Objective: Mechanical evaluation of 2 suture constructs for extracapsular stifle stabilization. Study Design: In vitro study. Sample Population: Crimped interlocking loop constructs (ILC) of 45 kg nylon leader line (NLL) and Orthofiber® (OF). Methods: ILC were tightened to 100 N, then crimp secured. Ramp to failure (n=10/group)—Data were derived from force/displacement plots. Stress–relaxation testing (n=10/group)—ILC's were nondestructively loaded and held at resultant displacement as force data were recorded. Incremental, cyclic loading (n=10/group)—ILC's were loaded (5 cycles/set) starting at 100 N and incrementally increased by 50 N (1 and 3 Hz protocols). Loop tension and elongation were recorded after each set. Results: Ramp to failure—initial loop tension was similar (NLL 75.5 ± 9.5 N; OF 68.7 ± 10.4 N, P=.140). Tested OF constructs were stiffer (NLL 125.7 ± 4.0; OF 234.6 ± 25.0 N/mm, P≤.001), had lower yield load (NLL 193.6 ± 13.8; OF 137.3 ± 94.3 N, P≤.001), lower peak load (NLL 873.7 ± 68.6; OF 653.6 ± 80.2 N, P≤.001), and lower elongation at failure (NLL 19.1 ± 1.4; OF 5.2 ± 1.0 mm, P≤.001) and at yield (NLL 1.52 ± 0.2; OF 0.3 ± 0.6 mm, P=.003) than NLL constructs. Yield in NLL ILC's was variable knot tightening/crimp slippage, but only crimp‐suture slippage in OF. Stress–relaxation testing—OF demonstrated greater relaxation. Incremental, cyclic loading—induced ILC elongation and tension loss in both groups, independent of loading frequency. NLL lost tension at lower rate, but elongated more than OF. Conclusions: NLL construct is mechanically superior to OF construct.  相似文献

Theileria parva antigens recognized by cytotoxic T lymphocytes (CTLs) are prime vaccine candidates against East Coast fever in cattle. A strategy for enhancing induction of parasite-specific T cell responses by increasing recruitment and activation of dendritic cells (DCs) at the immunization site by administration of bovine Flt3L and GM-CSF prior to inoculation with DNA vaccine constructs and MVA boost was evaluated. Analysis of immune responses showed induction of significant T. parva-specific proliferation, and IFN-γ-secreting CD4(+) and CD8(+) T cell responses in immunized cattle. However, antigen-specific CTLs were not detected. Following lethal challenge, 5/12 immunized cattle survived by day 21, whereas all the negative controls had to be euthanized due to severe disease, indicating a protective effect of the vaccine (p<0.05). The study demonstrated the potential of this technology to elicit significant MHC class II and class I restricted IFN-γ-secreting CD4(+) and CD8(+) T cells to defined vaccine candidate antigens in a natural host, but also underscores the need to improve strategies for eliciting protective CTL responses.  相似文献

Composting inside high-rise, caged layer facilities can produce atmospheric ammonia (NH3) concentrations exceeding standards for human and poultry health. Control measures that reduce NH3 volatilization are necessary for in-house composting to be sustainable. Due to differences specific to in-house composting — low carbon to nitrogen ratios of composting material, continuous manure addition, and frequent turning — it is not known whether NH3 control measures used previously for poultry manure will work. The objectives of this study were to evaluate various amendment and process controls on NH3 produced during simulated in-house composting in the lab, and to evaluate select chemical control measures during composting inside a high-rise layer facility. Ten amendments (aluminum sulfate; chloride salts of aluminum, calcium, magnesium, and potassium; gypsum; sodium bisulfate; zeolite (clinoptilolite); straw; and cellulose) and four process controls (moisture; temperature; turning frequency; and particle size) were evaluated in lab incubations in 1 L vessels wherein samples of poultry manure compost were incubated to simulate composting. Vials of boric acid solution were used to capture NH3 evolved during incubations. With the exception of zeolite and cellulose, all amendments reduced NH3 capture. Low moisture and temperature also reduced NH3 capture, although managing temperature and moisture to achieve low NHg would adversely impact microbial activity and other desired benefits of composting. When evaluated inhouse, aluminum sulfate, calcium chloride and magnesium chloride did not reduce NH evolution from compost measured on three different dates with a gas sensor. Spatial variability along treated segments of windrow apparently masked amendment effects. At the end of a six-week composting cycle, total nitrogen content was higher in compost treated with aluminum sulfate than control or chloride salt treatments. Aluminum sulfate and process controls such as moisture content, carbon source and particle size have potential to reduce NH3 loss from poultry manure composted inside high-rise layer structures. In-house compost management to reduce NH3 volatilization must consider the cost of amendments, effectiveness, and impacts on the composting process.  相似文献

In soybean, manual cross-pollination to produce large quantities of F1 hybrid seed for yield trials is difficult and time-consuming. Conversely, insect-mediated cross-pollination has been shown to produce large quantities of hybrid seed in soybean and could facilitate the identification of heterotic patterns. The objective of our study was: (1) evaluate F1 hybrid soybean plants from single crosses for yield and agronomic traits over several environments and (2) compare hybrid performance of the single crosses to lines developed from three-way crosses and backcrosses. In 2003, F1 seed of single-crosses and their parent lines were evaluated in replicated experiments at three locations. Also in 2003, three-way crosses, and BC1F1 seed were produced. In 2004, three-way crosses, BC1F1 crosses, and their parent lines were evaluated at one location. High-parent heterosis (HPH) in single-crosses for grain yield ranged from −41.11% to +11.19%; for protein content from −4.34% to +3.53%, and for oil content from −13.22% to −0.84%. In three-way crosses, HPH for yield ranged from −25.21% to −4.50%, for protein from −2.72% to +1.92%, and for oil from −5.87% to −1.20%. For BC1F1 crosses, HPH for yield ranged from −15.65% to +41.97%, for protein from −2.57% to +1.69%, and for oil from −2.47% to +2.22%. Although positive heterosis levels were observed across all populations tested to determine the economic feasibility it is imperative that more tests of more cross-combinations be evaluated in replicated environments. Extensive research in different environments must be conducted to determine what parental combinations will produce the highest heterosis levels, and to develop criteria for selecting the parents with the best combining ability. This will be important to maximize agronomic performance that can economically justify the use of hybrids in soybean production.  相似文献

A 3-year-old male pygmy goat was evaluated because of a grade III/V plateau pansystolic murmur that was auscultated over the tricuspid valve area and a grade II/V plateau pansystolic murmur that was auscultated over the left heart base. Echocardiography revealed a large right atrium and ventricle, an atrial septal defect, and a dysplastic tricuspid valve displaced apically into the right ventricle. Contrast echocardiography and color-flow Doppler echocardiography revealed flow from right to left through the atrial septal defect and severe tricuspid regurgitation. A diagnosis of tricuspid valve dysplasia and severe tricuspid regurgitation was made. This anomaly (Ebstein's anomaly) of the tricuspid valve should be considered in goats that have systolic, with or without diastolic, murmurs over the tricuspic valve. Two-dimensional echocardiography can confirm the diagnosis of the anomaly. The associated tricuspid regurgitation and interatrial right-to-left shunt flow can be detected and semiquantitated by use of contrast, continuous wave, and color-flow Doppler echocardiography.  相似文献

The antibiograms of 408 Salmonella species isolated from large animals were collected during a three year study from 1981 through 1983. The predominant Salmonella serogroup among these isolates was group B. A consistently high percentage of all isolates were resistant to ampicillin and tetracycline. A pattern of increasing resistance to chloramphenicol and gentamicin was documented for serogroup B isolates while the susceptibility of the isolates to neomycin increased. There was a decrease in the incidence of susceptibility to sulfamethoxazole-trimethoprim among the group E isolates. These changes were not as remarkable, nor as alarming, as the overall decreased susceptibility to chloramphenicol and gentamicin. An evaluation of the principles concerning use of antimicrobial agents in veterinary medicine for treatment of Salmonella infections is recommended.  相似文献
