Four foliar and two stem-base pathogens were inoculated onto wheat plants grown in different substrates in pot experiments. Soils from four different UK locations were each treated in three ways: (i) straw incorporated in the field at 10 t ha−1 several months previously; (ii) silicon fertilization at 100 mg L−1 during the experiment; and (iii) no amendments. A sand and vermiculite mix was used with and without silicon amendment. The silicon treatment increased plant silica concentrations in all experiments, but incorporating straw was not associated with raised plant silica concentrations. Blumeria graminis and Puccinia recondita were inoculated by shaking infected plants over the test plants, followed by suitable humid periods. The silicon treatment reduced powdery mildew ( B. graminis ) substantially in sand and vermiculite and in two of the soils, but there were no effects on the slight infection by brown rust ( P. recondita ). Phaeosphaeria nodorum and Mycosphaerella graminicola were inoculated as conidial suspensions. Leaf spot caused by P. nodorum was reduced in silicon-amended sand and vermiculite; soil was not tested. Symptoms of septoria leaf blotch caused by M. graminicola were reduced by silicon amendment in a severely infected sand and vermiculite experiment but not in soil or a slightly infected sand and vermiculite experiment. Oculimacula yallundae (eyespot) and Fusarium culmorum (brown foot rot) were inoculated as agar plugs on the stem base. Severity of O. yallundae was reduced by silicon amendment of two of the soils but not sand and vermiculite; brown foot rot symptoms caused by F. culmorum were unaffected by silicon amendment. The straw treatment reduced severity of powdery mildew but did not detectably affect the other pathogens. Both straw and silicon treatments appeared to increase plant resistance to all diseases only under high disease pressure.  相似文献

Lithuania has requested that its whole territory should be recognized by the EU as a protected zone for Erwinia amylovora . Fireblight monitoring was performed in 1998/2002 with the aim of detecting and identifying the bacterium, and of determining its distribution in the country. The study consisted of periodic surveys (at least twice a year) of nurseries, orchards, collective farms and host plants, growing individually or in small groups, as well as the surrounding zone within a radius of 250 m. Tests, under conditions of quality control, were applied to host plants with and without symptoms, using detection methods such as ELISA and immunofluorescence (with polyclonal antibodies), semi-selective plating and pathogenicity.  相似文献

Stratospheric ozone (O3) depletion has led to increased terrestrial ultraviolet‐B (UV‐B) radiation (290–320 nm). Leaves exposed to this radiation produce UV‐absorbing compounds in the epidermal cells, which protect plants from UV‐B damage. To determine the role of UV‐absorbing compounds in the UV‐B sensitivity of weeds (common chickweed (Stellaria media), downy brome (Bromus tectorum), green smartweed (Polygonum scabrum), redroot pigweed (Amaranthus retroflexus), spotted cat’s‐ear (Hypochoeris radicata), and stork’s‐bill (Erodium cicutarium)) seedlings were exposed to 0, 4 (field ambient), 7 (18% O3 depletion) and 11 (37% O3 depletion) kJ m?2 d?1 of biologically effective UV‐B radiation in a greenhouse. Ultraviolet‐absorbing compounds were extracted from the second true‐leaf (0.5 cm2 samples) with methanol : distilled water : HCl (79 : 20 : 1) in an 85°C water bath for 15 min, and the absorbance of the extracts measured at 300 nm. The shoot dry biomass was recorded to determine the susceptibility to UV‐B radiation. Common chickweed was the most sensitive and green smartweed the least sensitive weed to UV‐B radiation. The latter accumulated more UV‐absorbing compounds and this accumulation occurred earlier compared with common chickweed. As UV‐BBE radiation levels increased from 0 to 11 kJ m?2 d?1, the green smartweed shoot biomass did not decline. However, the biomass of all five susceptible species declined despite an increase in the UV‐absorbing compounds in response to increased UV‐B radiation. Therefore, formation of a ‘UV‐screen’ in these species is not sufficient to fully prevent UV‐B damage. When the concentration of UV‐absorbing compounds in the six species was plotted against their susceptibility to UV‐B radiation, no relationship was observed. Thus, while the accumulation of UV‐absorbing compounds may be a major factor in the protection of certain species against UV‐B radiation and may offer some degree of defence in other species, it does not explain UV‐B susceptibility differences in weedy species in general.  相似文献

Fireblight (Erwinia amylovora) appeared in Hungary in 1996. Most damage occurred on apple, pear, quince and medlar, and also on the ornamentals Pyracantha, Sorbus, Cotoneaster and Crataegus. In 1996–2006, an official programme for elimination of infected parts of plants started in Békés county. This mainly concerned trees in towns and villages, since there are few pome‐fruit orchards in the county. Work teams under official direction pruned back or cut down trees. In total, some 13 000 trees were pruned back and nearly 11 000 were cut down. In addition, 21 villages were subjected to special phytosanitary measures. Infection decreased considerably between 1996 and 2002, but over 90% of the inhabited areas in the county remained subject to special measures, because of the very dispersed occurrence of fireblight.  相似文献

Plant resistance to insect herbivory involves gene expression in response to wounding and the detection of insect elicitors in oral secretions (Kessler and Baldwin, 2002, Ann. Rev. Plant/ Biol. 53: 299–328). However, crawling insect larvae stimulate the synthesis of 4-aminobutyrate within minutes and imprints of larval footsteps can be visualized within seconds through superoxide production or transient increases in chlorophyll fluorescence (Bown et al., 2002, Plant Physiol. 129: 1430–1434). Here cryo-scanning electron microscopy was used to demonstrate that larval feet, which are equipped with a perimeter row of hook-like crochets, damage leaf tissue and result in larval footprints. Staining for cell death shows that areas of wounding correspond to footsteps detected through increased chlorophyll fluorescence. Superoxide production in response to footsteps was inhibited by diphenyleneiodonium, an inhibitor of the plasma membrane NADPH oxidase enzyme. Inhibition of superoxide production, however, did not eliminate the detection of cell death. The results demonstrate that larval footsteps damage leaf tissue, and initiate rapid local responses which are not dependent on herbivory or oral secretions. It is proposed that superoxide production at the wound site prevents opportunistic pathogen infection.  相似文献

A study was carried out to demonstrate that Rice yellow mottle virus (RYMV), a virus known to be transmitted by beetles, can spread between rice plants by direct leaf contact caused by wind. Almost all healthy plants surrounding an infected plant became infected when exposed to a fan blowing for 15 min at a distance of 50 cm. Spread of RYMV by plant contact, mediated by wind, was also demonstrated in field experiments, the extent of spread depending on plant density. Infection was almost 10 times higher in plots with a density of 33 plants m−2 than in plots with 16 plants m−2. Less spread was observed in plots protected by 1·5 m high windscreens. It is suggested that wind-mediated spread of RYMV may result from abrasive contact between leaves of plants.  相似文献

A new virus named Nootka lupine vein-clearing virus (NLVCV) was isolated from Lupinus nootkatensis plants that were confined to a relatively small area in the Talkeetna mountains of south-central Alaska. Annual surveys (2000–03) consistently found leaf symptoms of pronounced vein clearing and mosaic on 3- to 4-week-old plants in late June. Spherical particles ≈30 nm in diameter were isolated from these leaves. Virions contained a single-stranded RNA of ≈4·0–4·2 kb and one species of capsid protein estimated to be ≈40 kDa. The double-stranded RNA profile from naturally infected leaves consisted of three major bands ≈4·2, 1·9 and 1·5 kbp. Protein extractions from either sap or virions of diseased plants reacted to polyclonal antiserum made against the virions in Western blot assays. A predicted PCR product ≈500 bp was synthesized from virion RNA using primers specific to the carmovirus RNA-dependent RNA polymerase (RDRP) gene. The nucleotide sequence of the amplified DNA did not match any known virus, but contained short regions of identity to several carmoviruses. Only species belonging to the Fabaceae were susceptible to NLVCV by mechanical inoculation. Based on dsRNA profile, size of virion RNA genome and capsid protein, and similarity of the RDRP gene to that of other carmoviruses, it is suggested that NLVCV is a member of the family Tombusviridae , and tentatively of the genus Carmovirus . As the host range, RDRP gene and dsRNA profile of NLVCV are different from those of known viruses, this is a newly described plant virus.  相似文献

Sperm concentration of wild yak bull is 2. 13 billion per milliliter. Survival time at 0-4 ℃ is 57hrs. After thawing survival time at 37 ℃ is 12 hrs. Resistance coefficient is 144,000. Abnormal sperm rate and acrosomal integrated rate of post-thawing is 9.17% and 87.53% respectively. Moving viscosity is 1. 169 cp. Total nitrogen is 1 437.7 mg/100 ml. Head of sperm is significantly shorter and end piece is significantly longer than that of domestic yak and yellow cattle. Activity of hyaluronidase is highly stronger than the domestic yak,and the activity of LDH is higher than the domestic yak by 48 %. Hence,the fertility of the wild yak bull is powerful and the artificial inseminated rate with domestic yak and yellow cattle is 88.9% and 71.58% respectively.  相似文献
