Cattle are a natural reservoir of the food-borne pathogen Escherichia coli O157:H7. Therefore, strategies that reduce E. coli O157:H7 prior to slaughter will reduce human exposures to this virulent pathogen. When bacteria that can anaerobically respire on nitrate (e.g., E. coli) are exposed to chlorate, they die because the intracellular enzyme nitrate reductase converts nitrate to nitrite, but also co-metabolically reduces chlorate to cytotoxic chlorite. Because chlorate is bactericidal only against nitrate reductase-positive bacteria, it has been suggested that chlorate supplementation be used as a strategy to reduce E. coli O157:H7 populations in cattle prior to harvest. Cattle (n = 8) were fed a feedlot-style high-grain diet experimentally infected with three strains of E. coli O157:H7. Cattle were given access to drinking water supplemented with 2.5 mM KNO3 and 100 mM NaCl (controls; n = 4) or 2.5 mM KNO3 and 100 mM NaClO3 (chlorate-treated; n = 4). Sodium chlorate treatment for 24 h reduced the population of all E. coli O157:H7 strains approximately two logs (10(4) to 10(2)) in the rumen and three logs (10(6) to 10(3)) in the feces. Chlorate treatment reduced total coliforms and generic E. coli from 106 to 10(4) in the rumen and by two logs throughout the rest of the gastrointestinal tract (ileum, cecum, colon, and rectum). Chlorate treatment reduced E. coli O157:H7 counts throughout the intestinal tract but did not alter total culturable anaerobic bacterial counts or the ruminal fermentation pattern. Therefore, it appears that chlorate supplementation is a viable potential strategy to reduce E. coli O157:H7 populations in cattle prior to harvest. 相似文献

Cultivation of the woody legume Sesbania sesban in agroforestry systems has been widely promoted for soil fertility improvement in Africa. Despite the existence of natural populations in eastern Zambia, considerable seed originating from Kenya has been introduced to the area, while further introductions from Malawi are planned. Large-scale anthropogenic seed dispersal raises concerns regarding genetic management, which were addressed here by random amplified polymorphic DNA (RAPD) analysis. Eleven populations of Sesbania from southern and East Africa were assessed, including S. sesban populations already or likely to be introduced into eastern Zambia and reference taxa. Seventeen primers revealed a very large number of polymorphisms (538) in surveyed material. Differentiation among S. sesban populations was unusually high for a woody perennial (70% of variation among populations), with all individuals stringently assigned to specific populations in cluster analysis, even for geographically proximate sites. High differentiation, combined with the unusually large number of polymorphisms determined, resulted in the identification of a number of population-diagnostic markers. High differentiation may reflect the restricted ecology of S. sesban, human disturbance and frequent natural fluctuations in niche availability. Hydrochory, posited as a mechanism for gene flow between populations located along the same drainage system, appears to be discounted. Geographic proximity was not always a good guide for genetic similarity among S. sesban populations. In particular, rather proximate populations from Zambia and Malawi appeared highly differentiated genetically, while Zambian populations were relatively similar to a geographically distant population from Namibia. Differentiation was consistent with the presence of two botanic varieties in the southern Africa region, vars. nubica and zambesiaca. Estimates of Nei's genetic diversity suggested that S. sesban populations from East Africa were most diverse. Overall, population diversity estimates were low, possibly due in part to frequent bottlenecks, short longevity and a short juvenile phase. The unusually large number of polymorphisms detected here provided an opportunity to test for data redundancy and primer selection methodologies during RAPD analysis. Combined data from a subset of eight primers selected according to three different approaches provided a very good overall fit to the complete data set (rM > 0.95). Different primer selection approaches influenced the reliability of clustering and diversity analysis, with a minimum of eight primers required for reliable ranking of diversity estimates. The implications of our data for conservation, cultivation and monitoring of S. sesban in the southern Africa region are discussed. It is now possible to use rigorous quantum scattering theory to perform accurate calculations on the detailed state-to-state dynamics of chemical reactions in the gas phase. The rapid growth of Atlantic salmon, Salmo salar L., culture in north-western Europe has given rise to concerns regarding the biological consequences of fish farm escapes on wild salmonid populations. Canthaxanthin, a carotenoid pigment additive to farmed salmon feed which is passed from females to their progeny, may be used as an indicator of the numbers of escaped farmed salmon which spawn in the wild. In the present study, thin-layer chromatography and high-performance liquid chromatography (HPLC) were used to screen emergent Atlantic salmon fry sampled from seven river catchments in Ireland for canthaxanthin. The incidence of fry containing canthaxanthin at greater than trace levels (<5% of total carotenoid pigment) was 0–4%, with an average of 1.7%, among the seven rivers sampled, indicating that the progeny of farmed salmon were present at similarly low frequencies. Canthaxanthin was detected at trace levels in an unexpectedly high proportion (35%) of salmon fry. Canthaxanthin was present at levels exceeding trace amounts in 24% of 21 non-anadromous brown trout, Salmo trutta L., sampled from six Irish rivers and present at trace levels in a further 57% of the fish, indicating that dietary canthaxanthin is freely available to salmonids in Irish rivers. The widespread presence of trace levels in salmon fry may be attributable, at least in part, to the increased sensitivity of the HPLC methods and to rapid dietary uptake during early post-emergence feeding. 相似文献

For the vast majority of mammalian genes, maternally- and paternally-derived alleles behave identically and are either expressed or repressed, regardless of whether they were inherited from egg or sperm. For imprinted genes, however, this is not the case. The alleles of imprinted genes are epigenetically modified in a parent-of-origin-specific manner and, as a consequence, maternally- and paternally-derived alleles behave differently. Typically one allele is expressed while the other is silent. Although relatively few in number, imprinted genes are the focus of intensive study, as they have important roles in embryonic development. Abnormal expression of imprinted genes results in growth disorders and is implicated in several clinical conditions. Most studies of imprinted genes have been performed in rodents or primates, with limited studies in other mammals such as bovine and opossum. We have recently demonstrated the existence of imprinted genes in the canine, by showing that the canine insulin-like growth factor 2 receptor gene ( IGF2R ) is monoallelically expressed, with predominant expression of the maternally-derived allele and repression of the paternally-inherited allele. Our ultimate goal is to characterize all imprinted genes in the canine, and to understand how they contribute to canine reproduction, development and disease. Proline is typically the most abundant amino acid present in grape juice and wine. The amount present is influenced by viticultural and winemaking factors and can be of diagnostic importance. A method for rapid routine quantitation of proline would therefore be of benefit for wine researchers and the industry in general. Colorimetric determination utilizing isatin as a derivatizing agent has previously been applied to plant extracts, biological fluids, and protein hydrolysates. In the current study, this method has been successfully adapted to grape juice and wine and proved to be sensitive to milligram per liter amounts of proline. At sugar concentrations above 60 g/L, interference from the isatin-proline reaction was observed, such that proline concentrations were considerably underestimated in grape juice and dessert wine. However, the method was robust for the analysis of fermentation samples and table wines. Results were within ±10% agreement with data generated from typical HPLC-based analyses. The isatin method is therefore considered suitable for the routine analysis required to support research into the utilization or release of proline by yeast during fermentation. 相似文献

AIM: To describe outcomes for horses diagnosed with incomplete, non-displaced fractures of the frontal plane of the radial facet (INFR) of the third carpal bone (C3) treated by placement of a lag screw across the fracture under arthroscopic guidance. METHODS: Horses (n=13) diagnosed with INFR and treated between December 1999 and January 2005 using a lag screw placed over the fracture were studied. For each case, five horses matched for sire, age and sex which were not known to have INFR were sought for comparison. Racing performance data were collected from a commercial online database. The racing performance of cases pre- and post-operatively, and of cases and matched horses in the post-operative period was compared. RESULTS: Sixteen INFR were found in the 13 horses. Radiographic evidence of healed fracture lines 2–4 months after surgery was seen in 11/16 (69%) fractures; 11/13 (85%) cases raced again after a median recovery period of 292 (range 149–681) days. Treatment was considered successful in 9/13 (69%) cases, which were still in training or had been retired for reasons other than lameness localised to the middle carpal joint at the end of the study period. Just 6/13 (46%) cases had raced prior to injury. The racing ability pre- and post-operatively of five cases was compared, three (60%) of which performed better post-operatively than they had before. There was no significant difference in racing longevity or ability post-operatively between patients and matched (control) horses. CONCLUSIONS: Post-operatively, there was little difference in the racing performance between horses diagnosed with INFR which had a lag screw placed across the fracture line and horses matched for sire, age and sex which were not known to have INFR. CLINICAL RELEVANCE: Horses which were diagnosed with INFR of C3 and had a lag screw placed across the fracture had a good prognosis for future racing performance. 相似文献

A reconstruction of past environmental change from Ecuador reveals the response of lower montane forest on the Andean flank in western Amazonia to glacial-interglacial global climate change. Radiometric dating of volcanic ash indicates that deposition occurred ~324,000 to 193,000 years ago during parts of Marine Isotope Stages 9, 7, and 6. Fossil pollen and wood preserved within organic sediments suggest that the composition of the forest altered radically in response to glacial-interglacial climate change. The presence of Podocarpus macrofossils ~1000 meters below the lower limit of their modern distribution indicates a relative cooling of at least 5°C during glacials and persistence of wet conditions. Interglacial deposits contain thermophilic palms suggesting warm and wet climates. Hence, global temperature change can radically alter vegetation communities and biodiversity in this region. 相似文献

The isozyme patterns of lactate dehydrogenases of various tissues were studied on 51 hagfish by starch-gel electrophoresis. Nine lactate dehydrogenase phenotypes were encountered, suggesting the coexistence of two alleles at each of the two separate gene loci. There apparently was no interaction between the products of these two separate loci. Even the products of two alleles at the same locus were apparently incapable of forming hybrid molecules, an indication of the possible monomeric nature of each lactate dehydrogenase molecule. 相似文献
