Natural products derived from micro-organisms, plants and animals possess inherent biological properties and are, therefore, an excellent source of lead chemicals on which to base synthesis programmes employing rational design techniques. Only a fraction of the fungal kingdom has been screened systematically for novel metabolites despite their suspected potential. Moreover, the fungi are generally manipulable in laboratory culture systems and, because of their developmental versatility, they offer tremendous potential as sources of novel metabolites. Primary assay systems based on biochemical targets provide an effective means of screening microbial products; such assay systems also efficiently support chemical purification of lead activities and rapid elucidation of new chemical templates. 相似文献

Specific Polymerase Chain Reaction Primers for the Detection of Plasmodiophora brassicae in Soil and Water 总被引:1,自引:0,他引:1
ABSTRACT The development of specific oligonucleotide primers for Plasmodiophora brassicae has led to a nested polymerase chain reaction (PCR) detection method for P. brassicae in soil and water. Initially, the PCR was used to amplify a section of the rDNA repeat. The PCR products were sequenced and the data used to design primers that were directed at the ribosomal RNA genes and internal transcribed spacer regions. Specificity was tested against more than 40 common soil organisms, host plants, and spore suspension contaminants, as well as P. brassicae isolates from around Australia and the world. Sensitivity was determined to be 0.1 fentograms (fg; 10(-15) g) for pure template and as low as 1,000 spores per g of potting mix. In soil, P. brassicae was detected in all soils where the inoculum was sufficient to result in clubroot symptoms. Also outlined is a simple method of DNA extraction from soil. 相似文献

A 7-year-old spayed female Cocker Spaniel was hospitalized with a history of chronic vomiting, anorexia, and weight loss. Laboratory abnormalities included leukocytosis, metabolic alkalosis, hypoglycemia, hypoproteinemia, and hyperinsulinemia. Gastroscopy and ultrasonography revealed multiple gastric masses and a possible pancreatic mass, respectively. Examination of tissues obtained at necropsy showed a pancreatic adenocarcinoma with hepatic metastasis, gastric hypertrophy, and multiple duodenal ulcers. Immunocytochemical staining of the neoplasia was positive for pancreatic polypeptide (PP) and insulin and negative for gastrin, calcitonin, adrenocorticotropic hormone (ACTH), serotonin, L-enkephalin, chromagranin, glucagon, and somatostatin. Subsequent serum gastrin and PP assays showed a fasting hypergastrinemia with a normal response of gastrin to provocative testing and extremely increased PP values. The high PP values may have resulted in the vomiting and gastrointestinal ulceration. A PP-secreting tumor has not previously been reported in the dog. 相似文献

Eighteen cats were anesthetized and were randomly assigned to 2 groups (9 cats/group). Cardiopulmonary resuscitation (CPR) was performed on each cat, with the cat in dorsal (group 1) or lateral (group 2) recumbency, by administering 5 external cardiac compressions/1 interposed (diastolic) ventilation (American Heart Association [AHA] technique; n = 3 cats/technique), simultaneous compression and ventilation (SCV; n = 3 cats/technique), or SCV with 1 interposed (diastolic) ventilation every tenth compression (SCV/DV; n = 3 cats/technique). Fourteen of the 18 cats were resuscitated. Central aortic and venous pressures were determined concurrently with lead II electrocardiography. Arterial blood samples were collected during the base-line period (after pressure fluctuations had stabilized, with the cats spontaneously breathing room air), 2.5 minutes after the onset of arrest (defined as the cessation of aortic pressure fluctuations), and after 10 minutes of CPR. Arterial blood gas values during the base-line period or during the period of arrest were not significantly different between group-1 and group-2 cats. After CPR, arterial pH and bicarbonate values were not significantly different between groups or between technique categories. The PaCO2 values were significantly lower in cats resuscitated by SCV or by SCV/DV than in cats resuscitated by AHA (P less than 0.05). The PaO2 values were significantly (P less than 0.001) higher in group-2 cats than in group-1 cats and were significantly (P less than 0.001) different between each technique category, with cats resuscitated by AHA having the lowest PaO2 and cats resuscitated by SCV/DV having the highest PaO2. Body position, CPR technique, sex, weight, or arterial blood gas values after CPR were not predictors of successful resuscitation. 相似文献

Diagnosis of myeloid leukosis induced by a recombinant avian leukosis virus in commercial white leghorn egg laying flocks 总被引:4,自引:0,他引:4
Commercial white leghorn egg layer flocks being used to produce fertile eggs for human vaccine production exhibited dramatically low peaks in egg production, two to four times higher than normal weekly mortality, and high numbers of cull, nonlaying birds after the onset of sexual maturity. These lower production characteristics could not be associated with management-related problems. Gross lesions of cull and fresh dead birds necropsied showed approximately 60% lacked ovarian activity and had lesions of a bacterial bursitis or synovitis, whereas the other 40% had tumors of the viscera but not of the bursa of Fabricius. Histologic examination of tumor-containing tissues showed lesions typical of myelocytomatosis. The diagnosis of myeloid leukosis was confirmed by the isolation of a recombinant avian leukosis virus (ALV) containing the LTR of subgroup J and the envelope of subgroup B ALV. A positive polymerase chain reaction with primers specific for the 3' untranslated region LTR confirmed the presence of LTR of ALV-J. The source of infection with this recombinant ALV was not determined; however, it is likely that commingling of the day-old egg-type chicks with ALV-J-infected meat-type chicks in a common hatchery had contributed to this outbreak. 相似文献

OBJECTIVE: To evaluate CSF in horses with confirmed West Nile virus encephalomyelitis. DESIGN: Retrospective study. ANIMALS: 30 horses. PROCEDURE: Results of CSF analyses from horses with acute neurologic signs attributed to West Nile virus infection that was confirmed by immunoglobulin M antibody capture ELISA were reviewed and analyzed. RESULTS: Among 30 CSF samples, findings in 8 (27%) were within reference ranges and in 22 (73%) were abnormal. Among the 22 abnormal samples, mononuclear pleocytosis was found in 16 (73%) and high protein concentration with nucleated cell count within reference range was found in 6 (27%) samples. A predominance of lymphocytes was found in 11 of 16 samples with mononuclear pleocytosis, and a predominance of large mononuclear cells was found in 5 of 16 samples. Sensitivities of analyses of CSF obtained from the lumbosacral and atlanto-occipital regions of the spinal cord were 89 and 50%, respectively. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that in horses with acute onset of neurologic signs caused by West Nile virus encephalomyelitis, findings in the CSF are likely to be abnormal, mononuclear pleocytosis with lymphocytic predominance may be most commonly observed, and CSF collected from the lumbosacral region may be abnormal more commonly than CSF collected from the atlanto-occipital region. 相似文献
