The effect of daily injections of human chorionic gonadotropin (HCG) on luteal maintenance in hysterectomized prepuberal gilts induced to ovulate and in hysterectomized mature gilts was studied. Twenty-four pre-puberal gilts, 120 to 130 d of age, were induced to ovulate with 1,000 IU pregnant mare serum gonadotropin followed 72 h later with 500 IU HCG. Nine of the 24 prepuberal gilts (bred controls) were artificially inseminated on d 0 (d 0 = d after HCG). Mature gilts that had displayed one or more estrous cycles of 17 to 22 d were used (d 0 = onset of estrus). All gilts, except the bred controls, were totally hysterectomized on d 6 to 9 and their corpora lutea (CL) marked with charcoal. From d 10 through 29, eight prepuberal and 10 mature hysterectomized gilts received daily injections of 500 IU HCG in saline while seven prepuberal and eight mature hysterectomized gilts received daily injections of saline vehicle. Jugular blood samples were quantitated by radioimmunoassay for estrogen and 13,14-dihydro-15-keto prostaglandin F2 alpha (PGFM), a metabolite of prostaglandin F2 alpha. One bred control gilt was pregnant on d 30, indicating that the prepuberal gilts used in the experiment were prepuberal. All mature gilts and six of seven prepuberal gilts that received saline had maintained CL to d 30. Eight of 10 mature gilts that received HCG had maintained CL to d 30, while only two of eight (P less than .05) prepuberal gilts that received HCG maintained CL to d 30. In experiment 1, nine prepuberal crossbred gilts 145 +/- 2 days of age and 90.3 +/- 1.6 kg body weight (BW) were hypophysial stalk-transected (HST) or sham-HST. Starting at 0800 on Day 1 (35 +/- 2 days after surgery), three sham-HST and two HST gilts received 3.5% sodium citrate vehicle (V) while two HST gilts and two sham-HST gilts received pulses of 2.5 micrograms GnRH every 45 min for 9 days via a jugular vein cannula. At 0800 on day 7, all gilts received 1,000 IU of pregnant mare serum gonadotropin (PMSG) im. Blood was sampled every 15 min from 0800 to 0845 on Days 1 through 6. On Day 10, ovarian morphology and ovarian and follicular fluid weights were recorded. In experiment 2, eight prepuberal crossbred gilts, 146 +/- 6 days of age and 79.5 +/- 1.5 kg BW, were HST or sham-HST. Starting at 0800 on Day 1 (7 +/- 4 days after surgery), two sham-HST and three HST gilts received V, while three HST gilts received pulses of 2.5 micrograms GnRH every 45 min for 8 days. At 1200 on Day 5, all gilts, including three unoperated controls (UC), received 1,000 IU of PMSG im. Blood was sampled from all but UC gilts every 15 min from 0800 to 0845 on Days 1 through 5. Ovarian data were obtained on Day 9. Four pregnant, primiparous, crossbred gilts and six gilts from the same population that had been ovariectomized (OVX) for approximately 3 wk were placed in individual pens in an enclosed building. Blood samples were collected every 30 min for 12 h from all gilts via an indwelling jugular vein cannula when the pregnant gilts were at d 30, 50, 70, 90, and 110 of gestation. Serum was quantified for LH and prolactin (PRL) by RIA. The OVX gilts served as controls to ensure that any variations in serum LH and PRL concentrations observed in the pregnant animals were not due to environmental factors unrelated to pregnancy. Within the pregnant gilts, mean serum LH concentrations, mean basal serum LH concentration, and mean serum LH peak height were similar on all days; however, number of LH peaks on d 30, 50, and 70 were greater (P < .05) than on d 90 and 110, and number of LH peaks on d 50 was greater (P < .05) than that on d 70. Within the pregnant gilts, mean serum PRL concentration, mean basal serum PRL concentration, and mean PRL peak height were greater (P < .001) on d 110 than on all other days; however, number of PRL peaks were similar among days. Parameters of LH and PRL secretion in the OVX and pregnant gilts varied independently. Results of this study indicated that 1) LH secretion does not vary appreciably throughout pregnancy and 2) PRL secretion does not vary significantly during the first 90 d of pregnancy, after which it increases markedly on or before 110 d.  相似文献

Seventeen Landrace X Large White primiparous sows that farrowed in August 1982 were fed ad libitum (AL, n = 8) or their intakes were restricted (R, n = 9) during lactation. Litter sizes were equalized after farrowing and pigs were not allowed creep feed. Pigs were weaned 23.8 +/- .4 d postpartum. On d 6, 12 and 20 postpartum, all sows were fasted for 16 h and blood samples were collected prior to feeding for analysis of plasma glucose (GLU), urea nitrogen (UN), free fatty acids (FFA), prolactin (PRL) and serum insulin (INS). On d -2, 2 and 4 from weaning, sows were fasted for 16 h and then blood samples were collected hourly from 0 to 6 postprandial for analysis of GLU, UN, FFA, PRL and INS. Serum for analysis of luteinizing hormone (LH), progesterone and estradiol was collected every 6 h from 1 d before until 12 d after weaning. Samples for LH were also collected at 15-min intervals for 3 h at -18, -6, 6, 18, 78, 102, 126, 150, 240 and 480 h from weaning. After weaning all sows were fed 1.8 kg X d-1, and were checked for estrus twice daily. Daily intakes of metabolizable energy (ME) during lactation were greater in AL (12,194 +/- 465 kcal) than in R sows (8,144 +/- 90 kcal). Compared with AL sows, R sows lost more weight and backfat during lactation and had higher postprandial UN levels 2 d before and 4 d after weaning. Reproductive performance and reproductive hormones were not affected by restriction of energy, but frequency of episodic release of LH prior to weaning was greater in sows that exhibited estrus after weaning (n = 12) than in anestrous sows (n = 5). After weaning, LH and estradiol concentrations were similar between estrous and anestrous sows until onset of the preovulatory increase in estradiol in the sows that exhibited estrus. Energy intake, body condition and productivity were similar between anestrous sows and sows that exhibited estrus. On d 12 and 20 of lactation, preprandial levels of GLU were greater and FFA were lower in anestrous than estrous sows. We conclude that restriction of feed intake during lactation affected body condition and metabolism of primiparous sows, but reproductive performance and productivity were not affected. Aberrations in partitioning of energy during lactation may predispose primiparous sows to postweaning anestrus, but the mechanisms by which this occurs have yet to be defined.  相似文献

Cortisol and luteinizing hormone (LH) were measured in serum after the administration of adrenocorticotropic hormone (ACTH) to suckled (S) and nonsuckled (NS) beef cows. Blood was sampled on 2 consecutive days every 2 weeks for four bleeding periods starting 14 days after calving. Cows were injected with 200 IU ACTH or saline in a 2-day switchback design. Serum was collected before ACTH or saline injection and at 30-min intervals thereafter for 8 hours. Average cortisol concentrations in serum were similar in S and NS cows (6.4 +/- .6 and 6.1 +/- .8 ng/ml, respectively) after saline. Average cortisol concentrations in serum collected during an 8-hr period after ACTH on days 14, 28, 42 and 56 postpartum were 24.7 +/- 2.4, 31.8 +/- 3.5, 36.4 +/- 4.2 and 40.7 +/- .5 ng/ml, respectively, for S cows, and 31.1 +/- 2.9, 44.7 +/- 5.2, 45.0 +/- 5.7 and 46.0 +/- 5.4 ng/ml, respectively, for NS cows. Cortisol response to ACTH, measured as area under the response curve, was greater (P less than .05) in NS than in S cows. Amount of cortisol released by 200 IU ACTH was maximal by days 28 to 29 postpartum in NS cows, but the response increased gradually between days 14 to 15 and days 56 to 57 in S cows. overall, LH in serum averaged .55 +/- .08 ng/ml for S cows and .92 +/- .06 ng/ml for NS cows after saline, and .49 +/- .07 ng/ml for S cows and .94 +/- .06 ng/ml for NS cows after ACth. Although mean and peak serum LH concentrations did not differ between cows given ACTH and those given saline, the number of LH peaks and the number of cows having LH after saline. Mean serum LH concentrations were lower (P less than. 05) in S than in NS cows at 28 days postpartum. The number of LH peaks was lower (P less than .05) and the magnitude of the largest LH peak tended to be lower (P less than .06) in S cows at all sampling periods.  相似文献
