Enhanced phytoextraction of heavy metals using chelating agents and agricultural crops is widely discussed as a remediation technique for agricultural soils contaminated with low mobile heavy metals. In this study, phytoextraction efficiency of Zea mays after single and split applications of EDTA was tested on the laboratory and the field scale. EDTA effectively increased the mobility of target heavy metals (Pb and Cd) in the soil solution. Split applications provided generally lower water-soluble levels of Pb and Cd both in the pot and the field experiment. Therefore, the risk of groundwater contamination may be reduced after split applications. Higher Pb and Cd mobilisation after single applications increased plant stress, phytotoxicity and reduced plant dry above-ground biomass production compared to corresponding split doses. Single doses enhanced plant uptake of Pb and Cd and the phytoremediation efficiency compared to corresponding split doses. Results of plant dry above-ground biomass and heavy metal uptake obtained from the pot experiment could be to some extent verified in the field experiment. Plant uptake of Pb and Cd was lower and biomass production dropped after EDTA additions in the field experiment. Remediation factors in the field experiment were in general significantly lower than in the pot experiment mainly due to the much higher mass of soil per plant under field conditions. This highlights the limitations when going from the lab to the field scale. The low phytoremediation efficiency in the field and the mobilisation of high amounts of Pb and Cd down the soil profile may make the use of EDTA and Z. mays not suitable for the remediation of severely heavy metal contaminated soils in a reasonable time frame and may result in substantial groundwater pollution under used crop management.  相似文献

Context

Species show different sensitivity to habitat loss and fragmentation depending on their specialization. Populations of a species at the range margin are generally assumed to be more stenoecious than populations at the core of the distribution and should therefore be more sensitive to habitat fragmentation.

Objectives

We evaluated the hypothesis that fragmentation effects species more strongly at the range periphery of their range compared to the core, resulting in lower genetic variability in comparable patch sizes and lower gene flow among populations.

Methods

We compared the genetic diversity and structure of five sand lizard (Lacerta agilis) populations at the margin of its range in Bulgaria and of 11 populations at the core of its distribution in Germany. We based the analysis on microsatellites, comprising 15 loci in Bulgaria and 12 in Germany.

Results

All diversity indices declined with patch size. For medium-sized patches all diversity indices were lower at the range periphery compared to the core, with two of them being significant. AICc based model selection showed strong support for core/periphery and patch size effects for observed and expected heterozygosity but only a patch size effect for allelic richness. There was no isolation-by-distance and each sampled population was allocated to a separate cluster with high probability for both countries, indicating that all populations are (almost) completely isolated.

Conclusion

Our study indicates an increased sensitivity of a species to fragmentation at the periphery compared to the core of its distribution. This differential sensitivity should be accounted for when prioritizing species based on their fragmentation sensitivity in landscape management.

The peculiarities of the survival and adaptation of deep-sea organisms raise interest in the study of their metabolites as promising drugs. In this work, the hemolytic, cytotoxic, antimicrobial, and enzyme-inhibitory activities of tentacle extracts from five species of sea anemones (Cnidaria, orders Actiniaria and Corallimorpharia) collected near the Kuril and Commander Islands of the Far East of Russia were evaluated for the first time. The extracts of Liponema brevicorne and Actinostola callosa demonstrated maximal hemolytic activity, while high cytotoxic activity against murine splenocytes and Ehrlich carcinoma cells was found in the extract of Actinostola faeculenta. The extracts of Corallimorphus cf. pilatus demonstrated the greatest activity against Ehrlich carcinoma cells but were not toxic to mouse spleen cells. Sea anemones C. cf. pilatus and Stomphia coccinea are promising sources of antimicrobial and antifungal compounds, being active against Gram-positive bacteria Bacillus subtilis, Staphylococcus aureus, and yeast Candida albicans. Moreover, all sea anemones contain α-galactosidase inhibitors. Peptide mass fingerprinting of L. brevicorne and C. cf. pilatus extracts provided a wide range of peptides, predominantly with molecular masses of 4000–5900 Da, which may belong to a known or new structural class of toxins. The obtained data allow concluding that deep-sea anemones are a promising source of compounds for drug discovery.  相似文献

Herpes simplex virus type 1 (HSV-1) is one of the most prevalent pathogens worldwide requiring the search for new candidates for the creation of antiherpetic drugs. The ability of sea urchin spinochromes—echinochrome A (EchA) and its aminated analogues, echinamines A (EamA) and B (EamB)—to inhibit different stages of HSV-1 infection in Vero cells and to reduce the virus-induced production of reactive oxygen species (ROS) was studied. We found that spinochromes exhibited maximum antiviral activity when HSV-1 was pretreated with these compounds, which indicated the direct effect of spinochromes on HSV-1 particles. EamB and EamA both showed the highest virucidal activity by inhibiting the HSV-1 plaque formation, with a selectivity index (SI) of 80.6 and 50.3, respectively, and a reduction in HSV-1 attachment to cells (SI of 8.5 and 5.8, respectively). EamA and EamB considerably suppressed the early induction of ROS due to the virus infection. The ability of the tested compounds to directly bind to the surface glycoprotein, gD, of HSV-1 was established in silico. The dock score of EchA, EamA, and EamB was −4.75, −5.09, and −5.19 kcal/mol, respectively, which correlated with the SI of the virucidal action of these compounds and explained their ability to suppress the attachment and penetration of the virus into the cells.  相似文献

Gliadins, and primarily α-gliadins containing several sequences such as aa 31-49, aa 56-88 (33-mer), aa 57-68, and aa 69-82, are critical in the induction of immune response or toxic reaction leading to the development of celiac disease (CLD). The role of IgA anti-gliadin antibodies (IgA AGA) is unknown. To this end, we prepared several humanized monoclonal IgA AGA using transgenic α1KI mice. Employing Pepscan with overlapping decapeptides of α-gliadin we observed a robust similarity between the specificity of humanized mouse monoclonal IgA AGA and IgA AGA from patients with florid CLD. The common immunodominant region included several sequential epitopes localized in the N-terminal part of α-gliadin (QFQGQQQPFPPQQPYPQPQPFP, aa 29-50, and QPFPSQQPYLQL, aa 47-58). Notably, IgA AGA produced by clones 8D12, 15B9, 9D12, and 18E2 had significant reactivity against sequences localized in the 33-mer, LQLQPFPQPQ (aa 56-65) and PQLPYPQPQPFL (aa 69-80). Humanized mouse monoclonal IgA AGA that have a known specificity are suitable as standard in ELISAs to detect serum IgA AGA of CLD patients and for studying the AGA pathogenic role in CLD, especially for analyzing the translocation of complex of specific IgA antibodies and individual gliadin peptides through enterocyte barrier.  相似文献

Host-seeking Dermacentor reticulatus ticks were detected by flagging method at 46 localities at south-east part of the Czech Republic, in the basins of rivers Morava and Dyje. Exact north-west distribution limits of D. reticulatus were defined in this area for the first time. Detailed prediction map of probabilities of D. reticulatus occurrence was obtained using GIS analysis. Spatial model delimited a south-north gradient in probability across the studied area, with highest probabilities above 0.8 in its southernmost part. Abundance of D. reticulatus varied markedly between localities in interval 0.33-222 of ticks per flag per hour. The highest abundances were in flooded areas at lower streams, towards upper streams abundance and density of these ticks decreased. Females prevailed in samples with population sex ratio of 0.413, significantly deviating from parity. Larvae and nymphs of this species were not detected by flagging. Although D. reticulatus range expansion probably did not reach such a degree as reported in other countries, these ticks became very abundant in some parts of studied area. Since spreading of vector-borne diseases became a problem in Europe, the knowledge of their exact recent geographic ranges is important for future modelling of their shift predictability.  相似文献

Babesia canis canis is the most frequent causative agent of canine babesiosis in Central Europe, frequently causing severe disease. Recently, many new endemic foci of this disease have been reported from European countries. Growing incidence of canine babesiosis was recorded also in Slovakia during the last decade, from first cases in eastern Slovakia ten years ago to recent cases all over the south of the country. We have used nested PCR-RFLP method to study prevalence of B. c. canis in its natural tick vector Dermacentor reticulatus, collected at three geographically isolated lowland areas of southern Slovakia situated in the southeast, southwest, and west of Slovakia, respectively. The highest prevalence of B. c. canis was observed in D. reticulatus from eastern Slovakia (14.7%; n=327), whereas the prevalence in southwest was significantly lower (2.3%; n=1205). Notably, all 874 D. reticulatus ticks collected at Záhorská ní?ina lowland (W Slovakia) were B. c. canis-negative. Recorded differences in Babesia prevalence concurs well with the shift in incidence of clinical cases of canine babesiosis as observed by vet practitioners. Presented results revealed that eastern Slovakia represents an area of high risk of B. c. canis infection, whereas western areas of the country still remain Babesia canis-free.  相似文献

Maintaining germplasm genetic integrity is a key objective of long-term ex situ conservation. Periodic regeneration, performed on limited plots with small number of individuals, increases the risk of genetic drift and genetic diversity changes. In this study, six accessions of white flowered, dry seed pea varieties (Pisum sativum L. subsp. sativum var. sativum): Bohatyr, Klatovsky zeleny, Hanák, Moravsky hrotovicky krajovy, Raman and Viktoria-75 and four accessions of colour flowered, fodder pea (P. sativum subsp. sativum var. arvense (L.) Poiret: Arvika, Česky banán, Moravská krajová and Niké, representing Czechoslovak varieties and landraces, bred over the last 40–80 years, were analyzed using ten microsatellite locus specific markers. Each accession was represented by 20 individual seeds of two temporally different samples, spanning the period of 20 or 40 years. Together with intra-accession variation (except of cv. Hanák), evidence of genetic changes, e.g. differences in allele frequencies as well as genetic composition of sample, was detected in six out of ten accessions (Arvika, Bohatyr, Česky banán, Moravsky hrotovicky krajovy, Moravská krajová and Raman). Evidence of genetic erosion was found in three accessions (Česky banán, Moravsky hrotovicky krajovy and Raman), while in another three (Arvika, Bohatyr and Moravská krajová) the level of diversity was found to have increased. Moreover in three samples of Bohatyr (2004) and Klatovsky zeleny (1963 and 2004), low levels of heterozygosity was detected. These results demonstrate that in pea, a self-pollinating and highly homozygous plant, the danger of the loss of genetic integrity exists. These findings are significant for long-term ex situ germplasm management.  相似文献

The cestode fauna of the darkspotted numbfish, Narcine maculata (Shaw) (Torpediniformes: Narcinidae), from Malaysian Bomrneo was examined for the first time. This work resulted in the discovery of a new genus and two new species of Anteroporidae (Lecanicephalidea). Sesquipedalapex comicus gen. n., sp. n. was erected on the basis of the peculiarities of its scolex, in particular its possession of an extremely long apical modification of the scolex proper, which readily distinguishes it from the other genus in the family. The genus is also distinct in its possession of acetabula that are in the form of suckers, rather than bothridiate in form. This species was found to deeply embed its elongate apical structure for much of its length within the intestinal mucosa, provoking a papilliform expansion of the outer wall of the spiral intestine at the site of attachment. The second new species, Anteropora klosmamorphis sp. n., is readily distinguished from its congeners on the basis of testis number and bothridial shape. Both new species are hyperapolytic. The diagnosis of Anteroporidae is amended to accommodate both new taxa. This increases the total number of genera in the family to two, and the total number of species to five.  相似文献

Microbial natural products are important for the understanding of microbial interactions, chemical defense and communication, and have also served as an inspirational source for numerous pharmaceutical drugs. Tropical marine cyanobacteria have been highlighted as a great source of new natural products, however, few reports have appeared wherein a multi-omics approach has been used to study their natural products potential (i.e., reports are often focused on an individual natural product and its biosynthesis). This study focuses on describing the natural product genetic potential as well as the expressed natural product molecules in benthic tropical cyanobacteria. We collected from several sites around the world and sequenced the genomes of 24 tropical filamentous marine cyanobacteria. The informatics program antiSMASH was used to annotate the major classes of gene clusters. BiG-SCAPE phylum-wide analysis revealed the most promising strains for natural product discovery among these cyanobacteria. LCMS/MS-based metabolomics highlighted the most abundant molecules and molecular classes among 10 of these marine cyanobacterial samples. We observed that despite many genes encoding for peptidic natural products, peptides were not as abundant as lipids and lipopeptides in the chemical extracts. Our results highlight a number of highly interesting biosynthetic gene clusters for genome mining among these cyanobacterial samples.  相似文献
