A case of a rapidly fatal disease in polo horses caused by African horsesickness (AHS) virus serotype 2 is described. The pattern of polo tournaments, environmental/management conditions (moist and warm), as well as probable importation strategies with no regard for import control and quarantine, favored introduction and spread of the virus. The outbreak, which involved a large number of horses, was characterized by severe respiratory distress, fever, supraorbital edema, and death. This is the first time a widespread epidemic of AHS has been reported in Nigeria and this is the first report of AHSV serotype 2 in the northern hemisphere. In addition, we amplified the complete genome of the virus using RNA extracted from clotted blood. This report indicates that AHS is expanding its geographical territory northwards and assuming a new microbial ecology. 相似文献

Two Triticum monococcum accessions were found to be highly resistant to the Russian wheat aphid. An attempt was made to transfer the resistance to common wheat through direct hybridization or by using bridge species. In each of the four crossing strategies tried, a gradual loss of resistance occurred as hybrids with higher ploidy levels were obtained. It appeared that the level of resistance observed was directly proportional to the ratio of the Triticum monococcum genome relative to other genomes. This would indicate suppression of the resistance gene(s) by the added genomes or dilution of its product(s) by those of homoeoloci. The degree of protection afforded by the gene(s) at the hexaploid level may prove to be small. Plants suspected to be homozygous for the resistance gene(s) were identified; however, further backcrossing to common wheat will be required to improve their agronomic types and meiotic regularities. 相似文献

Detection of bovine viral diarrhea virus by spot hybridization with probes prepared from cloned cDNA sequences 总被引:1,自引:0,他引:1
A cytopathic strain of bovine viral diarrhea virus (BVDV) was purified from infected cell culture fluids by isopycnic density-gradient centrifugation. Genomic RNA was extracted and tailed with adenine residues at the 3' end with poly-A polymerase. Double-stranded complementary DNA (cDNA) was synthesized, using the poly-A-tailed RNA as a template and oligo-dT as a primer, and then cloned into the pUC9 plasmid. Virus-specific cDNA sequences, varying in length from 0.5 to 2.5 kilobases (kb), were obtained. One BVDV-specific sequence of cloned cDNA, 1.1 kb in length and with an internal Pst I restriction endonuclease cleavage site, was selected for use as a probe. The cloned cDNA insert was removed from the plasmid either with or without flanking plasmid sequences and labeled with 32P-nucleotides by nick translation for use as hybridization probes for BVDV. The performance of probes of smaller fragments of the insert was compared to that of the intact sequence in hybridization assays. In addition, 2 methods of specimen preparation were compared to establish optimum parameters for hybridization. The hybridization assay was 10-100 times more sensitive than infectivity assays for BVDV in infected cell cultures. Freezing of specimens reduced by 10-fold the sensitivity of hybridization for BVDV target sequences. The probes prepared from the cloned cDNA hybridized with all cytopathic and noncytopathic BVDV strains tested but not with uninfected cell cultures, cellular ribosomal RNA, bovine coronavirus, bluetongue virus, or bovine adenovirus 3. The coastal shelf of the Gulf of Oman experiences periodic upwelling events during the summer months that are driven by the southwest monsoon. It is unclear what role these events play in the spatial and temporal distribution of the region’s fish assemblage. We carried out trials on two different video techniques to characterize the habitat and fish assemblage along the continental shelf margin near Muscat, Oman. Exploratory surveys with a drift stereo‐video revealed three main habitat types: Sand, Reef and Megabenthos. Three areas were chosen for additional sampling using stereo‐BRUVS (‘baited’ remote underwater‐video systems). On two separate occasions (November 2005 and March 2006) replicate stereo‐BRUVS were deployed in each area stratified by the main habitat types. For each teleost and elasmobranch species encountered on the video, an estimate of total body length and the relative abundance (MaxNi) was made. The stereo‐BRUVS recorded a wide range of demersal and pelagic teleosts including species of conservation interest such as sharks, rays and groupers. The drift stereo‐video recorded significantly fewer species than the stereo‐BRUVS (N = 15 versus N = 43). Species diversity from the stereo‐BRUVS increased by 96% in March 2006 (N = 41) compared to November 2005 (N = 23), a pattern consistent at all three areas. The structure of the overall fish assemblage (using canonical analysis of principal coordinates analysis) was highly variable both in time and space. There was ample evidence of strong habitat associations, particularly with depth and seasonal shifts in abundance and diversity. We argue the upward migration of oxygen‐depleted water into the shallow depths during the late monsoon displaces the demersal fish community along this coast. 相似文献

The kinetics of antibody formation in Holstein heifers after primary and secondary intranasal inoculation of bovine herpesvirus-1 (BHV-1) and after BHV-1-induced abortion was determined. Sera were fractionated by gel filtration and ion-exchange chromatography. The antibody activity within serum immunoglobulin (Ig) isotypes was assessed, using a plaque-reduction neutralization assay. The primary immune response to BHV-1 infection was characterized by the appearance of IgM and IgG antibodies in serum by postinoculation day (PID) 7. Maximal IgG antibody activity occurred at PID 35 in nonpregnant heifers and at PID 14 in pregnant heifers. Thereafter, IgG antibody activity declined slowly in both groups of heifers. Maximal IgM antibody activity occurred at PID 14 in both groups of heifers and declined rapidly thereafter. The IgG antibody activity during primary immune responses was restricted to the IgG1 subclass. Secondary responses were characterized by anamnestic IgG antibody responses. Antibody activity was present within the IgG1 and IgG2 subclasses during secondary immune responses, but the increase in antibody activity during this period was primarily in the IgG2 subclass. Secondary IgM antibody formation was elicited by abortion induced by the intra-amniotic inoculation of BHV-1, but not by reexposure by the intranasal route. Abortion occurred in 1 heifer 28 days after intranasal BHV-1 inoculation. Abortion in this heifer was not associated with a secondary antibody response. The nature of BHV-1 antigenic exposure in the bovine determined the relative distribution of anti-BHV-1 antibody activity in serum IgM, IgG1, and IgG2. The African buffalo (Syncerus caffer) is a natural reservoir host for both pathogenic and non-pathogenic Theileria species. These often occur naturally as mixed infections in buffalo. Although the benign and mildly pathogenic forms do not have any significant economic importance, their presence could complicate the interpretation of diagnostic test results aimed at the specific diagnosis of the pathogenic Theileria parva in cattle and buffalo in South Africa. The 18S rRNA gene has been used as the target in a quantitative real-time PCR (qPCR) assay for the detection of T. parva infections. However, the extent of sequence variation within this gene in the non-pathogenic Theileria spp. of the Africa buffalo is not well known. The aim of this study was, therefore, to characterise the full-length 18S rRNA genes of Theileria mutans, Theileria sp. (strain MSD) and T. velifera and to determine the possible influence of any sequence variation on the specific detection of T. parva using the 18S rRNA qPCR. The reverse line blot (RLB) hybridization assay was used to select samples which either tested positive for several different Theileria spp., or which hybridised only with the Babesia/Theileria genus-specific probe and not with any of the Babesia or Theileria species-specific probes. The full-length 18S rRNA genes from 14 samples, originating from 13 buffalo and one bovine from different localities in South Africa, were amplified, cloned and the resulting recombinants sequenced. Variations in the 18S rRNA gene sequences were identified in T. mutans, Theileria sp. (strain MSD) and T. velifera, with the greatest diversity observed amongst the T. mutans variants. This variation possibly explained why the RLB hybridization assay failed to detect T. mutans and T. velifera in some of the analysed samples. 相似文献

The effects of canine parvovirus (CPV) infection in dogs with hemolytic anemia was compared with the clinical effects of human parvovirus-induced aplastic anemia in human beings with chronic regenerative anemias. Phenylhydrazine was used to induce a transient, severe, hemolytic anemia in dogs to evaluate the effects of CPV infection on rapidly dividing bone marrow precursors. Erythrocyte colony-forming unit bone marrow cultures and cytologic examination of bone marrow were used to determine the effects of CPV infection on erythroid bone marrow precursors. The induced hemolytic anemia regenerated rapidly and although the bone marrow was infected, it was determined that CPV infection did not induce a detectable decrease in erythroid progenitors in dogs with severe hemolytic anemia. 相似文献

Human epidermal growth factor receptor type 2 (c‐erbB‐2), an oncoprotein with potential prognostic marker and therapeutic use, is overexpressed in several human and animal tumours. But information regarding this molecule in feline tumours is scarce. This study aimed to assess the changes in the immunohistochemical expression of c‐erbB‐2 in feline endometrial adenocarcinomas (FEA) compared to normal endometrium. An immunohistochemistry assay using a specific antibody against c‐erbB‐2 was performed in FEA samples (n = 34) and in normal endometrium in the follicular (FS; n = 12) and luteal (LS; n = 11) stages. In FEA, the c‐erbB‐2 immunoexpression was assessed in neoplastic epithelial cells whilst in normal endometria it was individually evaluated in the surface and the superficial and deep glandular epithelia (SE, SGE and DGE, respectively). In FS and in LS, all the epithelia were positive for c‐erbB‐2; positivity was higher in the SE and the SGE than in DGE. Twenty of the 34 FEA samples (58.8%) were positive for c‐erbB‐2 immunolabelling. Nevertheless, its expression was higher in all the epithelia in the FS compared to FEA (p ≤ 0.0001) or the LS (p = 0.016). The results presented herein suggest that c‐erbB‐2 molecule is differently expressed in the feline endometrium through the oestrous cycle and though it may also be involved in feline endometrial carcinogenesis, a question remains unanswered on the importance of additional pathways of epithelial proliferation in the neoplastic changes in feline endometrium. 相似文献
