Fatty acids are the natural ligands associated with the bovine milk lipocalin, β-lactoglobulin (BLG), and were identified by means of mass spectrometry. The naturally bound ligands were found to contribute to the stability of the proteins toward denaturation by both temperature and chaotropes. To assess the nature of the structural regions involved in this stabilization, the thermodynamic and kinetic aspects of the stability of various structural regions of the proteins were studied in the presence of bound palmitate, which is the most abundant natural ligand. Binding of a single palmitate molecule was found to affect not only the stability of the calyx region, where palmitate is bound, but also that of the region at the hydrophobic interface between the barrel itself and the long helix in the protein structure, where the thiol group of Cys121 is buried. This region is known to be essential for the stability of the BLG dimer and is relevant to the generation of "reactive monomers" that are involved in covalent and noncovalent polymerization of BLG and in the formation of covalent adducts with other milk proteins.  相似文献

In this article, we present equations derived for the prediction of the aboveground tree volume and phytomass for twenty-five of the most important forest species growing in Italy. These equations result from ongoing research aiming to fill a gap in the models available at the national scale. With regard to volume, the results are particularly important for thirteen species or groups of species that were once scaled with models, conventionally assumed as reference models, available for other species. In Italy, phytomass models had never been constructed at the national level before. For any single tree, specific equations allow estimations of the following tree components to be made: stem and large branches (for either volume or phytomass), small branches (phytomass), stump (phytomass) and the whole tree phytomass. The models have been constructed on the basis of nearly 1,300 sampling units (sample trees). Although these equations must be considered intermediate results of the ongoing research because only half the scheduled number of samples has been collected, they have already been used in the practice, for example in the estimates reported in the recently published second national forest inventory.  相似文献

In this study, the association between virulence genotypes and phylogenetic groups among Escherichia (E.) coli isolates obtained from pet dogs and cats with cystitis was detected, and fingerprinting methods were used to explore the relationship among strains. Forty uropathogenic E. coli (UPEC) isolated from dogs (n = 30) and cats (n = 10) in Italy were analysed by polymerase chain reaction (PCR) for the presence of virulence factors and their classification into phylogenetic groups. The same strains were characterized by repetitive extragenic palindromic (REP)- and enterobacterial repetitive intergenic consensus (ERIC)-PCR techniques. We found a high number of virulence factors such as fimbriae A, S fimbriae (sfa) and cytotoxic necrotizing factor 1 (cnf1) significantly associated with phylogenetic group B2. We demonstrated a high correlation between α-hemolysin A and pyelonephritis C, sfa, and cnf1 operons, confirming the presence of pathogenicity islands in these strains. In addition, UPEC belonging to group B2 harboured a greater number of virulence factors than strains from phylogenetic groups A, B1, and D. REP- and ERIC-PCR grouped the UPEC isolates into two major clusters, the former grouping E. coli strains belonging to phylogenetic group B2 and D, the latter grouping those belonging to groups A and B1. Given the significant genetic variability among the UPEC strains found in our study, it can be hypothesized that no specific genotype is responsible for cystitis in cats or dogs.  相似文献

ABSTRACT: In dogs Helicobacter spp. are found in all gastric regions usually localized in the surface mucus, gastric glands and parietal cells. The aim of this study was to detail the distribution of Helicobacter spp. in the fundic mucosa of asymptomatic Beagle dogs and their intracellular localization within parietal cells, in order to evaluate species-specific pathogenetic effects on gastric cells. The presence of Helicobacter spp. was investigated by immunohistochemistry, TEM, and PCR in the fundic mucosa of six Beagle dogs. Helicobacter spp. were found in all dogs examined, and H. bizzozeronii and H. felis were identified by PCR and confirmed by TEM. In the lumen of the fundic glands, co-localization was common. H. bizzozeronii was present in larger numbers than H. felis in both intraluminal and intraparietal localization. The amounts of H. bizzozeronii were similar in superficial and basal portions of the glands. H. felis was predominantly localized in the superficial portions of gastric glands but almost absent from the base. Within parietal cells, most Helicobacter organisms were intracanalicular, but intact and degenerate Helicobacter organisms were also visualized free in the cytoplasm or in secondary lysosomes. No specific degenerative lesions were found in infected parietal cells. Helicobacter organisms were also observed within macrophages in the lamina propria. In conclusion, there is a differential distribution of H. bizzozeronii and H. felis in the fundic mucosa of Beagle dogs, and their intracellular localization in parietal cells and macrophages suggests novel pathogenic scenarios for the development of immune response and maintenance of chronic gastritis in dogs.  相似文献

The effects of humic acids extracted from two commercially‐available products (CP‐A prepared from peat and CP‐B prepared from leonardite) on the growth and mineral nutrition of tomato plants (Lycopersicon esculentum L.) in hydroponics culture were tested at concentrations of 20 and 50 mg L‐1. Both the humic acids tested stimulated plants growth. The CP‐A stimulated only root growth, especially at 20 mg L‐1 [23% and 22% increase over the control, on fresh weight basis (f.w.b.), and dry weight basis (d.w.b.), respectively]. In contrast, CP‐B showed a positive effect on both shoots and roots, especially at 50 mg L‐1 (shoots: 8% and 9% increase over the control; roots: 18% and 16% increase over the control, on f.w.b. and d.w.b., respectively). Total ion uptake by the plants was affected by the two products. In particular, CP‐A showed an increase in the uptake of nitrogen (N), phosphorus (P), iron (Fe), and copper (Cu), whereas, CP‐B showed positive effects for N, P, and Fe uptake. The change in the Fe content was the most appreciable effect on mineral nutrition (CP‐A: 41% and 33% increase over the control for 20 mg L‐1 and 50 mg L‐1 respectively; CP‐B: 31% and 46% increase over the control for 20 mg L‐1 and 50 mg L‐1, respectively). Increases in Fe concentration in the plant roots were especially pronounced (CP‐A: 113% and 123% increases with respect to controls for the 20 mg L‐1 and 50 mg L‐1 treatments; CP‐B: 135% and 161% increases with respect to the control for 20 mg L‐1 and 50 mg L‐1 treatments). On the basis of the current experiments and from evidence in the literature, reduction of Fe3+ to Fe2+ by humic acid is considered as a possibility to explain a higher Fe availability for the plants.  相似文献

Cold-adapted enzymes produced by psychrophilic organisms have elevated catalytic activities at low temperatures compared to their mesophilic counterparts. This is largely due to amino acids changes in the protein sequence that often confer increased molecular flexibility in the cold. Comparison of structural changes between psychrophilic and mesophilic enzymes often reveal molecular cold adaptation. In the present study, we performed an in-silico comparative analysis of 104 hydrolytic enzymes belonging to the family of lipases from two evolutionary close marine ciliate species: The Antarctic psychrophilic Euplotes focardii and the mesophilic Euplotes crassus. By applying bioinformatics approaches, we compared amino acid composition and predicted secondary and tertiary structures of these lipases to extract relevant information relative to cold adaptation. Our results not only confirm the importance of several previous recognized amino acid substitutions for cold adaptation, as the preference for small amino acid, but also identify some new factors correlated with the secondary structure possibly responsible for enhanced enzyme activity at low temperatures. This study emphasizes the subtle sequence and structural modifications that may help to transform mesophilic into psychrophilic enzymes for industrial applications by protein engineering.  相似文献

Headspace sorptive extraction (HSSE) and stir bar sorptive extraction (SBSE), two recently introduced solventless enrichment techniques, have been applied to the analysis of the headspace of Arabica roasted coffee and of the headspace of the brew and of the brew itself. In both HSSE and SBSE enrichment is performed on a thick film of poly(dimethylsiloxane) (PDMS) coated onto a magnet incorporated in a glass jacket. Sampling is done by placing the PDMS stir bar in the headspace (gas phase extraction or HSSE) or by immersing it in the liquid (liquid phase extraction or SBSE). The stir bar is then thermally desorbed on-line with capillary GC-MS. The performance of HSSE and SBSE have been compared through the determination of the recoveries and relative abundances of 16 components of the coffee volatile fraction to classical static headspace (S-HS) and to headspace and in-sample solid phase microextraction (HS-SPME and IS-SPME, respectively) applying the fibers PDMS 100 microm, Carbowax/divinylbenzene 65 microm (CW/DVB), Carboxen/PDMS 75 microm(CAR/PDMS), polyacrylate 85 microm(PA), PDMS/divinylbenzene 65 microm(PDMS/DVB), and Carboxen/divinylbenzene/PDMS 50-30 microm(CAR/PDMS/DVB). In all cases, HSSE and SBSE gave higher recoveries, and this is entirely due to the high amount of PDMS applied.  相似文献

Patterns of Abies cephalonica Loudon seedling recruitment at its westernmost geographical limit (western Greece), were studied by monitoring their emergence, survival, and establishment for up to four years in the microhabitats to which seeds are dispersed. Seedlings were monitored on 200 permanent sampling plots established in 11 different locations covering the entire strictly protected Mount Aenos National Park (Cephalonia). Annual seedling mortality rate was estimated in relation to habitat characteristics as well as to their number of cotyledons. A. cephalonica presented the typical behaviour of a masting species in the number of seedlings produced over the four years of study. Of the habitat characteristics studied, ground cover, elevation, soil depth, slope and canopy openness were the main parameters that classified seedling habitat with respect to their emergence. Seedling survival was higher in years of low seedling production. Seedlings with higher cotyledon numbers exhibited higher survival. Our data indicate that A. cephalonica is characterized by remarkable reproductive plasticity, which is expressed in the number of seedlings produced each year and those surviving with respect to its habitat characteristics. Preserving the species implies preserving its habitat. In this sense, Mount Aenos National Park may be the most important protected area for the endemic fir in view of preserving its integrity through pre-emptive management, in the light of current and future threats.  相似文献

Antimicrobial peptides (AMPs) are found widespread in nature and possess antimicrobial and immunomodulatory activities. Due to their multifunctional properties, these peptides are a focus of growing body of interest and have been characterized in several fish species. Due to their similarities in amino-acid composition and amphipathic design, it has been suggested that neuropeptides may be directly involved in the innate immune response against pathogen intruders. In this review, we report the molecular characterization of the fish-specific AMP piscidin1, the production of an antibody raised against this peptide and the immunohistochemical identification of this peptide and enkephalins in the neuroepithelial cells (NECs) in the gill of several teleost fish species living in different habitats. In spite of the abundant literature on Piscidin1, the biological role of this peptide in fish visceral organs remains poorly explored, as well as the role of the neuropeptides in neuroimmune interaction in fish. The NECs, by their role as sensors of hypoxia changes in the external environments, in combination with their endocrine nature and secretion of immunomodulatory substances would influence various types of immune cells that contain piscidin, such as mast cells and eosinophils, both showing interaction with the nervous system. The discovery of piscidins in the gill and skin, their diversity and their role in the regulation of immune response will lead to better selection of these immunomodulatory molecules as drug targets to retain antimicrobial barrier function and for aquaculture therapy in the future.  相似文献

Wild blueberries are a rich source of polyphenols and other compounds that are highly metabolized by the intestinal microbiota and may, at the same time, affect the intestinal environment itself. A repeated-measure, crossover dietary intervention on human volunteers was designed to study the effect of six week consumption of a wild blueberry ( Vaccinium angustifolium ) drink, versus a placebo drink, in modulating the intestinal microbiota. Relative to total eubacteria, Bifidobacterium spp. significantly increased following blueberry treatment (P ≤ 0.05), while Lactobacillus acidophilus increased after both treatments (P ≤ 0.05). No significant differences were observed for Bacteroides spp., Prevotella spp., Enterococcus spp., and Clostridium coccoides . Bifidobacteria, which have been largely proposed to be of benefit for the host, appeared to be selectively favored suggesting an important role for the polyphenols and fiber present in wild blueberries. Results obtained suggest that regular consumption of a wild blueberry drink can positively modulate the composition of the intestinal microbiota.  相似文献
