Somatic hybrids of Sinapis alba+Brassica juncea (Sal Sal AABB) were synthesized by protoplast electrofusion. They were true genomic allopolyploids since they possessed 60 chromosomes, i.e. the sum of S. alba (2n= 24) and B. juncea (2n= 36) chromosomes. Chromosome pairing was predominantly bivalent with the occasional occurrence of multivalents in the pollen mother cells at diakinesis and metaphase I. Hybrids were completely pollen-sterile, but produced seeds on back-crossing with B. juncea and B. campestris. A total of 37 BC1 plants were raised from two somatic hybrids (JS-1 and JS-2) and 24 of these were analysed cytologically. The 22 plants originating from the pollinations of somatic hybrids with B. juncea showed a chromosome configuration of 18II+12I and had 42–86% pollen fertility. Two plants from the backcrosses of the somatic hybrid with B. campestris formed 10II +20I, and had 0–4% fertile pollen. Total DNA analysis by probing with pTA71 carrying a full-length 18S–25S rDNA fragment of the wheat nuclear genome revealed that the two somatic hybrids possessed all the characteristic bands of both the species, confirming their hybridity. Probing with the mitochondrial coxI and atp9 genes indicated mitochondrial genome recombination in the hybrids. Hybridization with chloroplast-specific psbD indicated that both the somatic hybrids possessed the cp genome of S. alba origin.  相似文献

The detection of DNA polymorphism in cultivated peanut (Arachis hypogaea L.) is reported here for the first time. The DNA amplification fingerprinting (DAF) and amplified fragment length polymorphism (AFLP) approaches were tested for their potential to detect genetic variation in peanut. The AFLP approach was more efficient as 43% of the primer combinations detected polymorphic DNA markers in contrast to 3% with the DAF approach. However, the number of polymorphic bands identified using primers selected in both approaches was comparable. In the DAF study, when 559 primers of varying types were screened, 17 (mostly 10-mer types) detected polymorphism producing an average of 3.7 polymorphic bands per primer with a total of 63 polymorphic markers. In the AFLP study, when 64 primer combinations (three selective nucleotides) corresponding to restriction enzymes Eco RI and Mse I were screened, 28 detected polymorphism. On an average, 6.7% of bands obtained from these 28 primer pairs were polymorphic resulting in a total of 111 AFLP markers. Our results demonstrate that both AFLP and DAF approaches can be employed to generate DNA markers in peanut and thus have potential in the marker-assisted genetic improvement and germplasm evaluation of this economically important crop. Summary Nineteen isolates of Melampsora medusae (Thüm.), collected from natural stands of Populus deltoides (Bartr.) along the lower Mississipi River Valley, were tested for the occurrence of physiologic races by inoculation of nine poplar clones. Eight distinct races were identified based upon differential responses on these clones. However, most of the isolates also differed in their aggressiveness (latent period and uredial number per leaf disk), and a significant isolate × cultivar interaction was observed for both traits. Within a specific geographic location, more than one race was present. Isolates from the northernmost sampling location (37°N latitude) appeared more aggressive than those from southern latitudes (34–36°N latitude). Thus wild pathosystems may be composed of variable pathogen populations differing in virulence and the apparent stability usually observed in such systems may be due to the genetic diversity of the host population in interaction with many epidemiological and ecological factors.The investigation reported in this paper (No. 86-8-96) is in connection with a project of the Kentucky Agricultural Experiment Station and is published with approval of the Director.  相似文献

The common carp (Cyprinus carpio L.) is one of the major aquaculture species, contributing nearly 35% to the inland fish production in Karnataka, India. Stocks collected from Hungary (2), Indonesia and Vietnam were assessed alongside two local stocks in a series of culture performance trials with the objective of setting up a base population for developing a breeding programme. The present study deals with the genetic divergence and polymorphism in these six stocks using random‐amplified polymorphic DNA (RAPD) markers. A total of 180 decamer random primers were screened for polymerase chain reaction (PCR) amplification (OPA 1‐20, OPB1‐20, OPC1‐20, OPD1‐20, OPE1‐20, OPF1‐20, OPG1‐20, OPP1‐20 and OPM1‐20). Eight primers were selected for analysis of common carp genotypes (OPA‐7, OPA‐20, OPB‐17, OPF‐10, OP F‐9, OPG‐4, OPG‐9 and OPP‐16). Out of 492 bands recorded, 57.1% were polymorphic. Stepwise regression analysis was carried out to find best combination markers affecting body weight (P<0.001). The results demonstrate major differences in the genetic structures between different stocks. Dendrogram data showed grouping of individuals according to stocks and corresponding data variables revealed the per cent homology within the stock and also found markers correlating to the body weight.  相似文献

A new cytoplasmic male-sterility system was developed in an oilseed Brassica, viz. B. juncea var. ‘Pusa Bold’ with the cytoplasmic background of a wild species, Diplotaxis siifolia, obtained through wide hybridization. The synthetic alloploid (D. siifolia×B. juncea: 2n = 56, D3D3AABB) was repeatedly backcrossed to B. juncea to achieve cytoplasmic substitution. The CMS plants resembled the cultivar in growth and morphology. The flowers had narrow sepals and petals and short, shrivelled anthers which failed to dehisce. The meiotic process appeared to be normal. The microspores degenerated at an early stage after tetrad formation. Female fertility in the CMS plants was as good as in the cultivar. Female transmission of sterility confirmed it to be cytoplasmically encoded.  相似文献

Cultivated peanut (Arachis hypogaea L.) consists of six botanical varieties. Identification of DNA markers associated with botanical varieties would be useful in plant genotyping, germplasm management, and evolutionary studies. We have developed 130 simple sequence repeat (SSR) markers in peanut, 38 of which were used in this study because of their ability in detecting genetic polymorphism among 24 peanut accessions. Eight SSR markers were found useful to classify botanical varieties. Among them, six SSR markers were specific to botanical varieties fastigiata and vulgaris, one to botanical varieties hypogaea and hirsuta, and one to botanical varieties peruviana, and aequatoriana. Also, three of them derived from peanut expressed sequence tags (ESTs) were associated with putative genes. As botanical varieties have different morphological traits and belong to different subspecies in A. hypogaea, these markers might be associated with genes involved in the expression of morphological traits. The results also suggested that SSRs (also called microsatellites) might play a role in shaping evolution of cultivated peanut. Multiplex PCR of botanical variety-specific markers could be applied to facilitate efficient genotyping of the peanut lines.  相似文献
