In human medicine, recombinant human thyroid-stimulating hormone (rhTSH) increases thyroid radioactive iodine uptake (RAIU), allowing radioiodine-131 (131I) dose reduction and greater efficacy in the treatment of differentiated thyroid cancer and multinodular goiter. The goal of this study was to evaluate the effect of rhTSH, administered 24 h and 48 h before radioiodine-123 (123I), on the thyroid RAIU in healthy dogs. Seven healthy euthyroid beagles were randomly allocated to 3 groups (2 groups of 2 dogs and 1 group of 3 dogs) in a prospective, blinded, crossover study. At Week 1, 1 group received 123I for a baseline RAIU; 1 group received 100 μg of rhTSH IV 24 h before 123I, and 1 group received 100 μg of rhTSH IV 48 h before 123I. All dogs received 37 MBq of radioactive 123I IV, and thyroid RAIU was determined 8 h, 24 h, and 48 h thereafter. The study was designed in such a manner that each dog received the 3 treatments and a wash-out period of 3 wk was respected in between. Blood samples were taken for measurement of serum total thyroxine (TT4) and thyrotropin (TSH) concentrations at baseline and 6 h, 12 h, 24 h, and 48 h after rhTSH administration. Recombinant human TSH caused no significant change on thyroid RAIU. The overall mean thyroid RAIU significantly decreased during the study independent of the treatment. Recombinant human TSH significantly increased serum TT4 concentration, which peaked 6 h after rhTSH administration. Compared to baseline, serum TSH concentration remained higher at 6 h, 12 h, 24 h, and 48 h. However, a statistically significant difference was reached only at 6 h and 12 h after rhTSH administration. No adverse effects of rhTSH were observed during the study. Further studies are needed to determine the best timing and dosage of administration of rhTSH in healthy and thyroid carcinoma dogs. 相似文献

This study aimed to analyse the effects of different water temperatures on the growth, survival and blood physiological parameters of Lophiosilurus alexandri. Juveniles measuring 12.6 ± 0.5 cm and weighing 30.6 ± 3.6 g were subjected to four water temperature regimes: 23, 26, 29 and 32°C with four replicates. The animals were fed twice daily with formulated diet. The experiment lasted for 35 days. Survival was 100% at all water temperatures, and the best temperature for growth was estimated to be 27.8°C using a quadratic regression model. The estimated temperatures for the best feed conversion and highest feed consumption rates were 26.2 and 28.8°C respectively. The fat visceral‐somatic index was highest at 32°C. The highest haemoglobin and haematocrit values were 29.4 and 32.1°C respectively. For plasma protein and cholesterol, the lowest values estimated were 23.9 and 24.1°C respectively. Glucose and liver nitrogen content increased with rising temperature. Higher values of total ammonia in the water were observed at higher temperatures. No effects of temperature were seen on the amounts of muscle dry matter, nitrogen, energy content and triglycerides or on the hepatosomatic index. Water temperature has an important role in the developmental and blood physiological parameters of juvenile L. alexandri, and the quadratic regression model showed that the appropriate temperature for growth is between 27 and 28°C. 相似文献

The majority of oil from oceanic oil spills (e.g. the recent accident in the Gulf of Mexico) converges on coastal ecosystems such as mangroves. Microorganisms are directly involved in biogeochemical cycles as key drivers of the degradation of many carbon sources, including petroleum hydrocarbons. When properly understood and managed, microorganisms provide a wide range of ecosystem services, such as bioremediation, and are a promising alternative for the recovery of impacted environments. Previous studies have been conducted with emphasis on developing and selecting strategies for bioremediation of mangroves, mostly in vitro, with few field applications described in the literature. Many factors can affect the success of bioremediation of oil in mangroves, including the presence and activity of the oil-degrading microorganisms in the sediment, availability and concentration of oil and nutrients, salinity, temperature and oil toxicity. More studies are needed to provide efficient bioremediation strategies to be applicable in large areas of mangroves impacted with oil. A major challenge to mangrove bioremediation is defining pollution levels and measuring recuperation of a mangrove. Typically, chemical parameters of pollution levels, such as polycyclic aromatic hydrocarbons (PAHs), are used but are extremely variable in field measurements. Therefore, meaningful mangrove monitoring strategies must be developed. This review will present the state of the art of bioremediation in oil-contaminated mangroves, new data about the use of different mangrove microcosms with and without tide simulation, the main factors that influence the success of bioremediation in mangroves and new prospects for the use of molecular tools to monitor the bioremediation process. We believe that in some environments, such as mangroves, bioremediation may be the most appropriate approach for cleanup. Because of the peculiarities and heterogeneity of these environments, which hinder the use of other physical and chemical analyses, we suggest that measuring plant recuperation should be considered with reduction in polycyclic aromatic hydrocarbons (PAHs). This is a crucial discussion because these key marine environments are threatened with worldwide disappearance. In Brazil, sour rot is an important postharvest disease on fruits and vegetables. Geotrichum candidum (synonym Galactomyces candidus) has been reported as the main species causing this disease. However, the identity of the causal agent is still uncertain. This research aimed to determine the identity of 165 fungal isolates associated with sour rot obtained from fruits and vegetables in Brazil, and to evaluate the effect of different temperatures on the incidence of sour rot on artificially inoculated tomato fruits. Based on the phylogenetic analyses of DNA sequences from the D1/D2 domain of the large subunit (LSU) rRNA gene and morphological analyses, 129 samples belonged to Galactomyces candidus, 15 to G. candidum var. citri-aurantii, 6 to G. phurueaense, 2 to Gal. pseudocandidus, 1 to Hyphopichia burtonii, 1 to H. khmerensis, 3 to Saccharomycopsis crataegensis, 1 to S. vini, 1 to Magnusiomyces tetrasperma, 1 to Trichosporon coremiiforme, and 1 to Zygoascus meyerae. Two new species were found, namely, Geotrichum solani (on potato) and Geotrichum spondiadis (on red mombin). All isolates were pathogenic when inoculated on healthy tomato fruits, including the new species of Geotrichum, which were also inoculated into their respective hosts, that is, potatoes and red mombins. To the best of our knowledge, this study is the first to report the presence of five other genera besides Geotrichum associated with sour rot on fruits and vegetables in Brazil, which demonstrates the diversity of fungi and yeasts associated with this disease. 相似文献

Meperidine has been shown to decrease lower esophageal sphincter tone in monkeys and people, to have little effect in cats, and to physically increase it in dogs. We hypothesized that administration of meperidine to dogs before anesthesia would decrease the probability of GER during the subsequent anesthetic. In this randomized, prospective clinical trial we aimed to determine the incidence of GER in dogs undergoing elective orthopedic surgery and receiving either meperidine or morphine prior to anesthesia. Dogs were admitted to the study, if they were healthy, with no history of vomiting or dysphagia. Dogs were fasted overnight. Dogs were received either morphine (0.66 mg kg–1 IM) or meperidine (8.8 mg kg–1 IM) with acepromazine. Anesthesia in all dogs included thiopental and isoflurane in oxygen. To measure esophageal pH a sensor-tipped catheter was placed with the tip 5–7 cm cranial to the lower esophageal sphincter, and connected to a computer for continual data collection. Dogs were observed for vomiting after pre-medication, and the pH of any fluid running from the mouth or nose during anesthesia was measured. Gastro-esophageal reflux was defined as a decrease in esophageal pH below 4 or an increase above 7.5 for greater than 15 seconds. One-way anova was used to test significance of differences between groups in parametric variables. Fisher's Exact test was used to test significance of differences in incidence between groups. In dogs receiving meperidine the incidence of vomiting was 0, and of GER was 31% (4/13), compared to 60% (6/10) and 60% (6/10), respectively in dogs receiving morphine. In this preliminary study, the administration of pre-anesthetic meperidine was associated with a 29% reduction in the absolute risk of GER compared to morphine. 相似文献

This study aimed to evaluate the effect of seminal plasma on bovine sperm cryopreservation and to assess the integrity of plasma and acrosomal membranes, mitochondrial potential, remodelling of F-actin cytoskeleton and sperm chromatin fragmentation during the cooling, equilibrium and freezing/thawing stages. Six ejaculates collected from seven Nelore bulls (n = 42) were used in this study. Each ejaculate was divided into two aliquots (with seminal plasma = SP group; without seminal plasma = NSP group) and packed to a final concentration of 50 × 106 sperm per straw. Statistical analyses were performed using SAS software (version 9.3), and p ≤ .05 was considered significant. A time effect was observed for all sperm characteristics (p < .05), except for chromatin fragmentation (p > .05). The presence of seminal plasma better preserved the acrosomal integrity (SP = 75.2% and NSP = 71.7%; p < .05) and also provided lower F-actin remodelling during cryopreservation process (SP = 29.9% and NSP = 32.4%; p < .05). Regarding to the cryopreservation stages, it was observed that cooling step induced higher remodelling of F-actin than the equilibrium and freezing/thawing stages (56.3%, 32.2% and 23.9%, respectively; p < .05). The equilibrium step had minor influence on overall sperm characteristics while the freezing/thawing stage was responsible for the highest percentage of damage in plasma membrane (−65.2%), acrosomal membrane (−34.0%) and mitochondrial potential (−48.1%). On the other hand, none of the cryopreservation stages affected chromatin integrity. It was concluded that the presence of seminal plasma provides increased acrosomal integrity and reduced remodelling of F-actin cytoskeleton. Higher F-actin remodelling is observed after the cooling step while the freezing/thawing step is most damaging to sperm membranes and mitochondrial potential during bovine sperm cryopreservation. 相似文献
