Samples of six common plant species collected in the old mining areas near Prince George, British Columbia (Canada) and Mount Amiata, Tuscany (Italy) show remarkable similarities in the variation of plant/soil Hg concentration ratio with soil Hg content irrespective of species or other biological differences. In contrast, plants sampled in the geothermally active areas of New Zealand, Hawaii and around Mount St. Helens exhibit more individuality in the concentration ratio to soil Hg relationship, but the relationships are distinctly different from the mine site specimens. This distinction is particularly evident when the same species of Equisetum and Plantago taken from these two different areas are compared. These and other data support the hypothesis that specific local environmental factors strongly influence the accumulation of Hg in plants even when the immediate soil concentrations are the same. This study aimed to investigate both the pharmacokinetic behavior and tolerance of methotrexate (MTX) in horses to design a specific dosing regimen as a new immunomodulatory drug for long-term treatment. To determine the primary plasma pharmacokinetic variables after single intravenous, subcutaneous or oral administration, six horses were administered 0.3 mg/kg MTX in a crossover design study. After a 10-week washout, MTX was administered subcutaneously to three of the six previously treated horses at a dose of 0.3 mg/kg once per week for 3 months. In both studies, MTX and metabolite concentrations were measured using LC-MS/MS. The absolute bioavailability of MTX was 73% following subcutaneous administration but less than 1% following oral administration. The plasma clearance was 1.54 ml min−1 kg−1 (extraction ratio = 2%). After 24 hr, plasma concentrations were below the LOQ. No adverse effects were noted except for a moderate reversible elevation in liver enzymes (GLDH). With regards to the main metabolites of MTX, very low concentrations of 7-hydroxy-MTX were found, whereas polyglutamated forms (mainly short chains) were found in red blood cells. A subcutaneous dose of 0.2 mg kg−1 week−1 may be safe and relevant in horses, although this has yet to be clinically confirmed. 相似文献

Badgers are a popular and protected species in England, despite their association with tuberculosis (Mycobacterium bovis infection) in cattle. Casualty badgers are commonly presented to veterinarians and wildlife rescue centres following injury, as a result of disease, or as orphans. Strict policies are adopted for their rehabilitation and release, with respect to the prevention of spread of tuberculosis, these policies differ between adult badgers and badger cubs. Adult badger casualties are not normally tested for M. bovis infection prior to release, but are instead kept in isolation and released back where found. A study of casualty adult badgers found 10% to be positive on a single serological test. These animals had a variety of clinical signs that had resulted in none of them being released back to the wild. Badger cubs are serologically tested for evidence of M. bovis infection on three occasions during rearing; 13% were found to test positive. Positive animals were examined at post-mortem and cultures made for M. bovis; 12.5% of serologically positive animals were found to be culture positive. We analyzed the effect of simplifying assumptions in canopy representation of radiation transfer models, comparing modeled diffuse non-interceptance and photosynthetic photon flux density with measurements at different layers of complex pine-broadleaved canopy with large seasonal variation of leaf area index. The most detailed model included clumping of trees (i.e.,?stand density) and a vertical specification of leaf angle distribution and shoot clumping. A less detailed model replaced the vertically specified variables with their means. The most parsimonious model accounted for neither shoot clumping nor stand density. The vertical specification of shoot clumping and leaf angle distribution only slightly improved vertical and seasonal openness and light estimates over using mean values. Further simplification had little effect on total absorbed light but was more risky for estimates of the vertical distributions of openness and light absorbed by the canopy, which will affect photosynthesis estimates due to the non-linearity of photosynthetic light response. Including woody surfaces in winter, when leaf area was low, was essential for reproducing the measurements correctly. A sensitivity analysis showed that ignoring (i)?shoot clumping could result in a substantial overestimation of total absorbed light with errors increasing with decreasing leaf area and (ii) stand density in sparse stands could lead to substantial overestimation of total absorbed light, and the effect is largely independent of leaf area. Also, (iii) the effect of changing leaf angle distribution increased with decreasing leaf area, and was larger and more persistent along the leaf area range with increasing shoot clumping. Overall, accounting for the effect of tree clumping on absorbed light is most important in stands composed of species where leaves are not very clumped (e.g., broadleaved). However, even in forests with highly clumped shoots (i.e., coniferous), an accurate estimation of absorbed light distribution in stands requires incorporation of stand density in the model. 相似文献

Six blastomere morphology parameters indicative of cell development abnormalities, egg diameter, dry weight, total lipid, lipid classes and fatty acids were determined for egg batches collected daily from three Atlantic cod (Gadus morhua) broodstock groups over the course of one spawning season. Egg batches were incubated to hatch and each morphological and biochemical parameter was tested as a predictor of hatching success. Five of the six blastomere morphology parameters were significantly positively correlated with each other. Correlation coefficients among several fatty acid parameters were also significant but correlation coefficients among the various lipid classes were mostly not significant. No significant correlations were found between blastomere morphology and lipid class or fatty acid parameters. Egg dry weight was negatively correlated with cell clarity, %docosahexanoic acid (DHA), DHA:eicosapentaenoic acid, and Σ polyunsaturated fatty acids. Fertilization success was not significantly correlated with any of the morphology or biochemistry parameters. Within‐population variability in several morphological and fatty acid parameters was related to elapsed time since onset of first spawning. However, the occurrence of such relationships with elapsed time was highly variable and inconsistent among the three broodstocks, typically being significant for only one or two broodstocks but not all three. Mean hatching success rates were high (>75%) in all three broodstocks but hatching success was not significantly related to any of the morphological or biochemical parameters nor to elapsed time from onset of first spawning. The implications of these results are discussed in terms of the utility of the various morphology and biochemistry parameters as measures of egg quality in marine finfish hatcheries. 相似文献

- Today, aquatic biodiversity suffers from many pressures linked to human activities, including climate change, which particularly affects alpine areas. Many alpine freshwater species have shifted their geographical distribution to colder areas, but a reduced availability of suitable habitats is also forecasted. New artificial water bodies could provide habitat enhancement opportunities, including small mountain reservoirs built to overcome a lack of snow during winter.
- To investigate the role of reservoirs as a habitat for freshwater invertebrates, a case study was conducted on eight reservoirs in the Swiss Alps. The study aimed to compare the water quality and freshwater biodiversity of the reservoirs with those of 39 natural and newly excavated ponds. Data were collected on physico‐chemistry, freshwater habitat structure, and aquatic insects (dragonflies and aquatic beetles).
- The study showed that the mountain reservoirs investigated did not differ from natural ponds in terms of surface area, conductivity, and trophic level. Similarly to natural ponds, reservoirs showed signs of impairment owing to surface run‐off carrying pollutants linked to ski tourism. They presented a low diversity of mesohabitats, and in particular lacked vegetation. Compared with natural ponds, the species richness in reservoirs was lower for dragonflies but not for beetles. At the regional scale, the community from the reservoirs was a subset of the natural ponds community, supporting 38% of the regional species richness for these two insect groups.
- The results suggest that mountain reservoirs are likely to be important for biodiversity in alpine areas, both as habitats and as stepping stones for species shifting their geographical range. These water bodies can be enhanced further by some nature‐friendly measures to maximize benefits for biodiversity, including margin revegetation or the creation of adjacent ponds. Ecological engineering needs to be innovative and promote freshwater biodiversity in artificial reservoirs.

Enzootic dermatophytosis in a shelter with approximately 140 cats was treated according to a protocol combining identification, isolation and treatment of subclinical carrier and affected animals in accordance with a three‐area system: healthy animals (no lesions and negative cultures), subclinical carrier animals (no lesions but with positive cultures) and clinically affected animals (lesions and positive cultures). The cats were examined and inspected under a Wood’s lamp and had samples taken for fungal culture every 2 weeks. Thirty‐three per cent of the cats had a positive fungal culture at the start of the study. Clinically affected animals and carriers were treated with a 0.2% enilconazole lotion (Imaverol®) twice a week and given itraconazole (Itrafungol®) 5 mg/kg SID orally every other week. The environment was treated once a day with a 1% bleach solution and once a week with a 0.6% enilconazole (Clinafarm®) solution. Treated animals were considered cured after two consecutive negative fungal cultures. All cats were cured within 56 days. Prophylactic measures against dermatophytosis were implemented for new arrivals consisting of individual quarantine and the systematic taking of fungal cultures. No relapses were observed based on the fungal cultures taken from the animals and the environment over the first 10 months. 相似文献

In recent years, identification of the microbial sources responsible for soil N2O production has substantially advanced with the development of isotope enrichment techniques, selective inhibitors, mathematical models and the discoveries of specific N-cycling functional genes. However, little information is available to effectively quantify the N2O produced from different microbial pathways (e.g. nitrification and denitrification). Here, a 15N-tracing incubation experiment was conducted under controlled laboratory conditions (50, 70 and 85% water-filled pore space (WFPS) at 25 and 35 °C). Nitrification was the main contributor to N2O production. At 50, 70 and 85% WFPS, nitrification contributed 87, 80 and 53% of total N2O production, respectively, at 25 °C, and 86, 74 and 33% at 35 °C. The proportion of nitrified N as N2O (P N2O) increased with temperature and moisture, except for 85% WFPS, when P N2O was lower at 35 °C than at 25 °C. Ammonia-oxidizing archaea (AOA) were the dominant ammonia oxidizers, but both AOA and ammonia-oxidizing bacteria (AOB) were related to N2O emitted from nitrification. AOA and AOB abundance was significantly influenced by soil moisture, more so than temperature, and decreased with increasing moisture content. Rates of degradation of simazine and metribuzin were measured under standard conditions in soil samples taken from different areas of a field. First-order half-lives for metribuzin degradation gave a coefficient of variation of 21·1% whereas those for simazine gave a coefficient of variation of less than 7%. Half-lives in soil samples taken from the same plot at different times were also more variable for metribuzin than for simazine. In a field experiment with simazine, there was considerable variation in initial deposition when the herbicide was applied with a commercial boom sprayer, and variability in soil residues increased with time after application. When the herbicide was applied with a knapsack sprayer, initial deposition was more even and variability in soil residues was always less than when applied with the boom sprayer. It is concluded that the principal cause of the variability in soil residues is unevenness in initial application and that interpretation of soil residue measurements must always make due allowance for the large amount of variation which is often encountered. 相似文献
