Feeding and palatability trials were conducted with four cultivars (cv) of switchgrass (Panicum virgatum L.), fertilized at three levels of N (0, 75, or 150 kg of N/ha) in 2 yr. Wether sheep had ad libitum access to chopped hays, and intake, apparent digestibility, particle passage rates, and concentrations of blood metabolites were determined. Palatability was measured with mature sheep in cafeteria trials. Nitrogen fertilization did not affect (P > .05) DM digestibility (DMD) or DMI, but there was a year x N interaction (P < .01) for NDF digestibility. Dry matter digestibility values for combined years and N levels were 56.9, 54.4, 56.6, and 57.9% (P < .01) for Pathfinder, New Jersey 50, Kentucky 1625, and Trailblazer cv, respectively; mean DMI values were 60.4, 60.8, 57.7, and 64.0 g/kg BW.75 (year x cv, P < .01). An apparently greater quality of Trailblazer was masked by weed invasion of N-fertilized stands of this cv in yr 2, with changes in hay composition. Lambs adapted to hay diets with time; mean DMI for cv and N levels combined increased (P < .001) from 50.5 to 71.4 g/kg BW.75 between wk 2 and 3 and wk 10 and 11, with no change in DMD. Intakes of NDF increased from 37.6 to 55.6 g/kg BW.75, an increase of 48%. Marker (Yb) measurements indicated little change in particle passage rates with treatment. There were no major differences in blood composition, apart from increases in blood urea N, as a result of N fertilization. Cafeteria trials showed preference by sheep for Trailblazer and KY 1625 compared with NJ 50 and Pathfinder, with a N x cv interaction (P < .01). Trailblazer was preferred to KY 1625 in a two-choice situation (P < .01). Results show relatively small effects of cv and N fertilization on quality of switchgrass and indicate the need for a lengthy period of adaptation by sheep to the feeding of a warm-season grass.  相似文献

Malignant catarrhal fever is a fatal lymphoproliferative and degenerative disease of ruminants. One causative agent is the gammaherpesvirus alcelaphine herpesvirus 1 (AHV-1), which produces no disease in its natural host, the wildebeest (Connochaetes species). Epidemiological evidence implicates sheep as the carrier of a similar virus. However, attempts to culture this virus from sheep or from animals affected with sheep-associated malignant catarrhal fever (SA-MCF) have failed. Lymphoblastoid cells have been propagated from cattle, deer and rabbits with SA-MCF. Although these cells show no evidence of viral particles or antigens, hybridisation experiments now show that they contain DNA sequences homologous to those of AHV-1. A genomic library was constructed from one of these lymphoblastoid cell lines and a clone identified which hybridised to cloned AHV-1 DNA. The authors believe that this clone contains part of the SA-MCF viral genome, and that the SA-MCF virus and AHV-1 are closely related gammaherpesviruses.  相似文献

OBJECTIVE: To evaluate the value of microarthroscopy in the equine midcarpal joint using the vital stains methylene blue, trypan blue, neutral red, and Janus green B to observe components of the synovial lamina propria, vascular architecture, and synoviocytes. STUDY DESIGN: Experimental. ANIMALS: Ten horses. METHODS: Microarthroscopy of left and right midcarpal joints was performed with and without vital staining of the synovium. Four vital stains (methylene blue, trypan blue, neutral red, and Janus green B) were evaluated, with each stain used in 5 joints. Synovial biopsy specimens were collected from the dorsomedial and dorsolateral aspects of the joint. RESULTS: All dyes were biocompatible. At x 60 without vital staining, synovial surface topography, vascular network, and translucency were observed. Intra-articular vital dyes improved evaluation of synovial surface topography. At x 150 with vital staining, individual synoviocytes were clearly identified with all dyes, except neutral red. Although methylene blue provided the best in vivo microscopic differentiation of the structure of the intima, trypan blue had superior retention in conventionally processed synovial biopsies. CONCLUSIONS: Methylene blue, trypan blue, neutral red, and Janus green B stains can be used safely for microarthroscopy. Good visualization of cells and vascular network can be obtained by microarthroscopy, and microarthroscopic evaluation of the synovium compares favorably with conventional histologic evaluation of biopsy specimens. CLINICAL RELEVANCE: Microarthroscopy may be beneficial in both research and clinical diagnosis of equine articular diseases.  相似文献

Renal biopsy often is required to establish a definitive diagnosis in dogs and cats with renal disease. In this retrospective study, we determined the complications of renal biopsy as well as factors that may be associated with development of complications and procurement of adequate renal biopsy specimens in 283 dogs and 65 cats. Data extracted from medical records at 4 institutions were evaluated using logistic regression. Proteinuria was the most common indication for renal biopsy in dogs. Complications were reported in 13.4 and 18.5% of dogs and cats, respectively. The most common complication was severe hemorrhage; hydronephrosis and death were uncommon. Dogs that developed complications after renal biopsy were more likely to have been 4 to < 7 years of age and > 9 years, to weigh < or = 5 kg, and to have serum creatinine concentrations > 5 mg/dL. The majority of biopsies from both dogs (87.6%) and cats (86.2%) were considered to be of satisfactory quality. Biopsies from dogs were more likely to be of high quality if they were obtained when the patient was under general anesthesia and more likely to contain only renal cortex if they were obtained by surgery. We concluded that renal biopsy is a relatively safe procedure, with a low frequency of severe complications. Hospital practices and patient variables have the potential to impact both the quality of the specimen obtained and the rate of complications.  相似文献
