Combinatorial screening of electrochemical catalysts by current-voltage methods can be unwieldy for large sample sizes. By converting the ions generated in an electrochemical half-cell reaction to a fluorescence signal, the most active compositions in a large electrode array have been identified. A fluorescent acid-base indicator was used to image high concentrations of hydrogen ions, which were generated in the electrooxidation of methanol. A 645-member electrode array containing five elements (platinum, ruthenium, osmium, iridium, and rhodium), 80 binary, 280 ternary, and 280 quaternary combinations was screened to identify the most active regions of phase space. Subsequent "zoom" screens pinpointed several very active compositions, some in ternary and quaternary regions that were bounded by rather inactive binaries. The best catalyst, platinum(44)/ruthenium(41)/osmium(10)/iridium(5) (numbers in parentheses are atomic percent), was significantly more active than platinum(50)/ruthenium(50) in a direct methanol fuel cell operating at 60 degreesC, even though the latter catalyst had about twice the surface area of the former.  相似文献

A single-electron transistor scanning electrometer (SETSE)-a scanned probe microscope capable of mapping static electric fields and charges with 100-nanometer spatial resolution and a charge sensitivity of a small fraction of an electron-has been developed. The active sensing element of the SETSE, a single-electron transistor fabricated at the end of a sharp glass tip, is scanned in close proximity across the sample surface. Images of the surface electric fields of a GaAs/AlxGa1-xAs heterostructure sample show individual photo-ionized charge sites and fluctuations in the dopant and surface-charge distribution on a length scale of 100 nanometers. The SETSE has been used to image and measure depleted regions, local capacitance, band bending, and contact potentials at submicrometer length scales on the surface of this semiconductor sample.  相似文献

把引起豌豆枯萎病的丁香假单胞菌丁香致病变种 Pseudomonas syringae pv.syringae 菌株3319和丁香假单胞菌豌豆致病变种 P.syringae pv.pisi 的模式菌株2452的染色体 DNA 分别用限制性核酸内切酶 HindⅢ完全消化,再用 T_4DNA 连接酶把消化好的 DNA 片段连接到用HindⅢ内切酶消化的质粒载体 pUC19上,然后把重组的质粒载体转化到大肠杆菌(E.coli RR1)中去克隆.把菌株3319和2452的 DNA 用 p~(32)标记做成探针,分别与克隆菌落杂交,筛选出只对菌株3319的 DNA 有特异性的1个重组 DNA 的克隆和对菌株2452的 DNA 有特异性的2个重组 DNA 克隆.再把这3个重组质粒 DNA 分别用 P~(32)标记做成探针,与菌株3319和2452的 DNA 杂交,也显示只对各自 DNA 来源的菌株具有特异性.这3个重组质粒中,有2个携带有菌株2452的 DNA 片段,1个为0.82 kb。另1个略小于0.58 kb,还有1个质粒携带有菌株3319的 DNA 片段,为0.85 kb.  相似文献

Background

Recent studies of anadromous salmon (Oncorhynchus spp.) on the Pacific Coast of North America indicate an important and previously unrecognized role of salmonid nutrients to terrestrial biota. However, the extent of this uptake by primary producers and consumers and the influences on community structure remain poorly described. We examine here the contribution of salmon nutrients to multiple taxa of riparian vegetation (Blechnum spicant, Menziesii ferruginea, Oplopanax horridus, Rubus spectabilis, Vaccinium alaskaense, V. parvifolium, Tsuga heterophylla) and measure foliar δ15N, total %N and plant community structure at two geographically separated watersheds in coastal British Columbia. To reduce potentially confounding effects of precipitation, substrate and other abiotic variables, we made comparisons across a sharp ecological boundary of salmon density that resulted from a waterfall barrier to salmon migration.

Results

δ15N and %N in foliage, and %cover of soil nitrogen indicators differed across the waterfall barrier to salmon at each watershed. δ15N values were enriched by 1.4‰ to 9.0‰ below the falls depending on species and watershed, providing a relative contribution of marine-derived nitrogen (MDN) to vegetation of 10% to 60%. %N in foliar tissues was slightly higher below the falls, with the majority of variance occurring between vegetation species. Community structure also differed with higher incidence of nitrogen-rich soil indicator species below the waterfalls.

Conclusions

Measures of δ15N, %N and vegetation cover indicate a consistent difference in the riparian community across a sharp ecological boundary of salmon density. The additional N source that salmon provide to nitrogen-limited habitats appears to have significant impacts on the N budget of riparian vegetation, which may increase primary productivity, and result in community shifts between sites with and without salmon access. This, in turn, may have cascading ecosystem effects in forests adjacent to salmon streams.

Objectives   To describe the morbidity and mortality patterns and identify factors associated with morbidity in pre-weaning llamas and alpacas.
Design   Cross-sectional observational study of 287 crias born on four farms in Ohio, USA
Procedure   Historical data representing all crias born over a 6-year period were obtained from two llama farms and two alpaca farms in Ohio. Multivariable generalised linear mixed effects regression models were used to identify factors associated with morbidity outcomes.
Results   In total, 105 (37%) of the llamas and alpacas had some reported morbidity during the pre-weaning period, and mortality rate was 2.1%. In addition, 51 (18%) of llamas and alpacas experienced morbidity because of infectious disease, and 47 (16%) experienced morbidity during the neonatal period. The three most commonly reported morbidity events were undifferentiated diarrhoea (23%), umbilical hernia (16%) and unspecified infectious disease (15%). Difficult birth was an important risk factor for pre-weaning morbidity in this population.
Conclusions   Camelid veterinarians and their clients can expect that pre-weaning health events are common among crias, although mortality is low. Crias experiencing difficult births may require additional monitoring for health events during the pre-weaning period.  相似文献

The aim of this study was to asses the variation in the morphology of the seminal epithelium in relation to natural photoperiod in male cats. Tom cats (n = 240) were castrated every other week throughout the year. Each testis was fixed in Bouin's solution and cut into sections. The percentage of tubules with round spermatids (RS), elongated spermatids (ES), tailed spermatids (TS), mature spermatids (MS) and the number of Sertoli cells (SC) and Leydig cells (LC) were recorded in each sample. Testicles from males during short days (SHD) had a higher percentage of tubules with RS and ES compared to testicles from males during long days (LHD, 31.3 ± 0.6 vs 2.1 ± 0.6%, p < 0.001; 30.9 ± 0.7 vs 11.0 ± 0.7%, p < 0.001). Conversely, testicles from males during SHD had a lower percentage of tubules with TS and MS compared to testicles from males during LHD (24.5 ± 0.8 vs 29.7 ± 0.8%, p < 0.01; 13.1 ± 1.2 vs 57.0 ± 1.2%, p < 0.01). Furthermore, testicles from males during SHD had a higher number of SC and lower number of LC compared to testicles from males during LHD (11.4 ± 0.1 vs 8.0 ± 0.1%, p < 0.01; 19.2 ± 1.0 vs 38.0 ± 1.0%, p < 0.01). In conclusion, there are seasonal changes in testis cell morphology in the tom which may be related to seasonal sperm production.  相似文献

在对沙棘木蠹蛾危害情况进行调查时,从死亡的沙棘树干内发现了罹病致死的沙棘木蠹蛾,通过对虫尸体内病原真菌的分离与鉴定,确定为球孢白僵菌(Beauveria bassiana)。经室内致病力测定,接种浓度4亿/mL、8亿/mL、12亿/mL,54 d的感病率分别是72.22%、83.33%、100%,证明球孢白僵菌是沙棘木蠹蛾的病原菌。同时表明浓度越高感病率越高。  相似文献
