Poor sperm viability post-thaw has resulted in constant research into methods of cryopreservation of canine semen. One factor that may be involved in poor viability is sperm oxidative stress caused by excessive formation of reactive oxygen species. The present study was performed in order to evaluate the effect of different concentrations of ascorbic acid (AA) and glutathione (Glu) added to an extender for the freeze-thawing of dog sperm. Semen from five mature dogs was collected and frozen in two studies. Prior to and after freezing, sperm motility, morphology and membrane status were examined. In addition, sperm motility was examined up to 120 min after thawing to evaluate thermo-resistance. In study I, semen was collected twice from each dog. On both occasions, semen was divided into three aliquots: control, Glu 1 m m and Glu 5 m m . In study II, semen was collected twice and divided into three aliquots; control, AA 50 μ m and AA 250 μ m . Initial sperm motility was significantly higher in sperm diluted with AA 50 μ m ; sperm longevity, however, measured by a thermal-resistance test (TRT), was higher for Glu treatments. Higher concentration of Glu produced significant improvement in TRT and membrane status, whereas higher concentration of AA had a negative impact in sperm longevity. Antioxidant supplementation to semen freezing extenders improved semen quality post-thaw. Moreover, Glu had the most beneficial effect when supplemented at 5 m m . 相似文献

Doppler velocity observations obtained by the Global Oscillation Network Group (GONG) instruments directly measure the nearly steady flows in the solar photosphere. The sun's differential rotation is accurately determined from single observations. The rotation profile with respect to latitude agrees well with previous measures, but it also shows a slight north-south asymmetry. Rotation profiles averaged over 27-day rotations of the sun reveal the torsional oscillation signal-weak, jetlike features, with amplitudes of 5 meters per second, that are associated with the sunspot latitude activity belts. A meridional circulation with a poleward flow of about 20 meters per second is also evident. Several characteristics of the surface flows suggest the presence of large convection cells. 相似文献

Arrest of cells in G0/G1 cell cycle phase is desired for nuclear transfer procedures. Serum starvation and cell cycle inhibitors are different ways to induce synchronization of the cell cycle. This study investigated the effects of serum starvation and cycloheximide (CHX) on the cell cycle of low (5th) and high (15th) passages fetal porcine fibroblasts. Cell cycle phases were determined using fluorescent activated cell sorting. Fifth passage fibroblast cultures had higher (p < 0.05) proportion of cells in G0/G1 only after 72 h of serum starvation (77.60 ± 0.65) when compared with non‐starved cells (71.44 ± 1.88). Serum starvation for all periods tested induced an increase (p < 0.05) on proportion of cells in G0/G1 on the 15th passage. No significant differences were observed on the 5th passage cultures exposed to CHX, although, on the 15th passage an increase on proportion of cells was observed after all periods of exposure (p < 0.05). These data indicates that high passage cells in vitro are more susceptible to serum starvation and CHX G0/G1 synchronization. 相似文献

We have investigated, under the normal conditions of local Chinese pig farming, castration of young male pigs by vaccination with a newly developed vaccine against gonadotrophin releasing hormone (GnRH). Because of the very early onset of puberty, long fattening period and relatively harsh circumstances in Chinese pig production, an investigation of the endocrine response of Chinese breeds to this type of vaccination was of particular interest. Fifteen crossbred boars (Yorkshire × Yanan) from three different litters were randomly assigned to three groups of five animals each. The first group was immunized at 13 weeks of age with a GnRH tandem dimer OVA‐conjugate in Specol and received a booster immunization 8 weeks later. The second group was injected with Specol alone and served as untreated controls. The remaining group was surgically castrated at the time of weaning (at 6 weeks of age). Pigs were fed ad libitum from weaning onwards. All animals were slaughtered at 31 weeks of age. Immunized boars had undetectable or low serum testosterone (0.09 ± 0.12 ng/ml), low fat androstenone (0.05 ± 0.01 μg/g) levels and very low testes weights (19.1 ± 4.3 g). Intact controls had much higher serum levels of testosterone (9.76 ± 4.81 ng/ml), fat androstenone levels (2.26 ± 0.87 μg/g) and testes weights (114.3 ± 29.41 g) at slaughter. Both the immunized and castrated group grew significantly faster than intact boars (p < 0.01). Average daily gains in immunized, castrated and intact animals were 0.69 ± 0.08, 0.63 ± 0.05 and 0.42 ± 0.07 kg (mean ± SD), respectively. The present data demonstrate for the first time that the newly developed anti‐GnRH vaccine works very well under practical Chinese pig farming conditions, and can be an attractive alternative to surgical castration. 相似文献

Forest vertebrates are critical to the subsistence of many tropical forest dwellers enjoying little or no access to other sources of animal protein. Yet the ecological and socioeconomic value of forest wildlife is being undermined as many large vertebrate populations are driven to local extinction by unsustainable hunting practices. Although large mammals that are preferred by hunters are widely variable in their morphology and ecology, they share a set of life history traits, which make them particularly vulnerable to overhunting. In this paper we compile data on game harvest from 31 tribal and nontribal settlements in Neotropical forests to examine how mammal assemblages are affected by the history of hunting within settlement catchment areas. The structure of hunter-kill profiles is related to settlement age and size in an attempt to understand how changes in hunting pressure may affect prey selectivity and the structure of residual game assemblages. There was a predictable shift from a few large-bodied to several small-bodied species harvested by increasingly older villages. Settlement persistence thus explained a significant proportion of the variation in mean body mass and species richness of mammals harvested. We conclude that differences in prey species profiles obtained by subsistence hunters of different ethnic groups can be largely explained by the local depletion status of game stocks, particularly large mammals, rather than by cultural factors. 相似文献

Two horses were diagnosed as having Mycobacterium ulcerans infections. The first was a 21-year-old Quarterhorse-cross mare living in Mallacoota (a coastal town near the border of New South Wales and Victoria, Australia) that presented with lichenification, hair-loss and oedema on a fetlock, which subsequently ulcerated, as well as a non-healing ulcer on the wither. The second horse was a 32 year-old Standardbred gelding from Nicholson, near Bairnsdale, Victoria, that had an ulcerated lesion on its caudal thigh. Histologically, there were characteristic changes seen with M. ulcerans infections in other species, including extensive necrosis without associated granulomatous inflammation. The organisms were seen in Ziehl-Neelsen-stained smears or sections of the lesions from both horses and were isolated in culture from the first horse. A definitive diagnosis was provided by real-time polymerase chain reaction targeting the M. ulcerans-specific insertion sequence, IS2404. Delayed identification of the infectious agent in the first case led to the use of suboptimal antimicrobial therapy, resulting in failure to control the infection and the horse was subsequently euthanased. The second horse was successfully treated following surgical debulking of the centre of the lesion and one session of aggressive cryosurgery. Mycobacterium ulcerans should be considered in the differential diagnosis of unexplained lichenification with oedematous and ulcerated skin lesions in horses living in regions where this organism is endemic. 相似文献

Ultraviolet light analysis of a fossil of the theropod dinosaur Scipionyx samniticus revealed that the liver subdivided the visceral cavity into distinct anterior pleuropericardial and posterior abdominal regions. In addition, Scipionyx apparently had diaphragmatic musculature and a dorsally attached posterior colon. These features provide evidence that diaphragm-assisted lung ventilation was present in theropods and that these dinosaurs may have used a pattern of exercise physiology unlike that in any group of living tetrapods. 相似文献

The role of climatic variation in regulating marine populations and communities is not well understood. To improve our knowledge, the sign, amplitude, and frequency of climatic and biotic variations should be compared as a necessary first step. It is shown that there have been large interannual and interdecadal sea-surface temperature changes off the West Coast of North America during the past 80 years. Interannual anomalies appear and disappear rather suddenly and synchronously along the entire coastline. The frequency of warm events has increased since 1977. Although extensive, serial, biological observations are often incomplete, it is clear that climate-ocean variations have disturbed and changed our coastal ecosystems. 相似文献

After nearly a century of recovery from overhunting, sea otter populations are in abrupt decline over large areas of western Alaska. Increased killer whale predation is the likely cause of these declines. Elevated sea urchin density and the consequent deforestation of kelp beds in the nearshore community demonstrate that the otter's keystone role has been reduced or eliminated. This chain of interactions was probably initiated by anthropogenic changes in the offshore oceanic ecosystem. 相似文献
