Evolution is an adaptive walk through a hypothetical fitness landscape, which depicts the relationship between genotypes and the fitness of each corresponding phenotype. We constructed an empirical fitness landscape for a catalytic RNA by combining next-generation sequencing, computational analysis, and "serial depletion," an in vitro selection protocol. By determining the reaction rate constant for every point mutant of a catalytic RNA, we demonstrated that abundance in serially depleted pools correlates with biochemical activity (correlation coefficient r = 0.67, standard score Z = 7.4). Therefore, enumeration of each genotype by deep sequencing yielded a fitness landscape containing ~10(7) unique sequences, without requiring measurement of the phenotypic fitness for each sequence. High-throughput mapping between genotype and phenotype may apply to artificial selections, host-pathogen interactions, and other biomedically relevant evolutionary phenomena.  相似文献

An outbreak of melioidosis, a bacterial infection caused by Pseudomonas pseudomallei, was identified in a batch of feral cynomolgus monkeys (Macaca fascicularis) imported to Britain from the Philippines. Thirteen confirmed or possible cases occurred among a batch of 50 animals. Subsequent investigations revealed that the infection was uncommon among imported primates from a variety of sources, although three other cases were identified in monkeys imported from Indonesia. The majority of the affected monkeys had splenic abscesses, and hepatic abscesses and infections of the soft tissues and skin were also frequently observed. Most of the infected animals had no clinical signs despite extensive abscesses, and the presence of infection was only suspected when they were shown to have serum antibodies to P pseudomallei by an enzyme-linked immunosorbent assay. Although there was no evidence of cross infection of other animals or human handlers, this outbreak is a reminder of the dangers of working with wild-caught primates and the potential for the establishment of environmental foci of melioidosis.  相似文献

AIM: To identify if there was a significant difference in the average daily liveweight gain (ADG) between Thoroughbred foals weaned using two weaning protocols commonly used in New Zealand.

METHODS: Sixteen Thoroughbred foals were blocked for sex and age, randomly allocated into progressive (187, SD 33 days; three colts, five fillies) or abrupt (182, SD 28 days; four colts, four fillies) weaning groups, and weighed every second day for 2 weeks either side of weaning, then fortnightly from birth to 480 (SD 31) days old. ADG was calculated to examine the short-term (10 days before weaning, 5 and 10 days post weaning) and long-term (0–6 and 6–16 months of age) effect of the two weaning treatments.

RESULTS: ADG was 1.10 (SD 0.091) kg/day before weaning (0–6 months of age) and 0.59 (SD 0.06) kg/day from weaning to 480 (SD 31) days old. At the start of weaning, liveweights of the progressive and abrupt weaning groups were 276.5 (SD 40.3) kg and 257 (SD 15) kg, respectively (p=0.23). For the 5-day period during weaning, irrespective of treatment, there was a significant decrease in ADG of –0.29 (SD 0.49) kg/day and –0.15 (SD 0.30) kg/day for progressive and abrupt weaning, respectively. There was no significant difference in ADG between weaning methods at any measurement period (short or long term) during and after weaning. However, there was large variation between foals in ADG in the 10 days after the weaning process, which may indicate variation in individual foals' responses to being weaned, rather than the weaning treatment.

CONCLUSIONS: Weaning, irrespective of method, was associated with a decrease in ADG in the first week after weaning. The method of weaning had no effect on post-weaning ADG either short term, 10 days after weaning, or long term up to 480 days of age. Practically, it may be more important to consider maturity and liveweight as criteria for weaning foals rather than age alone.  相似文献

An outbreak of equine influenza (EI) caused by influenza A H3N8 subtype virus occurred in the Australian states of Queensland and New South Wales in August 2007. Infection in the Australian horse population was associated with the introduction of infection by horses from overseas. The first case of EI in Queensland was detected on 25 August 2007 at an equestrian sporting event. Infection subsequently spread locally and to other clusters through horse movements prior to the implementation of an official standstill. There were five main clusters of infected properties during this outbreak and several outliers, which were investigated to find the potential mechanism of disease spread. To contain the outbreak, Queensland was divided into infection status zones, with different movement controls applied to each zone. Vaccination was implemented strategically in infected areas and within horse subpopulations. Control and eventual eradication of EI from Queensland was achieved through a combination of quarantine, biosecurity measures, movement control, rapid diagnostic testing and vaccination.  相似文献

Blood gastrin and pepsinogen responses to a single infection with 100,000 Ostertagia ostertagi infective larvae in lactating dairy cows were investigated. None of the infected cows showed signs of clinical ostertagiasis, nor was there any difference in live weight gain, milk yield or faecal egg count between groups. Pepsinogen levels of the infected group were significantly elevated between days 3 and 24 after infection (peak 1041 mU tyrosine; day 14). In contrast, there was no significant difference in blood gastrin levels between infected and control animals suggesting that few adult worms had become established in the former group. These data are compared with the increases in both gastrin and pepsinogen levels recorded in susceptible calves exposed to the same level, pattern and strain of ostertagia infection in a previous experiment. It is suggested that gastrin assay may be of value in adult cattle for indicating when elevated pepsinogen levels are merely associated with a rise in larval intake and not with the establishment of large adult worm burdens.  相似文献
