We test previous claims that the bacteria Vibrio alginolyticus produces tetrodotoxin (TTX) when living in symbiosis with the nemertean Lineus longissimus by a setup with bacteria cultivation for TTX production. Toxicity experiments on the shore crab, Carcinus maenas, demonstrated the presence of a paralytic toxin, but evidence from LC-MS and electrophysiological measurements of voltage-gated sodium channel–dependent nerve conductance in male Wistar rat tissue showed conclusively that this effect did not originate from TTX. However, a compound of similar molecular weight was found, albeit apparently non-toxic, and with different LC retention time and MS/MS fragmentation pattern than those of TTX. We conclude that C. maenas paralysis and death likely emanate from a compound <5 kDa, and via a different mechanism of action than that of TTX. The similarity in mass between TTX and the Vibrio-produced low-molecular-weight, non-toxic compound invokes that thorough analysis is required when assessing TTX production. Based on our findings, we suggest that re-examination of some published claims of TTX production may be warranted. 相似文献

Maturing male and female Atlantic salmon (Salmo salar L.) were held under three temperature regimes for 10 weeks between September and December: warm (constant 14–16 °C), ambient (decreasing from 11 to 5 °C), and cold (decreasing from 7 to 3 °C). Blood samples were analyzed for plasma steroid levels, and the fish were inspected for the presence of expressible milt (total volume and spermatocrit) and ovulation weekly. Samples of eggs were dry-fertilized with milt stripped from three males held at the same temperatures and incubated until the eyed stage. In females, levels of plasma testosterone (T) and 17β-oestradiol (E2) dropped as ovulation approached, concurrent with a rapid increase in levels of plasma 17α,20β-dihydroxy-4-pregnen-3-one (17,20β-P). In males, levels of T and 11-ketotestosterone (11-KT) peaked 2–3 weeks after the first appearance of expressible milt, while levels of 17,20β-P increased steadily and did not exhibit a definite peak. Exposure of females to cold water amplified and advanced the profiles of all three steroids compared with the ambient group, and increased the survival rates to the eyed egg stage. Cold water had no immediate effect on the male steroid profiles, but later, higher levels of 17,20β-P were evident compared with both the ambient controls and the warm water group, while the effects on 11-KT and T were more variable. Exposure to warm water completely inhibited both milt production and ovulation. Moreover, warm water modulated the steroid profiles of the males with lower 11-KT levels compared with ambient controls and lower 17,20β-P level compared with cold-water-treated males. In females, warm water resulted in total inhibition of the peri-ovulatory peak in 17,20β-P and prevented the normal decline of T and E2 levels associated with ovulation. The findings of the present study are highly relevant for broodstock management in aquaculture, as well in understanding the impact of climate change/temperature variability on wild salmon spawning. 相似文献

- Dredging blue mussels (Mytilus edulis) and thus removing structural elements, inducing resuspension of sediment as well as reducing filtration capacity, will inevitably affect the ecosystem. The study demonstrates that the impacts of fishing can be reduced through gear developments.
- A new light dredge was tested on commercial vessels using two different experimental setups. First, a twin haul experiment tested the standard gear (i.e., a Dutch dredge) against the light dredge by fishing the two gears side by side onboard the same vessel. Second, a single dredge experiment tested the absolute performance of the two gears by fishing in areas with a known blue mussel density.
- Results from the twin haul experiment demonstrate that the weight of sediment retained in the gear per square metre fished is 49% less in the light dredge compared with the Dutch dredge which will reduce resuspension of sediment at the surface. Also, the drag resistance of the light dredge was significantly less (177.1 vs. 202.7 kg m‐1). In the twin haul experiment no significant difference was found in the catch per unit effort (CPUE) of the two gears. The single dredge experiment, on the other hand, demonstrated a significant increase in CPUE exceeding 200% when using the light dredge.
- Seafloor tracks made by the two dredges could not be distinguished by use of side‐scan sonar and the tracks were still detectable 2 months after fishing.
- It was concluded that replacement of the Dutch dredge with the light dredge would reduce the impact of the fishery on the ecosystem by (i) reducing resuspension of sediment, (ii) reducing fuel consumption, and (iii) potentially reducing energy transfer to the sediment through a reduced gear drag resistance. A potential increase in catch efficiency may reduce the area affected.
- Fishing with the light dredge is discussed in relation to management of Natura 2000 sites.

Optimal water quality is considered as being a restriction for marine copepod cultures for live feed. There is a lack of knowledge on the water‐quality conditions in copepod cultures and the effect on copepods. Few studies have investigated the effect of ammonia on copepods, and fewer reports No Observed Effect Concentrations (NOEC) and Lowest Observed Effect Concentrations (LOEC), which provides safety levels before cultures are affected. This study investigates the tolerance of Acartia tonsa nauplii and adults to ammonia, using mortality as the endpoint after 24, 48 and 72 h of exposure. Nauplii were exposed to levels from 0 to 5127 μg NH3 L?1 and adults to levels from 0 to 8481 μg NH3 L?1. Nauplii NOEC was 30 μg NH3 L?1 and LOEC was 81 μg NH3 L?1. Adult NOEC was 477 μg NH3 L?1 and LOEC was 1789 μg NH3 L?1. 50% Lethal Concentrations (LC50) for nauplii of 48 and 72 h was 1257 and 220 μg NH3 L?1. LC50 for adults was 2370 (24 h), 972 (48 h) and 770 (72 h). Combining NOEC with excretion rates of NH4/NH3 a model was developed to calculate densities in batch cultures. We recommend that for batch cultures of A. tonsa, NH3 is kept below NOEC for nauplii and that levels of NH3 together with pH are monitored weekly. 相似文献

Among farmers there is a growing interest for adoption of reduced tillage practices, which has accentuated the need to understand the consequences for soil nutrient dynamics and losses. A laboratory study was conducted with structurally intact soil cores collected from two depths, 0–4 and 14–18 cm, within tillage experiments on contrasting soil types, both experiments with soil under mouldboard ploughing (MP) or shallow tillage (ST). The soil cores were adjusted to one of seven matric potentials ranging from ?1500 to ?15 hPa. The extent and regulation of nitrous oxide (N2O) evolution as a function of tillage, depth and soil characteristics was studied by measurement of N2O and CO2 evolution rates, as well as nitrifying and denitrifying potentials, and subsequent data analysis by multiple linear regression models. At both sites, compaction of ST soil below the depth of tillage was significant. The vertical distribution of N2O evolution was different in MP and ST soil, but no main effect of tillage on N2O evolution was observed. Effects of soil variables on N2O evolution were analysed using volumetric water content, water-filled pore space, or relative gas diffusivity (RD) to represent the effect of soil water. Using RD weakened interactions with tillage and C availability and strengthened main effects, suggesting that RD may provide a more general representation of the water effect. At 0–4 cm depth, N2O evolution was related to NO3? availability in the soil with 5.1% C, but to C availability in the soil with 1.5% C. The contrasting patterns of dependencies in the different environments support the interpretation of reduced tillage and soil water content as indirect controls, via diffusional constraints, of N2O evolution. 相似文献

The contents of alkylresorcinols (AR) were analyzed in 131 winter wheats, 20 spring wheats, 10 durum wheats, 5 spelt wheats, and 10 early cultivated forms of wheat (5 diploid einkorn and 5 tetraploid emmer), which are part of the HEALTHGRAIN diversity screen. AR were analyzed by gas chromatography (GC), which provides both total contents and relative homologue compositions, as well as with a Fast Blue colorimetric method that provides only total contents but which is fast and easily screens a large number of samples. There was considerable variation in the total AR content analyzed with GC: winter wheat (220-652 microg/g of dm), spring wheat (254-537 microg/g of dm), durum wheat (194-531 microg/g of dm), spelt (490-741 microg/g of dm), einkorn (545-654 microg/g of dm), and emmer wheat (531-714 microg/g of dm). The relative AR homologue composition was different for different types of wheat, with a C17:0 to C21:0 ratio of 0.1 for winter, spring, and spelt wheats, 0.04 for einkorn and emmer wheat, and 0.01 for durum wheat. The total AR content analyzed with the Fast Blue method was lower than that analyzed with GC but there was a good correlation between the two methods (R(2) = 0.76). 相似文献

Production of Atlantic salmon smolts in recirculation aquaculture systems (RAS) is growing, and novel production protocols using continuous light in RAS are being implemented in the industry. In the present study, Atlantic Salmon parr were exposed to either a traditional protocol (short-day winter signal [12:12 L:D] for 6 weeks) or to continuous light. Both photoperiods were applied in freshwater (FW) and brackish water RAS. Salmon from all treatments were transferred to seawater pens at 200 and 600 g and grown until slaughter size. A control group was smoltified with a 6-week short-day winter signal and kept in FW until sea transfer at 100 g. Continuous light gave a higher growth rate in RAS but reduced feed intake and growth and increased feed conversion ratio during the first 8 weeks in seawater. However, at slaughter, fish exposed to continuous light was bigger than fish given a winter signal because of the higher growth rate in RAS. Slaughter weight was lowest in fish transferred to sea at 600 g, despite having the highest day-degree sum during their life span. The best performing group was the control group transferred at 100 g. All treatments handled transfer to seawater and survival and maturation were not affected by the treatments in RAS. The immune status was examined with a multigene expression assay on BioMark HD platform from parr stage to 5–7 months after seawater transfer. Overall, there was no significant effect of photoperiod or salinity on the expression of the selected immune genes. In sum, the results from this study indicate that using continuous light in RAS may have negative effects on performance shortly after transfer in fish transferred to sea at 200 g, whereas at 600 g, all treatments had reduced growth after transfer irrespective of treatment in RAS. 相似文献

Fly-ash particles accumulate in sediments and can be used to assess spatial distribution and temporal trends of atmospheric deposition of pollutants derived from high temperature combustion of fossil fuels. Previous work has concerned fly-ash derived from oil and coal. Oil-shale is the main fossil fuel used in Estonia and a major source of atmospheric pollution in the Baltic states. To assess if oil-shale power plants produce specific fly-ash particles, we used scanning electron microscopy (SEM) and energy dispersive X-ray analysis (EDX) to compare fly-ash particles from oil-shale combustion with particles from oil and coal combustion. Two types were analysed, large black (10–30 μm) and small glassy (<5 μm) spheroidal particles. Although particle morphology to some extent is indicative of the fuel burnt, morphological characters are not sufficient to differentiate between particles of different origin. However, our results indicate that with EDX analysis the fly-ash from oil-shale can be distinguished from oil and coal derived particles in environmental samples. Concentrations of large black and small glassy spheroidal fly-ash particles in a sediment core from an Estonian lake showed similar trends to oil-shale combustion statistics from Estonian power plants. 相似文献

Remediation of 150 000 m3 of sediment containing400 kg PCB was performed by suction dredging in asmall lake in Sweden in the summers of 1993 and 1994.Upon project completion calculations indicated that97% of the PCB was deposited in a landfill withsubsequent low transport to the surroundings, andthat 2.9 kg was left in the lake sediment.Concentrations of PCB that were measured weekly in thewater leaving the lake were 12 ng/L (geometric mean)during dredging, not exceeding 30 ng/L and notsignificantly higher than in investigations preceedingthe remedial action. The concentration of PCB wasnegatively correlated to water discharge andpositively correlated to water temperature. PCBtransport in the river was correlated to waterdischarge but not to the dredging activity.
