The structural changes in the cell wall and delignification pattern caused by Trametes versicolor and Trametes hirsuta in the sap wood of Leucaena leucocephala were examined by light and confocal laser scanning microscopy. The in vitro decay test was conducted for 12 weeks. Both species of Trametes used in this study caused anatomical characteristics specific to simultaneous white rot. Formation of boreholes, erosion troughs, erosion channels with U-shaped notches in tangential sections and thinning of cell walls were evident in the wood inoculated with each of the fungal species. Cell separation due to removal of middle lamellae occurred at the early stages of infection (30 d) with T. versicolor. In contrast, middle lamellae remained intact in wood inoculated with T. hirsuta and showed cell separation due to degradation of the outer layer of the secondary wall and degradation of the middle lamellae observed only in severely decayed wood at late stages. Confocal microscopy revealed the delignification pattern particularly from cell corners and vessel walls at an advanced stage of decay, indicating strong ligninolytic activity of both species in the sapwood of L. leucocephala.  相似文献

Human filariasis caused by Brugia malayi is still a public health problem in many countries of Asia including India, Indonesia, Malaysia and Thailand. The World Health Organization (WHO) has targeted to eliminate filariasis by the year 2020 by Mass annual single dose Diethylcarbamazine Administration (MDA). Results of the MDA programme after the first phase was less satisfactory than expected. Malayan filariasis caused by B. malayi is endemic in the south of Thailand where domestic cat serves as the major reservoir host. There is no report about the occurrence of B. malayi in dogs. The present work was carried out to find out the incidence of microfilariasis in dogs and also to detect the presence of human filarial infection in dogs, if any. One hundred dogs above 6 months of age presented to the veterinary college Hospital, Mannuthy, Kerala, with clinical signs suggestive of microfilariasis - fever, anorexia, conjunctivitis, limb and scrotal oedema - were screened for microfilariae by wet film examination. Positive cases were subjected to Giemsa staining, histochemical staining and molecular techniques. Results of the study showed that 80% of dogs had microfilariasis; out of which 20% had sheathed microfilaria. Giemsa and histochemical staining character, PCR and sequencing confirmed it as B. malayi. High prevalence of B. malayi in dogs in this study emphasized the possible role of dogs in transmission of human filariasis.  相似文献

Agrobacterium-mediated genetic transformation was performed using embryonic axes explants of pigeon pea. Both legume pod borer resistant gene (cry1Ac) and plant selectable marker neomycine phosphor transferase (nptII) genes under the constitutive expression of the cauliflower mosaic virus 35S promoter (CaMV35S) assembled in pPZP211 binary vector were used for the experiments. An optimum average of 44.61% successfully hardened dot blot Southern hybridization positive plants were obtained on co-cultivation media supplemented with 200 μM acetosyringone without L-cysteine. The increased transformation efficiency from a baseline of 11.53% without acetosyringone to 44.61% with acetosyringone was further declined with the addition of different concentrations of L-cysteine to co-cultivation media. Transgenic shoots were selected on 50 and 75 mg L−1 kanamycin. Rooting efficiency was 100% on half-strength Murashige and Skoog medium with 20 g L−1 sucrose and 0.5 mg L−1 indole butyric acid in the absence of kanamycin. Furthermore, 100% seed setting was found among all the transgenic events. The plants obtained were subjected to multi- and nochoice tests to determine the behavioral responses and mortality through Helicoverpa armigera bioassays on the leaf and relate their relationship with the expression of cry1Ac protein which was found to be less in leaf as compared to the floral buds, anther, pod, and seed.  相似文献

The anatomical changes occurring during transition from juvenile to mature wood were studied in the branch and main trunk of Leucaena leucocephala. To confirm the wood maturity in the 5-yr-old tree, the anatomical features were compared with the adult wood collected from the main trunk of a 15-yr-old tree. In both the branch and wood of the trunk, most of the anatomical features—such as increase in dimensions of fibers and vessel elements, and decrease in frequency of vessel elements and rays—were gradual during radial growth from pith to periphery. A noticeable anatomical change is observed in the transition of triseriate rays to multiseriate rays in the region of wood which is about 5.5 cm away from the pith. The ray dimensions and anatomical features of other elements from the periphery of the branch and 5-yr-old trunk xylem match with that of 15-yr-old main trunk wood. This indicates that in both the branches and main trunk of Leucaena, transition from juvenile to adult or mature wood occurs after the trees have attained 14 cm radial growth.  相似文献

In vitro laboratory decay tests on Ailanthus excelsa Roxb. wood revealed that I. hispidus exhibits a combination of both white-rot and soft-rot patterns of wood decay. Early stages of wood decay showed dissolution of the middle lamella as well as defibration and localized delignification of fiber walls; vessels, axial, and ray parenchyma remained unaltered. Delignification commenced from the middle lamellae at the cell corners without any marked effect on the primary and secondary wall layers. In later stages of growth, the species produces typical soft-rot decay pattern by forming erosion channels through the S2 layers of fiber walls, transverse bore holes in the cell walls, and erosion channels alongside/following the orientation of cellulose microfibrils. The rays showed signs of cell wall alterations only after the extensive damage to the fiber walls. After 120 days of incubation, the vessels also showed localized delignification, the erosion of pits, and separation from associated xylem elements. The extensive weight losses under natural and in vitro decayed wood as well as the very soft nature of severely degraded wood indicate that I. hispidus alters wood strength and stiffness.  相似文献

Cotton (Gossypium sp.) is a major crop grown under rainfed conditions in Vertisols and associated soils in semi-arid tropical (SAT) regions of Peninsular India. In recent years, cotton productivity has declined due to various biophysical factors including pest and diseases, seasonal water stress soil degradation and poor crop management practices. In this study, we compare two methods for evaluating the suitability of Vertisols for cotton in contrasting two agro-ecological regions viz., sub-humid moist (SHM) region and semi-arid dry(SAD) were characterized. Twelve cotton growing Vertisols (seven from SHM and five from SAD) were evaluated for their suitability for cotton cultivation using soil quality index (SQI) and modified Sys-FAO method. SQIs were calculated using the weighted additive index from transformed scores of selected indicators by principal component analysis. For Sys-FAO method both biophysical and soil characteristics were considered for suitability evaluation. We found that the soils of SHM region were moderately suitable for cotton cultivation with soil moisture as the major limiting factor, whereas the soils of SAD region are marginally suitable due to high exchangeable sodium percentage and poor hydraulic conductivity. From this, it may be concluded that the weighted SQI has better agreement with the cotton yield.  相似文献

The influence of position of cuttings, auxin treatments and seasonal variation on adventitious rooting of Dendrocalamus asper, a multipurpose edible bamboo of horticultural value, was examined. Single node proximal (culm) and distal (culm-branch) cuttings were collected during the months of April–June and treated for 24 h with water or 2 mM auxin (indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) and naphthalene acetic acid (NAA)). The treated cuttings were planted horizontally in sand beds and maintained under misting conditions for 2 months. Culm cuttings exhibited better adventitious rooting than culm-branch cuttings and the month of May proved to be the best for rooting of both types of cuttings. Auxin treatments also influenced adventitious rooting and their effectiveness was in the order: NAA>IBA>IAA. The interaction among auxin, position of cuttings and month of collection was also found to be significant on root induction and differentiation. Single node culm cuttings, without auxin treatment, resulted in 98% plantlets in May and 67% plantlets in June, and emerged as a suitable material for large-scale vegetative propagation. Administration of IAA or NAA to abundant culm-branch cuttings of low commercial utility (collected in May) also produces ≥50% plantlets.  相似文献

1. Two experiments were conducted to examine the effect of feeding higher supplemental vitamin E (VE) concentrations on male reproductive variables, fertility and hatchability of quails. 2. In experiment 1, sixty 5-week-old male quails, reared in individual cages, were fed male breeder diets supplemented with 0, 75, 150, 225 or 300 IU alpha-tocopherol acetate/kg. At 10 weeks, observations on cloacal gland size, foam production, testes weight and blood samples for testosterone estimation were taken. 3. In experiment 2, 50 male and 50 female cage-reared quails (5 weeks old) were fed male breeder and layer rations, respectively, supplemented with 0, 75, 150, 225 or 300 IU/kg. At 13 weeks, 9 different mating pairs (5 pairs/group) were formed. Group 1 contained one control male and one control female in each of 5 pens. In groups 2 to 5, control males were paired with females on supplements of 75, 150, 225 or 300 IU/kg. In groups 6 to 9, control females were paired with males on supplements of 75, 150, 225 and 300 IU/kg. During subsequent adaptation and egg collection periods, each of 10 d, control layer ration was fed to all groups. All the eggs laid during the 10-d egg collection period were incubated artificially to estimate fertility and hatchability. The trial was repeated at 15 weeks. 4. Adult male quails receiving moderate supplemental VE (75 and 150 IU/kg) had a higher cloacal gland index, quantity of foam secretion, testicular weight and plasma testosterone than quails fed on either VE-deficient or more highly supplemented diets (225 and 300 IU alpha-tocopherol acetate/kg). 5. It was concluded that a supplement of 75 IU VE/kg in maize/soybean diet could provide the best reproductive performance of male breeder quails. Cloacal gland index, quantity of foam secretion, testicular weight and plasma testosterone can serve as indirect indicators of testicular activity and fertilising ability in quails. Supplemental VE did not affect the fertility and hatchability of male and female Japanese quails.  相似文献

One new glycoside derivative from syringic acid and one new phenol glycoside, curculigoside E (1) and orchioside D (2), were isolated and characterized from the rootstock of Curculigo orchioides collected in the Nawalparasi District (Nepal). Soil samples were collected from rhizosphere of litchi-growing areas of North-Western Himalayan Region (NWHR) of India, for finding qualitative and quantitative differences in arbuscular mycorrhizal (AM) fungi and Azotobacter chroococcum. These samples were taken from plants being grown in different cultivation types namely, weed control with weedicides or tillage; orchard floor either clear or with cover crops; intercropping with cereals and legumes. Qualitative and quantitative differences were noticed with different cultivation types and a marked reduction in the AM fungi was observed in orchards where chemicals were used for weed control and intensive farming system was used on the orchard floor. AM fungi were generally abundant in the soils with range pH 5.5–6.6. Among different AM fungi retrieved from the soils, Glomus spp. was most dominant. Fifteen AM fungal species were isolated, identified and characterized and along with their ability to colonize the roots. In the soil samples, a marked variation in viable bacterial count of A. chroococcum was also noticed due to varied physico-chemical characteristics of the orchard soils. The changes in AM fungal species composition can be attributed to changes in soil chemical properties resulting from cultural practices such as ploughing, application of chemical fertilizers and weedicides. An experiment was also conducted to study the comparative efficacy of four dominant and frequently occurring indigenous AM species namely, Glomus fasciculatum (Thaxter sensu Gerdemann), G. magnicaulis (Hall), G. mosseae (Nicol. & Gerd.), Gigaspora heterogamma (Nicol. & Gerd.) and two A. chroococcum strains viz., AZ1 and AZ2 singly and in dual combination to evaluate their effect in air-layers system. Dual inoculation of G. fasciculatum and AZ1 increased total root length of air-layered shoots by 81.39% over uninoculated control. These studies indicated that indigenously isolated AM fungal species and A. chroococcum strains can be used for air-layering for better adaptation under specific agro-climatic and ecological zone conditions.  相似文献
