Neoparamoeba pemaquidensis believed to be the most prevalent parasite of Atlantic salmon industry in Australia. In the present study, the in vitro effects of crude extract of garlic and metronidazole were investigated using a primary culture toxicity assay. Garlic extract appeared to be completely effective at killing a cultured strain (NP251002) of Neoparamoeba pemaquidensis in vitro at a dilution of 1:100 with in 24 h. The number of viable Amoebae after using garlic extract in lower dilutions (1:200, 1:400, 1:800, 1:1000) for 24 h, also were significantly lower than in the control group. Garlic extract was also efficacious at killing wild type Amoebae that isolated from the diseased fish showing clinical signs of AGD. Metronidazole had no clear effect against Neoparamoeba pemaquidensis (NP251002) even in a concentration of 50 mg L(-1) for 24 h. However some morphological changes have occurred in metronidazole-treated Amoebae after 5 days of exposure.  相似文献

Atlantic surfclams (Spisula solidissima), among the largest extant non‐symbiotic clam species in the world, live in dense aggregations along the Middle Atlantic Bight (MAB) continental shelf. The food resources that support these populations are poorly understood. An individual‐based model that simulates the growth of post‐settlement surfclams was used to investigate the quantity of food needed to maintain existing surfclam populations along the MAB continental shelf. Food inputs to the model were based on measured near‐bottom water‐column chlorophyll concentrations. Simulations showed that these water‐column food sources supported only 65% of the observed body mass of a standard large surfclam (160‐mm shell length). Additional simulations using benthic food sources to supplement water‐column food sources by 20% generated surfclams that grew to observed size and biomass and exhibited spawn timing consistent with the known surfclam spawning season. The simulation results suggest that measured water‐column chlorophyll concentrations may underestimate the food available to the continental shelf benthos. Large continental shelf bivalves are an essential resource for fisheries and higher trophic level consumers. Understanding available and utilized food resources is important for predicting long‐term impacts of climate change on benthic secondary production and fishery yield on the continental shelf.  相似文献

In order to develop a microsatellite typing system for Lepeophtheirus salmonis (Krøyer), a DNA preparation method for individual sea lice suitable for analysis by polymerase chain reaction (PCR) was designed, and the DNA sequences of 50 L. salmonis microsatellite elements were determined. The microsatellites were composed of 60% perfect, 25% imperfect, and 15% compound repeats. Based on the flanking DNA sequences, four microsatellite‐PCR assays were optimized and used in a pilot study to analyse L. salmonis samples collected in Ireland, Norway and Scotland. Two of the microsatellite‐PCR assays targeted polymorphic loci amplifying seven and 10 alleles respectively. The results showed that microsatellite‐PCR typing could detect genetic variation both within and between the L. salmonis groups, and also was capable of amplifying group‐specific alleles.  相似文献

Different synthetic forms of supplemental methionine (Met) are produced commercially by various manufacturers. Limited information exists on the bioavailability of these different forms in commercially relevant fish species. The objective of this study was to compare the relative bioavailability of L‐methionine and a methionine hydroxy analogue calcium salt (MHA‐Ca) to DL‐methionine in a commercially relevant species, rainbow trout, using a linear slope‐ratio assay. In addition, the ability of the rainbow trout to effectively convert the D‐isomer of methionine into the biologically active L‐isomer was studied by comparing the relative bioavailability of L‐methionine to DL‐methionine. Nine (9) diets were created from a methionine‐deficient basal diet which was supplemented with graded equimolar levels of methionine (0.1%, 0.2%, 0.3% of diet by weight) from either DL‐methionine, L‐methionine and methionine hydroxy analogue calcium salt (MHA‐Ca). Diets were fed in triplicate (IBW = 24.0 ± 0.6 g) for 12 weeks using a pair‐feeding protocol. The basal diet was formulated to be deficient in both methionine (0.5%) and cysteine (0.3%), whereas meeting all other known nutritional requirements of rainbow trout. MHA‐Ca was found to be less available (P < 0.05) than DL‐methionine with relative bioavailability values of 69%, 60% and 73% based upon weight gain, growth rate (TGC) and retained nitrogen values respectively. No significant differences (P > 0.05) in bioavailability were found between DL‐methionine and L‐methionine. These findings lead to the conclusion that differences in bioavailability do exist between sources of synthetic methionine, additionally findings suggest that rainbow trout are able to effectively convert the D‐isomer of methionine into the L‐isomer without significantly affecting fish performance.  相似文献

During the austral summer of 1969-1970 bones of Lower Triassic vertebrates were excavated from coarse quartzose sandstones forming stream channel deposits of the Fremouw Formation at Coalsack Bluff, in the Transantarctic Mountains of Antarctica. This is the first assemblage of fossil tetrapods of significant geologic age to be found on the Antarctic Continent. The fossils include labyrinthodont amphibians, presumed thecodont reptiles, and therapsid reptiles, including the definitive genus, Lystrosaurus. This genus is typical of the Lower Triassic of southern Africa, and is also found in India and China. Lystrosaurus and associated vertebrates found in Antarctica were land-living animals: therefore their presence on the South Polar Continent would seem to indicate the contiguity of Antarctica, Africa, and India in Early Triassic times.  相似文献

This study examined the efficacy of bithionol as a prophylactic or therapeutic oral treatment for Atlantic salmon (AS), Salmo salar , affected by amoebic gill disease (AGD). Furthermore, it explored the interaction of bithionol oral therapy with the current standard treatment (a freshwater bath for at least 3 h). The efficacy of three medicated feeds was determined in the trial by feeding AGD-affected AS at 1% body weight (BW) day−1 either oil coated commercial feed (control) or prophylactic and therapeutic bithionol at 25 mg kg−1 feed. Feeding commenced 2 weeks prior to exposure to Neoparamoeba spp. at 300 cells L−1 and continued for 49 days post-exposure (PE). Bithionol when fed as a 2-week prophylactic or therapeutic treatment at 25 mg kg−1 feed delayed the onset of AGD pathology and reduced the percentage of gill filaments with lesions. Administration of a 3-h freshwater bath at 28 days PE significantly reduced amoeba numbers to a similar level across all treatments; in contrast, gross gill score and percent lesioned filaments were reduced to different extents, the control having a significantly higher score than both bithionol treatments. Following the freshwater bath, clinical signs of AGD increased at a similar level across all treatments, albeit controls were significantly higher than the bithionol treatments immediately following freshwater treatment. This study demonstrated that bithionol at 25 mg kg−1 feed, when fed as a 2-week prophylactic or a therapeutic treatment, delayed and reduced the intensity of AGD pathology and warrants further investigation as a treatment for AGD-affected AS.  相似文献

Eyespot is an economically important stem‐base disease of wheat caused by two fungal species: Oculimacula yallundae and Oculimacula acuformis. This study investigated the efficacy of two sources of resistance, viz. the genes Pch1, introgressed into hexaploid wheat from Aegilops ventricosa, and Pch2, identified in wheat cv. Cappelle Desprez, against O. yallundae and O. acuformis separately. In a series of seedling bioassays Pch1 was found to be highly effective against both species. Although Pch2 was found to confer resistance against both pathogen species, it was significantly less effective against penetration from O. yallundae than O. acuformis. Furthermore, a quantitative trait locus (QTL) analysis was not able to locate any resistance to O. yallundae on chromosome 7A of Cappelle Desprez. This has important implications for the use of Pch2 in commercial cultivars as it is necessary to have genes that confer resistance to both pathogens for effective eyespot control. In addition, a set of 22 T. monococcum accessions was screened for resistance to both O. yallundae and O. acuformis to identify potentially novel resistances and to assess the accessions for evidence of differential resistance to the eyespot species. Significant differences in resistance to the two pathogens were identified in four of these lines, providing evidence for differential resistance in T. monococcum. This study demonstrates that future screening for novel sources of eyespot resistance should investigate both pathogen species.  相似文献
