The purpose of this report is to discuss the use of topical 1% 5‐fluorouracil as a sole therapy for canine corneal squamous cell carcinoma (SCC). A 12‐year‐old castrated male pug was evaluated for a well‐demarcated, central, 3 mm in diameter, pale pink, raised, right corneal mass. An incisional biopsy was obtained using a #64 beaver blade after topical anesthesia and without sedation. A definitive diagnosis of corneal SCC was obtained after histopathologic evaluation of the biopsy. Topical 1% 5‐fluorouracil ointment was applied to the right eye four times daily for 2 weeks followed by no treatment for 2 weeks, then treatment again twice daily for 2 weeks. The cornea remained free of recurrence 10 months after cessation of treatment. In dogs affected with corneal SCC, topical 1% 5‐fluorouracil monotherapy may be a viable and cost‐effective treatment option with minimal side effects. This chemotherapy agent may also have an effect on corneal pigmentation. Chronic cyclosporine therapy did not contribute to the pathogenesis of corneal SCC in the case described.  相似文献

The objectives of this study were to investigate the influence of ram age on structural and functional competence of frozen–thawed spermatozoa and to test the hypothesis that increasing number of sperm bound to the zona pellucida in vitro was associated with decreasing in vivo fertility of frozen semen. Rams were allocated into two groups. Each group consisted of five rams aged either 1–2 years (young) or 4–5 years (mature). Three successive ejaculates were collected from each ram using an artificial vagina. Only ejaculates of ≥ 2.5 × 109 sperm/ml and 80% sperm progressive motility were pooled per ram, diluted with Bioxcell® medium and frozen in 0.25 ml straws. The end points of post‐thawing semen evaluation were computer‐assisted cell motility analysis, sperm capacitation (chlortetracycline assay), simultaneous assessment of plasma membrane integrity, mitochondrial membrane potential and condensation status of nucleus, per‐cell analysis of lipid peroxidation using C11‐BODIPY581/591, sperm‐hemizona binding (HZB) ability and sperm fertility after laparoscopic insemination of ewes (n = 114) in the progestagen‐synchronized oestrus. The results showed that mature rams had significantly lower values of sperm hyperactivated motility and peroxidized sperm, higher percentages of live non‐capacitated sperm and sperm cells with intact plasma membrane, functional mitochondria and condensed chromatin, as well as, greater lambing rate and ewe prolificacy. Sperm HZB binding ability was higher (p < 0.05) for young than for mature rams. Significant correlations were found between number of spermatozoa bound to the zona pellucida and semen fertility (r = ?0.63 to ?0.71). In conclusion, mature rams have better semen quality and in vivo fertility than young rams. Cryocapacitation can be involved in decreasing ram semen fertility as evidenced by the high number of spermatozoa bound to the zona pellucida in vitro.  相似文献

Goal, Scope and Background  Temperature and soil moisture content are important environmental variables in bioremediation technologies. Optimizing these variables in-situ would enhance and maintain remediation of hazardous wastes during cold winter seasons or in cold regions and may lead to reduced maintenance and/or cost. The effect of elevated temperature and soil moisture on bioremediation efficiency was investigated using a laboratory mesocosm approach. Selected polycyclic aromatic hydrocarbons (PAHs) and phenols degradation in contaminated flooded soils, commonly found in Superfund sites situated in coastal plains sediments/soils, were evaluated in the mesocosms. Material and Methods  Four laboratory mesocosm treatments in triplicate simulating in-situ bioremediation of contaminated site soils using an immobilized microbe bioreactor system, i.e., bioplug, were established to evaluate temperature effects. Elevated temperature treatments of site soils with and without contaminant-specific microorganisms were established at a temperature of 42±2°C. Similarly, treatment of site soils with and without contaminant-specific microorganisms were established at an ambient temperature of 21±1°C. Composite samples were analyzed for selected PAHs and chlorinated phenols to determine rates of mineralization and overall remediation efficiency for different temperature regimes. Results  Mesocosm studies indicated that the high temperature inoculated treatment demonstrated a significant reduction in mean total PAHs and total phenols with a kinetic rate (KR) of 76±13 ng g−1 d−1 in 49 days (approximately 84% reduction; p<0.01) The KR for low temperature inoculated treatment was 54±1 ng g−1 d−1 in 49 days (approximately 66% reduction; p<0.01). High temperature non-inoculated mesocosms exhibited significant mineralization of all constituents with KR of 15±6 ng g−1 d−1 (approximately 65% reduction; p<0.01) in 49d compared to 54% reduction for low temperature non-inoculated treatment with KR of 12±3 ng g−1 d−1 (p=0.1794). Phenol compounds in inoculated treatments were also significantly reduced (65%, p<0.01) at elevated temperatures compared to ambient (52%, p<0.01). Discussion  Increased bioavailability and desorption were noted for elevated temperature and moisture in the soil laboratory mesocosms simulating a field in situ remediation protocol. This protocol employing the application of immobilized microflora indicated that in situ systems provide an economical advantage if optimal elevated temperature and moisture are controlled properly. Results also suggested that temperature and moisture optimization needs to be combined with efficient nutrients delivery systems for impacted soils/sediments. Conclusions  The study demonstrated that temperature and soil moisture contents are important factors in the success of in-situ bioremediation techniques at hazardous waste sites situated in a coastal zone. Kinetic rates were significantly enhanced to remediate known recalcitrant compounds (PAHs and phenols) in aged soil. Recommendations and Perspectives  The placement of a preferred microbial consortia such as an immobilized microbial population in an entrained bioreactor, i.e., bioplug, can significantly reduce constituents of concern in a timely manner for contaminated soils/sediments. However, frequent monitoring of the soil temperature, moisture content, nutrient level, and dissolved oxygen is necessary to achieve predictable kinetic rates of mineralization. The California horn shark (Heterodontus francisci), instrumented with a pericardial catheter and a ventral-aortic flow probe, was studied to determine the effect of complete pericardial chamber evacuation on the time course for restitution of pericardial fluid-volume and pressure, and the effects of both fluid removal and its restitution on cardiac output. Prior to evacuation, pericardial pressure was –0.02±0.02 kPa, and cardiac output was 18±2 ml min–1 kg–1. Evacuation reduced pericardial pressure to –0.73±0.14 kPa, and increased cardiac output to 23±4 ml min–1 kg–1. The time course for restoration of post-evacuation pressure is described by a non-linear asymptotic function. A large percentage of the pericardial pressure and volume recovery occurred within the first hour, while, complete restoration of pre-withdrawal conditions required about 11 h. Pericardial pressure-volume relationships, determined by incremental infusion of small volumes of elasmobranch saline into the pericardium, confirm previous findings that the operating pericardial pressure in the horn shark is at or near ambient pressure and that both pericardial fluid volume and cardiac stroke volume influence horn shark pericardial pressure.  相似文献

Compositional information for lignins in food is rare and concentrated on cereal grains and brans. As lignins are suspected to have important health roles in the dietary fiber complex, the confusing current information derived from nonspecific lignin determination methods needs to be augmented by diagnostic structural studies. For this study, lignin fractions were isolated from kiwi, pear, rhubarb, and, for comparison, wheat bran insoluble dietary fiber. Clean pear and kiwi lignin isolates allowed for substantive structural profiling, but it is suggested that the significance of lignin in wheat has been overestimated by reliance on nonspecific analytical methods. Volume integration of NMR contours in two-dimensional (13)C-(1)H correlation spectra shows that pear and wheat lignins have comparable guaiacyl and syringyl contributions and that kiwi lignins are particularly guaiacyl-rich (approximately 94% guaiacyl) and suggest that rhubarb lignins, which could not be isolated from contaminating materials, are as syringyl-rich (approximately 96% syringyl) as lignins from any known natural or transgenic fiber source. Typical lignin structures, including those newly NMR-validated (glycerols, spirodienones, and dibenzodioxocins), and resinols implicated as possible mammalian lignan precursors in the gut are demonstrated via their NMR correlation spectra in the fruit and vegetable samples. A novel putative benzodioxane structure appears to be associated with the kiwi lignin. It is concluded that the fruits and vegetables examined contain authentic lignins and that the detailed structural analysis exposes limitations of currently accepted analytical methods.  相似文献

The growth of plants in nutrient solutions is an invaluable tool for mineral nutrition studies. Successful growth of plants in nutrient solutions require special attention and consideration. Details and helpful ideas for the growth of plants in nutrient solutions are often omitted in publications where techniques like these are used. The objective of this report is to focus on some of the concerns, successes, experiences, and problems noted for the growth of sorghum and corn in nutrient solutions. Topics discussed are nutrient solution composition, pH of nutrient solutions, phosphorus concentrations, sources of Fe in solutions, plus several suggestions and comments for successful growth of sorghum and corn in nutrient solutions.  相似文献

Phosphorus (P) forms were sequentially extracted from peat derived soils (Eutric Histosols and Gleysols) at eight sites in Saxony‐Anhalt (Germany) to disclose general differences in P pools between mineral and organic soils and to investigate effects of peat humification and oxidation in conjunction with land use and soil management on the P status of soils. Overall 29 samples providing a wide variety of basic chemical properties were subjected to the Hedley fractionation. The Histosol topsoils contained more total P (Pt) (1345 ± 666 mg kg—1) than the Gleysol topsoils (648 ± 237 mg kg—1). The predominant extractable fractions were H2SO4‐P (36—63 % of Pt) in calcareous and NaOH‐Po (0—46 % of Pt) in non‐calcareous Histosols. These soils had large pools of residual P (13—93 % of Pt). Larger contents and proportions of Po and of labile P fractions generally distinguished organic from mineral soils. Regression analyses indicated that poorly crystalline pedogenic oxides and organic matter were binding partners for extractable and non‐extractable P. Intensive management that promotes peat humification and oxidation results in disproportional enrichments of labile P fractions (resin‐P, NaHCO3‐Pi, and NaHCO3‐Po). These changes in P chemistry must be considered for a sustainable management of landscapes with Histosols and associated peat derived soils.  相似文献

Peripheral blood smears from four adult reindeer (Rangifer tarandus) were examined after staining with Romanowsky's stain and cytochemical stains, including alpha-napthyl butyrate esterase (alpha-NBE), Sudan black B (SBB), chloroacetate esterase (CAE) and alkaline phosphatase (ALP). Romanowsky-stained eosinophils, neutrophils, lymphocytes and monocytes resembled those of cattle, sheep and goats. Basophils had two different staining patterns with Romanowsky's stain. Basophils that we termed "grey basophils" were similar in appearance to grey eosinophils in Greyhound dogs, with medium blue-grey to lavender-grey cytoplasm containing varying numbers of clear vacuoles or granules and variable numbers of small, intensely basophilic, perinuclear granules. The second basophil staining pattern was more typical of ruminant basophils, with uniform, pale to dark basophilic cytoplasmic granules. Basophils stained positive for alpha-NBE, SBB, CAE, and ALP. Eosinophils stained positive for SBB, and were negative for alpha-NBE, CAE, and ALP. Neutrophils were negative for SBB, CAE, and ALP. Monocytes stained positive for alpha-NBE, were rarely positive for CAE and SBB, and were negative for ALP. Transmission electron microscopy revealed matrix within all granulocytes granules, including those of basophils.  相似文献

The potential of a new kind of light-induced pesticide action was evaluated for a broad variety (32) of natural photosensitizers and their thione derivatives. The latter were synthesized to shift the absorption spectra towards the visible region and to increase the triplet and singlet oxygen quantum yields. Qualitative and quantitative evaluation of growth inhibition of Fusarium culmorum (F. G. Smith) Sacc. produced by these photosensitizers under UVA and visible light was performed on silica gel plates and in liquid medium. The results show that the phototoxicity per excited molecule of the thione derivatives using UVA light was similar to that of their parent natural compounds. On the other hand, only the thione derivatives were photoactive under visible light irradiation. These compounds show encouraging levels of phototoxicity against F. culmorum, both in liquid culture and on silica plates, and may have potential for use as photoactive pesticides.  相似文献
