AIMS: To evaluate a multivalent leptospiral and clostridial vaccine for prevention of renal colonisation and urinary shedding in sheep, following experimental challenge with New Zealand strains of Leptospira borgpetersenii serovar Hardjo type Hardjobovis and L. interrogans serovar Pomona.
METHODS: Two separate but similarly designed studies were conducted. In both studies, Romney-cross lambs, aged 9–11 weeks, were randomly allocated to a vaccinated group and a control group. Vaccinated lambs each received two 1.5-mL S/C doses of a multivalent leptospiral and clostridial vaccine, 4 weeks apart, and animals in the control groups received the same dose of saline. Groups of 12 vaccinated and 12 control lambs were randomly selected in each study for challenge with serovars Hardjo or Pomona. Challenge was initiated 16 weeks following the second vaccination with three daily doses of live leptospires by intranasal and conjunctival routes. Following challenge, urine samples were collected weekly for 6 weeks, for dark field microscopy and leptospiral culture; 6 weeks after challenge the lambs were slaughtered and kidneys collected for leptospiral culture.
RESULTS: In lambs challenged with serovar Hardjo, 8/12 unvaccinated lambs had ≥1 urine or kidney sample that was positive for leptospires following culture, compared with 0/12 lambs in the vaccinated group (p=0.001). In lambs challenged with serovar Pomona, 9/12 unvaccinated lambs had ≥1 urine or kidney sample that was positive following culture, compared with 0/12 lambs in the vaccinated group (p<0.001). Prevention of renal colonisation and urinary shedding, expressed as the prevented fraction, was 100 (95% CI=61.7–100)% and 100 (95% CI=68.3–100)% against challenge with serovars Hardjo and Pomona, respectively, at 4 months after vaccination.
CONCLUSIONS AND CLINICAL RELEVANCE: Use of a multivalent leptospiral and clostridial vaccine demonstrated protection against challenge from New Zealand strains of serovars of Hardjo and Pomona 4 months after vaccination in lambs first vaccinated at 9–11 weeks of age. Further studies are required to assess the duration of immunity against challenge in sheep. 相似文献
Splitting of the sun's global oscillation frequencies by large-scale flows can be used to investigate how rotation varies with radius and latitude within the solar interior. The nearly uninterrupted observations by the Global Oscillation Network Group (GONG) yield oscillation power spectra with high duty cycles and high signal-to-noise ratios. Frequency splittings derived from GONG observations confirm that the variation of rotation rate with latitude seen at the surface carries through much of the convection zone, at the base of which is an adjustment layer leading to latitudinally independent rotation at greater depths. A distinctive shear layer just below the surface is discernible at low to mid-latitudes. 相似文献
Over the past decade, lambing percentages have risen in conjunction with a rise in the percentage of multiple lambs born. Multiple-born lambs are smaller than their singleton counterparts and are particularly susceptible to starvation-exposure. Any technique that can increase the birthweight or thermoregulatory capability, or both, of otherwise lightweight lambs has the potential to substantially increase survival of multiple-born lambs. In the United Kingdom under housed conditions, shearing during pregnancy has been shown to increase both the birthweight and thermoregulatory capability of newborn lambs. However, shearing during pregnancy under pastoral conditions has failed to consistently affect the newborn lamb’s thermoregulatory capability. In contrast, under New Zealand’s pastoral conditions shearing during pregnancy has been found to increase birthweight, but results have been inconsistent in both magnitude and birth-rank specificity. Increase in feed intake by the dam, types of shearing comb used and changes in gestation length do not explain the variation observed. When studies involving shearing during pregnancy are collated it becomes apparent that there are two criteria that must be met to achieve a birthweight response. Firstly, the dam must have the potential to respond and secondly, the dam must have the means to respond. Any increase in lamb survival through shearing during pregnancy would be predominantly through an increase in birthweight of otherwise lightweight lambs. However, in the few studies conducted to date, an increase in birthweight has not resulted in a statistically significant increase in survival. For an increase in birthweight to have a positive effect on lamb survival, lambs must be otherwise destined to be born within a birthweight range in which survival rate is below optimum (<4.0 kg), and the increase in birthweight observed must move a significant proportion of otherwise lightweight lambs into a higher range of survival rate. 相似文献
Tropical forest fragmentation threatens biodiversity, yet basic information on population responses for major groups such as plants is lacking. Hypervariable genetic markers were used to reconstruct a population-level pedigree in fragmented tropical forest for the tree Symphonia globulifera. Though seedlings occurred only in remnant forest, the pedigree showed that most seedlings had been produced by sequentially fewer adults in pasture, creating a genetic bottleneck. The pedigree also implicated shifts in the foraging of animals that disperse pollen and seed in a secondary constriction of the bottleneck. These results suggest that tropical conservation strategies should anticipate complex, cryptic responses to fragmentation. 相似文献
We combined two protein-marking enzyme-linked immunosorbent assays (ELISA) with a predator gut content ELISA to monitor the movement and feeding activity of commercially-purchased Hippodamia convergens Guèrin-Mèneville (Coleoptera: Coccinellidae) under realistic field conditions during two field seasons in central Arizona. The protein-marking ELISAs were used to differentiate released H. convergens from the native beetles. Commercially purchased beetles marked with rabbit immunoglobulin G (IgG) were released into cotton fields and chicken IgG marked beetles were released into adjacent cantaloupe fields. Results showed that the total native beetle abundance in each crop was about the same size. The recovery rates after 15 days for the released beetles were less than 1.0% over all the releases, indicating that they dispersed readily from the release site. Of the recaptured beetles containing rabbit IgG (cotton), 82.2% were recovered in cotton and 11.8% moved to cantaloupe. Of those containing chicken IgG (cantaloupe), 66.5% were recovered in cantaloupe and 33.5% moved to cotton. A predator gut content ELISA was used to determine if there were differences in the frequency of predation of released versus indigenous H. convergens on the silverleaf whitefly, Bemisia argentifolii Bellows &; Perring (Homoptera: Aleyrodidae). The proportion of beetles containing whitefly antigens was always higher for the released beetles than for their native counterparts. Our results demonstrate an approach to combine protein marking and predator gut content ELISAs that allows the simultaneous comparison of feeding and intercrop movement of native and commercially-obtained biological control agents. 相似文献
AIMS: To determine the effects of liveweight and condition score of ewes at mating, and shearing mid-pregnancy, on birthweights and growth rates of twin lambs from birth to the time of weaning, and; to determine whether any effect of condition score is independent of liveweight. METHODS: Romney ewes (n=435) which conceived within a 17-day mating period and successfully gave birth to and reared twin lambs to weaning were used. Ewes were managed during pregnancy so that their total liveweight increased at a level equivalent to the expected growth of the conceptuses. Approximately half of the ewes were shorn 70 days after the mid-point of mating, and the remainder were left unshorn, at which time they had 6-months' wool growth. All lambs were weighed within 12 h of birth and again at weaning. RESULTS: There was a significant linear relationship (R2=0.99) between mean group liveweight and condition score of ewes at mating (difference of 7.3 kg per 1.0 unit condition score). Liveweight of ewes at mating had small but significant (p<0.05) positive effects on both birthweights and growth rates of twin lambs to weaning. Ewes that had condition scores of 3.5–4.0 (scale 0–5) at mating gave birth to significantly (p<0.05) lighter lambs than ewes that had condition scores <3.5. The condition score of ewes at mating was positively associated with growth rates of lambs, although this effect was largely explained by variation in the liveweight of ewes at mating. Twin lambs born to ewes shorn mid-pregnancy were significantly (p<0.05) heavier at birth (by 0.4 kg) and at weaning (p<0.05) than those born to unshorn ewes. The heavier weaning weights of lambs born to ewes shorn mid-pregnancy were due to a heavier birthweights rather than increased growth rate. CONCLUSIONS: Liveweight and condition score of ewes at mating had small positive effects on growth rates to weaning of twin lambs. Shearing mid-pregnancy increased birthweights and weaning weights of twin lambs. To achieve high weaning weights of twin lambs, farmers should ensure ewes attain high liveweights and condition scores at mating and shear mid-pregnancy. It is speculated that the effects of liveweight and condition score of ewes at mating on weaning weights of twin lambs would be greater than those observed in the present study under conditions in which nutrition of the dam is below optimum. 相似文献
We have described the efforts of the Australian administration of the Territory of Papua and New Guinea (TPNG) to establish a veterinary service and viable animal industries. These efforts began with planning before the end of World War II in 1945 and continued until independence in 1975. Whereas pre-war cattle had mostly been used to control grass on plantations, post-war, the objective was to use the country's extensive, unoccupied grasslands for cattle production. During this period, the cattle population increased from 4000 to more than 150,000. The greatest success was achieved in herds with crosses of Bos indicus and Bos taurus owned by expatriates. The only serious disease constraint on production was myiasis due to Chrysomya bezziana, a parasite throughout New Guinea and South-east Asia. Attempts were made to increase the productivity of the indigenous pig population. Success was limited by a failure to manage inadequate nutrition and internal parasites and to fully understand the cultural aspects of pig ownership. Similar problems inhibited chicken production in villages. The serious viral infections of pigs and birds were absent from TPNG, but a border with Indonesian Papua represents a potential route for the incursion of animal diseases exotic to both Papua New Guinea and to Australia. 相似文献
