This study determined the lutein level in various green leafy vegetables (GLVs) and the influence of olive and sunflower oils on the postprandial plasma and eye response of dietary lutein in adult rats, previously induced with lutein depletion (LD). Fresh GLVs (n = 35) were assessed for lutein (L) and its isomer zeaxanthin (Z) levels by high-performance liquid chromatography and liquid chromatography-mass spectrometry. Among GLVs analyzed, Commelina benghalensis L. contained a higher level of L + Z (183 mg/100 g dry wt) and was used as a lutein source for feeding studies. Rats with LD were fed a diet containing powdered C. benghalensis (2.69 mg lutein/kg diet) with either olive oil (OO group), sunflower oil (SFO group), or groundnut oil (GNO group) for 16 days. The L + Z levels of the OO group were markedly (p > 0.05) higher than those of SFO and GNO groups, in plasma (37.6 and 40.9%) and eyes (22.7 and 30.8%), respectively. These results suggest that oleic acid or OO can be used as a suitable fat source to modulate the absorption of dietary lutein to manage age-related macular degeneration.  相似文献

Fusarium wilt (FW) and Sterility mosaic disease (SMD) are important biotic constraints to pigeonpea production worldwide. Host plant resistance is the most durable and economical way to manage these diseases. A pigeonpea mini-core collection consisting of 146 germplasm accessions developed from a core collection of 1290 accessions from 53 countries was evaluated to identify sources of resistance to FW and SMD under artificial field epiphytotic conditions during 2007–08 and 2008–09 crop seasons. Resistant sources identified in the field were confirmed in the greenhouse using a root dip screening technique for FW and a leaf stapling technique for SMD. Six accessions (originated from India and Italy were found resistant to FW (<10% mean disease incidence). High level of resistance to SMD was found in 24 accessions (mean incidence <10%). These SMD resistant accessions originated from India, Italy, Kenya, Nepal, Nigeria, Philippines and United Kingdom. Combined resistance to FW and SMD was found in five accessions (ICPs 6739, 8860, 11015, 13304 and 14819). These diverse accessions that are resistant to FW or SMD will be useful to the pigeonpea resistance breeding program.  相似文献

Parathion hydrolase (PH), also known as organophosphorus acid anhydrase, hydrolyses the triester linkage found in organophosphates including organophosphate pesticides and the nerve gas sarin. The enzyme is reported to be membrane-associated and the immature protein has a signal sequence of 29 amino acids. In experiments designed to examine the post-translational processing of the enzyme and to assess the distribution of the precursor and mature forms of the protein, we induced expression of the Flavobacterium balustinum PH structural gene, opd, in Escherichia coli strain BL21. Western blotting revealed that the induced PH was predominantly membrane-associated in E. coli but a protein band equivalent in size to mature PH was also found to be induced specifically in periplasmic fractions. This periplasmic protein was not PH, as it did not cross-react in Western blots, and N-terminal sequencing of the induced protein showed it to have 100% homology to the outer membrane protein OmpF.  相似文献

1. A study was conducted to evaluate the individual and combined effects of aflatoxin B1 (AF), ochratoxin A (OA) and T-2 toxin (T-2) on performance, organ morphology serum biochemistry and haematology of broiler chickens and the efficacy of esterified-glucomannan (E-GM), a cell wall derivative of Saccharomyces cerevisiae1026 in their counteraction. 2. Two dietary inclusion rates of AF (0 and 0.3 mg/kg), OA (0 and 2 mg/kg), T-2 (0 and 3 mg/kg) and E-GM (0 and 1 g/kg) were tested in a 2 x 2 x 2 x 2 factorial manner on a total of 960 broiler chickens from 1 to 35 d of age in an open sided deep litter pen house. 3. Body weight and food intake were depressed by all the mycotoxins, OA being the most toxic during early life. 4. Weights of kidney and adrenals were increased by AF and OA. Liver weight was increased by AF (17.8%), while OA increased gizzard weight (14.6%) and reduced bone ash content (8.1%). T-2 toxin showed no effect on these variables. 5. Serum cholesterol content was decreased and activity of serum gamma glutamyl transferase (GGT) was increased by AF and OA while serum protein content was decreased by AF. These effects were more pronounced at 21 d than at 35 d of age. Inconsistent responses were seen in the other variables: blood urea nitrogen (BUN) content, activities of serum alanine amino transferase and aspertate amino transferase. Blood haemoglobin content was depressed by AF and T-2, whereas blood coagulation time was prolonged by OA. 6. Significant interactions were observed between any 2 toxins for their additive effects on body weight, food intake, bone ash content and serum GGT activity at 21 d. Conversely, antagonistic interactions were observed among any 2 of the toxins for their effects on variables such as serum protein and serum cholesterol content. Simultaneous feeding of all 3 mycotoxins did not show increased toxicity above that seen with any 2. 7. Esterified-glucomannan increased body weight (2.26%) and food intake (1.6%), decreased weights of liver (32.5%) and adrenals (18.9%) and activity of serum GGT (8.7%), and increased serum protein (14.7%), cholesterol (21.9%), BUN (20.8%) and blood haemoglobin (3.1%) content, indicating its possible beneficial effect on mycotoxicosis in broiler chickens.  相似文献

Canine adenovirus type 2 (CAV2) has been proposed for recombinant vaccines to control rabies in wild animals. To evaluate the suitability of CAV2 as a safe vector for the genetically engineered vaccines, seven wild-caught raccoons (three males and four females) were administered CAV2 per os. Two of the animals were euthanatized on each of post-infection days 3, 6, and 14, and one was euthanatized on day 21. Two other control raccoons (a male and a female) were also euthanatized on day 21. Microscopic pulmonary lesions of multifocal necrotizing bronchiolitis with basophilic intranuclear inclusions were seen in 3/4 raccoons euthanatized on post-infection days 3 and 6. Ultrastructural examination of lungs with pulmonary lesions revealed hexagonal viral particles characteristic of adenoviruses. CAV2 is potentially pathogenic for raccoons, and this susceptibility should be of concern to developers of recombinant vaccines who intend to use CAV2 as a vaccine vector.  相似文献
