To preserve Jeju black cattle (JBC; endangered native Korean cattle), a pair of
cattle, namely a post-death cloned JBC bull and cow, were produced by somatic cell
nuclear transfer (SCNT) in a previous study. In the present study, we examined the
in vitro fertilization and reproductive potentials of these
post-death cloned animals. Sperm motility, in vitro fertilization
and developmental capacity were examined in a post-death cloned bull (Heuk Oll Dolee)
and an extinct nuclear donor bull (BK94-13). We assessed reproductive ability in
another post-death cloned cow (Heuk Woo Sunee) using cloned sperm for artificial
insemination (AI). There were no differences in sperm motility or developmental
potential of in vitro fertilized embryos between the post-death
cloned bull and its extinct nuclear donor bull; however, the embryo development ratio
was slightly higher in the cloned sperm group than in the nuclear donor sperm group.
After one attempt at AI, the post-death cloned JBC cow became pregnant, and gestation
proceeded normally until day 287. From this post-death cloned sire and dam, a JBC
male calf (Heuk Woo Dolee) was delivered naturally (weight, 25 kg). The genetic
paternity/maternity of the cloned JBC bull and cow with regard to their offspring was
confirmed using International Society for Animal Genetics standard microsatellite
markers. Presently, Heuk Woo Dolee is 5 months of age and growing normally. In
addition, there were no significant differences in blood chemistry among the
post-death cloned JBC bull, the cow, their offspring and cattle bred by AI. This is
the first report showing that a pair of cattle, namely, a post-death cloned JBC bull
and cow, had normal fertility. Therefore, SCNT can be used effectively to increase

The aim of the present study was to investigate the effect of Eucommia ulmoides leaf (EUL) supplementation on the growth performance, blood and meat quality parameters in growing and finishing pigs. Ninety gilts (L × LW × D, 20 kg initialBW) were housed 10 per pen in a front‐open building with three replicate pens per treatment. Experimental treatment was started from the beginning of the growing stage (20 ± 3 kg) by supplementing EUL at 0(C), 3(T1) and 5% (T2) to the growing and finishing diet. Pigs were slaughtered by electrical stunning at 105 ± 3 kg live weight. Average daily feed intake (ADFI, kg/day) decreased (P < 0.05) by addition of EUL in growth performance, average daily gain (ADG, kg/day) was lower (P < 0.05) in T1 and T2 than in C. In hematology, leukocytes (WBC, 103/mm3) decreased (P < 0.05) in T1 and T2 than in C. Erythrocytes (RBC, 106/mm3), hemoglobin (HGB, g/dL) and hematocrit (HCT, %) increased (P < 0.05) in T1 and T2 than in C. Platelet (PLT, 103/mm3) was lower (P < 0.05) in T2 than in C and T1. In biochemical composition of serum, total protein (g/dL), r‐GTP (μ/L), total cholesterol (mg/dL) and triglycerides (mg/dL) were lower (P < 0.05) in T1 and T2 than in C. On longissimus dorsi muscle, crude protein was higher (P < 0.05) in T1 than in C. Crude ash was higher (P < 0.05) in T1 and T2 than in C. Yellow to blue color scale (CIE b*) in meat color was higher (P < 0.05) in T2 than in C. CIE b* in back fat color was higher (P < 0.05) in T2 than in the other treatments. In sensory evaluation scores for fresh meat, the values of meat color, fat color, drip loss and marbling were not significantly affected by addition of EUL. In cooked meat, the values of chewiness and overall acceptability were higher (P < 0.05) in T1 and T2 than in C. The results indicate that the addition of EUL affected growth performance, blood parameters and meat quality parameters in growing and finishing pigs. 相似文献

Rodent models of focal cerebral ischemia that do not require craniotomy have been
developed by intraluminal suture middle cerebral artery occlusion (MCAo). Mouse MCAo
models have been widely used and extended to genetic studies of cell death or recovery
mechanisms. Therefore, we compared surgery-related parameters and techniques between such
rats and mice. In rodent MCAo models, has to be considered body temperature during the
operative period, as well as the need for the use of a standardized tip in terms of the
outer diameter of probes. Induction of focal cerebral ischemia was measured by
neurological dysfunction parameters. Our methods could induce stable moderate-severity
ischemic brain injury models and histological alteration at 24 hr after MCAo surgery.
Moreover approximately 80% (rats) and 85% (mice) survival ratios were shown indicating
with model engineering success. Finally, we described and compared major parameters
between rats and mice, including probe size, thread insert length, operation and occlusion

This study aimed to investigate the effect of mydriasis using topical rocuronium bromide on electroretinography (ERG) in domestic pigeons (Columba livia). Scotopic mixed rod and cone, photopic cone, and photopic flicker ERG were performed on nine eyes of nine healthy adult pigeons under sedation. Each pigeon underwent two sets of ERG recordings. First, without the induction of mydriasis (control) and the second time with the induction of mydriasis using topical rocuronium bromide (treatment). The results were compared using either the Student’s t-test or Wilcoxon rank-sum test, where a P-value of <0.05 was considered statistically significant. No significant differences were observed in the a- and b-wave implicit times and amplitudes during scotopic ERG between the two groups. The a- and b-wave amplitudes in the photopic cone were significantly higher in the treatment group (63.83 ± 32.33 and 191.75 ± 94.46 µV) compared to the control group (46.15 ± 27.60 and 116.76 ± 70.65 µV; P=0.045 and P=0.032, respectively). The photopic flicker amplitude was also significantly higher in the treatment group (76.23 ± 48.56 µV) than in the control group (42.18 ± 31.18 µV; P=0.044). No statistically significant differences were observed in the photopic cone and flicker implicit times between both groups. In conclusions, mydriasis induced by rocuronium bromide in pigeon resulting in higher amplitudes during the photopic ERG but not scotopic ERG. 相似文献

This study was conducted to investigate the efficacy of rocuronium bromide as mydriatic agent in domestic pigeons (Columba livia). This study was done in two phases. In the first phase, rocuronium bromide (0.20 mg/20 µl) was topically instilled to the right eye (OD) of eight domestic pigeons. Pupil diameter was measured before instillation (T0), and at 5 (T05) and 10 (T10) min after instillation, and every 10 min thereafter until 160 (T160) min. Pupillary light reflex (PLR) was assessed using a scoring system at the same time points. In the second phase, the same dosage was instilled twice in the span of 10 min into both eyes (OU) of four pigeons (eight eyes). Measurements were done accordingly. The iris color in the first phase were: gravel, pearl and bull eye. All irises in the second phase were bull eye. Mydriasis were observed in 6/8 (75%) pigeons in the first phase. Maximal mydriasis was observed at T30 (mean pupil diameter=4.62 ± 0.13 mm). Pupil diameter in the treated eye was significantly different from contralateral eye and from T0 since T05 (P=0.017 and P=0.006, respectively)−T120 (P=0.043 and P=0.044, respectively). PLR was disappeared from T10 (P=0.034) to T90 (P=0.041). In the second phase, mydriasis was only observed in 2/8 eyes. ABSTRACT: A Vietnamese fish sauce Nuoc mam used in the present study was rich in oligopeptides of which nitrogen content accounted for 20% of total nitrogen. The high-molecular-weight peptide fractions fractionated by ion-exchange chromatographies and ultrafiltration enhanced sweetness and umami as well as sourness and bitterness of the fish sauce, and increased several flavor characteristics including continuity, first taste and aftertaste. Thus, it is apparent that large amounts of peptides produced during the long-term fermentation of fish sauce are responsible for the complicated taste of the sauce. From each fraction, 17 peptides in total were isolated and determined for their amino acid sequences. These di-, tri- and tetra-peptides synthesized by solid- or liquid-phase method gave any one of bitter, sour, umami taste, or practically no taste in the absence of salt. In the presence of 0.3% NaCl, however, almost all peptides showed sweet and umami tastes. Other than these 17 peptides, there existed many other kinds of peptides in the fish sauce. We investigated the influence of pneumoperitoneum#(PP) and postural change under
inhalation anesthesia with isoflurane, which is routinely used in dogs, on the
cardiovascular and respiratory systems. As test animals, 6 adult beagles were used. To
induce anesthesia, atropine, butorphanol and propofol were intravenously injected.
Anesthesia was maintained with 1.3 MAC (1.7%) isoflurane. The following were the
experiment conditions: I:E ratio, 1:1.9; tidal air exchange, 20 ml/kg;
and ventilation frequency, 14 times/min. Respiration was regulated so that the
PaCO2 was approximately 35 to 40 mmHg before the start of the experiment. PP
with CO2 (intraperitoneal pressure 15 mmHg) and a postural change (15°C) was
performed during the experiment. As parameters of circulatory kinetics, heart rate (HR),
mean aortic pressure (MAP), mean pulmonary arterial pressure (MPAP), central venous
pressure (CVP), femoral venous pressure (FVP) and cardiac output (CO) were measured. As
parameters of respiratory kinetics, airway pressure (PAW) and blood gas (BG) were
measured. There were significant increases in HR, MAP, MPAP, CVP, FVP, CO, PAW and
PaCO2 after PP in the horizontal position. There were significant increases
in CVP, FVP, PAW and PaCO2 after PP in the Trendelenburg position. There were
significant increases in the MPAP, CVP, FVP, PAW and PaCO2 after PP in the
inverse Trendelenburg position. There was a significant difference in FVP after PP between
the Trendelenburg position and inverse Trendelenburg position. The results of this
experiment suggest that appropriate anesthesia control, such as changing the ventilation
conditions after PP, is required for laparoscopic surgery under inhalation anesthesia with
