Quantitative autoradiography of [3H]muscimol binding in rat brain   总被引:3,自引:0,他引:3  
A simple quantitative autoradiographic technique for the study of neurotransmitter receptors that includes the use of a tritium-sensitive film permits saturation, kinetic, and competition studies of brain samples as small as 0.01 cubic millimeter. This technique was used to study [3H]muscimol binding in rat brain. Unilateral gamma-aminobutyric acid receptor supersensitivity was observed in the substantia nigra pars reticulata after production of localized lesions of the ipsilateral corpus striatum.  相似文献

The brain is generally considered immunoprivileged, although increasing examples of immunological responses to brain antigens, neuronal expression of major histocompatibility class I genes, and neurological autoimmunity have been recognized. An adeno-associated virus (AAV) vaccine generated autoantibodies that targeted a specific brain protein, the NR1 subunit of the N-methyl-D-aspartate (NMDA) receptor. After peroral administration of the AAV vaccine, transgene expression persisted for at least 5 months and was associated with a robust humoral response in the absence of a significant cell-mediated response. This single-dose vaccine was associated with strong anti-epileptic and neuroprotective activity in rats for both a kainate-induced seizure model and also a middle cerebral artery occlusion stroke model at 1 to 5 months following vaccination. Thus, a vaccination strategy targeting brain proteins is feasible and may have therapeutic potential for neurological disorders.  相似文献

Observations of the 1.4- to 2.4-micrometer spectrum of Pluto reveal absorptions of carbon monoxide and nitrogen ices and confirm the presence of solid methane. Frozen nitrogen is more abundant than the other two ices by a factor of about 50; gaseous nitrogen must therefore be the major atmospheric constituent. The absence of carbon dioxide absorptions is one of several differences between the spectra of Pluto and Triton in this region. Both worlds carry information about the composition of the solar nebula and the processes by which icy planetesimals formed.  相似文献

Wild blueberry (Vaccinium angustifolium Ait.) fields in the north east Canada are naturally grown in a course textured thin layer of soil and below this layer is a soilless layer of gravel. The root zone depth of this crop varies from 10 to 15 cm. Investigating the depth to the gravel layer below the course textured soil is advantageous, as it affects the water holding capacity of the root zone. Water and nutrient management are the two primary determinants of crop yield and the amount of leaching. The objective of this study was to estimate the depth to the gravel layer using DualEM-2 instrument. A C++ program written in Visual Studio 2010 was used to develop mathematical models for estimating the depth to the gravel layer from the outputs of DualEM-2 sensor. Two wild blueberry fields were selected in central Nova Scotia, Canada to evaluate the performance of DualEM-2 instrument in estimating the rootzone depth above the gravel layer. The mid points of squares created by grid lines were used as the sampling points at each experimental site. The actual depth to the interface was measured manually at selected grid points (n = 50). The apparent ground conductivity (ECa) values of DualEM-2 were recorded and the depth to the interface was estimated for the same sampling points within the selected fields. The fruit yield samples were also collected from the same grid points to identify the impact of the depth to the gravel layer on crop yield. After calibrations, comprehensive surveys were conducted and the actual and estimated depths to the interface were established. The interpolated maps of fruit yield, and the actual (zin) and estimated (\( {\text{z}}_{\text{in}}^{*} \)) depths to the interface were created in ArcGIS 10 software. Results indicated that the zin was significantly correlated with \( {\text{z}}_{\text{in}}^{*} \) for the North River (R 2 = 0.73; RMSE = 0.27 m) and the Carmel (R 2 = 0.45; RMSE = 0.20 m) sites. Results revealed that the areas with shallow depth to the gravel layer were low yielding, indicating that the variation in the depth to the gravel layer can have an impact on crop productivity. Non-destructive estimations of the depth to the gravel layer can be used to develop erosion control strategies, which will result in an increased crop production.  相似文献

The altitude profiles of temperature and pressure measured during the descent of the four Pioneer Venus probes show small contrast below the clouds but significant differences within the clouds at altitudes from 45 to 61 kilometers. At 60 kilometers, the probe which entered at 59.3 degrees north latitude sensed temperatures 25 K below those of the lower latitude probes, and a sizable difference persisted down to and slightly below the cloud base. It also sensed pressure below those of the other probes by as much as 49 millibars at a mean pressure of 200 millibars. The measured pressure differences are consistent with cyclostrophic balance of zonal winds ranging from 130 +/- 20 meters per second at 60 kilometers to 60 +/- 17 meters per second at 40 kilometers, with evidence in addition of a nonaxisymmetric component of the winds. The clouds were found to be 10 to 20 K warmer than the extended profiles of the lower atmosphere, and the middle cloud is convectively unstable. Both phenomena are attributed to the absorption of thermal radiation from below. Above the clouds, in the lower stratosphere, the lapse rate decreases abruptly to 3.5 K per kilometer, and a superimposed wave is evident. At 100 kilometers, the temperature is minimum, with a mean value of about 170 K.  相似文献

Although lightning has been seen on other planets, including Jupiter, polar lightning has been known only on Earth. Optical observations from the New Horizons spacecraft have identified lightning at high latitudes above Jupiter up to 80 degrees N and 74 degrees S. Lightning rates and optical powers were similar at each pole, and the mean optical flux is comparable to that at nonpolar latitudes, which is consistent with the notion that internal heat is the main driver of convection. Both near-infrared and ground-based 5-micrometer thermal imagery reveal that cloud cover has thinned substantially since the 2000 Cassini flyby, particularly in the turbulent wake of the Great Red Spot and in the southern half of the equatorial region, demonstrating that vertical dynamical processes are time-varying on seasonal scales at mid- and low latitudes on Jupiter.  相似文献

Changes in the distribution of dry matter and nonstructural carbohydrates in various parts of young ‘Fuyu’ persimmon (Diospyros kaki) were examined with 3- and 4-year-old trees with (fruited) and without fruits (defruited). The effect of such changes was then monitored with regard to the magnitude of new growth the following year. From June 15 to November 1, fruiting significantly decreased the rate of dry weight accumulation in perennial parts of the tree. Dry matter was partitioned the most to fruits (68–72%) and the least to the roots. Of the total dry weight, root accounted for 8–27% in fruited and 58–62% in defruited trees. During this period, soluble sugars were three times more in fruited than in defruited trees, but more than 95% of sugars were in the fruits. Starch content increased in defruited trees, more than 93% of which being distributed to the perennial parts. Compared with fruited trees, defruiting in the previous year increased new shoot growth and the number of fruits the following year, with a 20–58% greater increase in dry matter. During the new growth from April 10 to June 10 the following year, root dry weight decreased by 30–32% in defruited trees, whereas it increased by 20–80% in the fruited ones. Soluble sugars in roots decreased by 8.8–19.7 g in defruited trees but increased by 9.7–12.3 g in fruited ones. Starch in roots decreased by 68–75.1 g in defruited trees but increased by 10.2–13.3 g in fruited ones. However, there were no significant differences in soluble sugars and starch in the newly grown parts. It was estimated that a 1-g difference in dry matter accumulated in the previous season resulted in a 116-mg and 256-mg difference in dry weight of newly grown parts, and a 6.5-mm and 17.5-mm new shoot in the following season for 3- and 4-year-old trees, respectively.  相似文献
