemon to treatment with pomegranate extract (PGE) were investigated. PGE, an alcoholic extract from pomegranate peel, was recently proposed as a means of effective alternative control against postharvest rots. In in vivo experiments, a significant reduction of rots caused by Penicillium digitatum and P. italicum was achieved on artificially inoculated fruits without direct contact between PGE and pathogens. On lemons both pathogens were completely inhibited by PGE at 12 g dry matter L?1 applied 12 and 24 h before the pathogen but a significant reduction of rots was also achieved by inoculating the pathogen immediately after PGE (0 h), indicating a very quick activation of defence responses. Lower, but significant, reductions were also obtained on grapefruits. An increase in reactive oxygen species (ROS) activity, reaching its peak after 24 h, was observed, in agreement with in vivo efficacy trials. Similarly, the expression of five genes involved in activation of defence responses in plants (CHI, CHS, MAPK, MAPKK and PAL) increased following PGE application. Based on the results of the present study, the high efficacy demonstrated for PGE in previous studies can be partially attributed to the induction of resistance in host tissues.  相似文献

One-day-old broiler chicks received cecal microflora (CM) cultured under aerobic, anaerobic conditions, or both (mixed) and were then infected with Salmonella Enteritidis, in order to compare the efficacy of these different types of culture in terms of the number of chicks infected, cecal colonization and faecal excretion of the challenging bacteria. Regardless of culture type, CM always led to a smaller number of S. Enteritidis for any of the parameters studied compared to untreated chicks. Aerobic CM demonstrated better efficacy in reducing the number of infected chicks and cecal colonization by S. Enteritidis, followed by mixed CM. No difference was observed in faecal excretion of S. Enteritidis between the chicks that received different types of CM culture.  相似文献

The efficacy of meso-dimercaptosuccinic acid (DMSA) (succimer) in treating avian lead intoxication was studied in a retrospective, nonrandomized, longitudinal study. Nineteen birds with moderate to high blood lead concentration and neurologic signs compatible with lead toxicity were treated with DMSA (30 mg/kg p.o., b.i.d.; n = 15) for a minimum of 7 days. In cases with severe neurologic signs, DMSA was supplemented with a single dose of edetate calcium disodium (<50.0 mg/kg of body weight i.m.; n = 4). Blood lead concentrations were measured two or more times (before and after treatment). Median blood lead concentration decreased (87%), neurologic signs were resolved, and there were no apparent adverse secondary effects.  相似文献

Thiafentanil oxalate, previously known as A-3080, is a synthetic opioid used for chemical immobilization of a variety of nondomestic hoofstock species. This study compared the combination of thiafentanil oxalate, medetomidine, and ketamine (TMK; 0.09 +/- 0.02 mg/kg, 0.01 +/- 0.003 mg/kg, and 1.36 +/- 0.33 mg/kg, respectively) with the combination of medetomidine and ketamine (MK; 0.09 +/- 0.02 mg/kg and 3.48 +/- 0.55 mg/kg, respectively) for anesthetization of 17 captive male axis deer (Axis axis) for vasectomy. Nine deer received TMK and eight deer received MK via projectile syringe during the months of January and February, 2005. Mean induction and arousal times, vital signs, and arterial blood gas values were monitored and compared. All animals received supplemental oxygen during the surgical procedure. Animals receiving TMK were reversed with naltrexone (100 mg/mg thiafentanil) and atipamazole (5 mg/mg medetomidine). Animals receiving MK were reversed with atipamazole (5 mg/mg medetomidine). Two MK animals and three TMK animals required supplementation with ketamine i.v. immediately upon handling. Six of the nine animals immobilized with TMK required intubation for positive-pressure ventilation. Two of these six animals also required isoflurane to maintain anesthesia. Mean induction time was 3.5 +/- 2.0 min in the TMK group, and 9.8 +/- 6.7 min in the MK group. Despite shorter mean induction times, animals anesthetized with TMK experienced unpredictable inductions, apnea, muscle rigidity, limb movement, and significant respiratory and metabolic lactic acidosis. MK resulted in smoother inductions, better respiratory function, and less adverse metabolic disturbances, and thus was considered superior to TMK for anesthesia in captive axis deer at the dosages tested.  相似文献

Two adult Wied's marmosets (Callithrix kuhlii) presented with jaundice, anemia, and weight loss. Death of one individual was attributed to renal tubular necrosis; liver and kidney were positive for Leptospira antigen by immunohistochemical (IHC) staining. The second animal was negative for antigen by IHC staining, but serologically positive for Leptospira borgpetersenii serovar ballum with an eightfold titer increase in paired samples, and was euthanized because of unresponsiveness to treatment. Environmental contamination by mice was suspected as the Leptospira source.  相似文献

To determine the inhibitory capacity of lactic acid bacteria due to the action of antagonistic substances, we tested 474 isolates of Lactobacillus from the crop and cecum of chickens against gram-positive and gram-negative indicator microorganisms by the spot-on-the-lawn and well-diffusion antagonism methods. Of the 474 isolates, 265 demonstrated antimicrobial activity against the indicator microorganisms. Isolates identified as L. reuteri, L. salivarius, or Lactobacillus spp. inhibited Enterococcus faecalis, E. faecium, Listeria monocytogenes, and Salmonella spp. but not L. casei, L. delbrueckii, L. fermentum, or L. helveticus by the well-diffusion simultaneous antagonism method under anaerobic incubation conditions. The antagonistic substances produced by some of the Lactobacillus isolates were inactivated after treatment by proteolytic enzymes, which suggested that the substances could be antimicrobial peptides or bacteriocins.  相似文献
