Evaluation of Trypsin Treatment on the Inactivation of Bovine Herpesvirus Type 1 on In Vitro Produced Pre-implantation Embryos

The aim of this study was to evaluate the efficiency of trypsin treatment on the inactivation of bovine herpesvirus type 1 (BoHV-1) on in vitro produced by fertilization and artificially infected bovine embryos. Bovine embryos on day 7 were exposed with 10 μl of BoHV-1, Los Angeles strain 10^7.5 TCID. These embryos and control embryos were divided in two groups: submitted to the sequential washes or to the trypsin treatment according to the International Embryo Transfer Society (IETS) guidelines. The embryos and the last washing drop of each group were used as inoculum to infect Madin Darby bovine kidney (MDBK) cells and submitted to nested PCR reaction using the primer that encodes the gene conserved region of virus glycoprotein gB. The data have shown that the control embryos and their last washing drop were negative. The exposed embryos that were treated with trypsin have shown positive results on the n-PCR and MDBK culture, and their last washing drop were negative. Our data have demonstrated that the trypsin treatment was not able to eliminate the BHV-1 of the embryos, suggesting an interaction between virus and embryo.     

Characterisation of haemolytic RTX toxins produced by Australian isolates of Actinobacillus pleuropneumoniae

The haemolytic RTX toxins of 27 isolates of Actinobacillus pleuropneumoniae, representing all serovars that have been isolated in Australia, were characterised. The quantity of protein secreted by these isolates into the media was not significantly different between serovars, but haemolytic activity was detected only in the unconcentrated supernatants from cultures of serovar 1 and 5 isolates. Haemolytic activity in supernatants of serovar 2, 3 and 7 isolates was detected only after the supernatants were concentrated. On Southern hybridisation blots, genomic DNA of serovar 1 and 5 isolates contained regions that were similar to the cloned structural genes for Apxl (apxIA) and for ApxII (apxIIA). In contrast, genomic DNA of serovar 2,3 and 7 isolates only contained regions similar to, if not identical with, the cloned apxIIA gene. The haemolytic activity of the culture supernatant depends on the type or composition of media and adaptability of the bacteria to in-vitro cultivation. Low passage cultures of A pleuropneumoniae, which were characterised by waxy colonies, produced significantly weaker haemolytic activity than A pleuropneumoniae after several passages in vitro.     

GONG Observations of Solar Surface Flows

Doppler velocity observations obtained by the Global Oscillation Network Group (GONG) instruments directly measure the nearly steady flows in the solar photosphere. The sun's differential rotation is accurately determined from single observations. The rotation profile with respect to latitude agrees well with previous measures, but it also shows a slight north-south asymmetry. Rotation profiles averaged over 27-day rotations of the sun reveal the torsional oscillation signal-weak, jetlike features, with amplitudes of 5 meters per second, that are associated with the sunspot latitude activity belts. A meridional circulation with a poleward flow of about 20 meters per second is also evident. Several characteristics of the surface flows suggest the presence of large convection cells.     

Serum antibody response to Mycoplasma hyopneumoniae measured by enzyme-linked immunosorbent assay after experimental and natural infection of pigs

Studies were conducted under experimental and field conditions to determine the effect of infection with M. hyopneumoniae on the immune response in serum as measured by ELISA. Following intratracheal challenge or contact exposure, serologically negative pigs derived from mycoplasma-free piggeries developed an immune response within 10 days. This response continued to rise for a further 50 days. In a field study in a commercial piggery, no animals (0/44) were observed to have M. hyopneumoniae antibodies at day 86 of life. However between day 86 and day 144, 97.7% (42/43) animals sero-converted. These results are discussed in terms of infection spread, particularly in the grower/finisher shed.
