We isolated Xanthomonas oryzae pv. oryzae mutants deficient in the phosphoenolpyruvate : carbohydrate phos-photransferase system, a major glucose transport system in
bacteria, using the glucose analogue 3-deoxy-3-fluoro-d-glucose (3FG). Glucose uptake by the mutants was decreased to 15–35% of the parental strain, and growth greatly decreased
in synthetic media containing glucose as a sole sugar source. Growth of the mutants in rice leaves was, however, similar to
the wild type. These findings suggest that glucose is not necessarily a major carbohydrate source for X. o. pv. oryzae in rice leaves.

To identify the character impact odorant of high-heat skim milk powder (HHSMP), a comparative study using ultrahigh-temperature (UHT) milk was performed. Aroma concentrate was prepared by column adsorption combined with simultaneous distillation-extraction. Aroma extract dilution analysis (AEDA) revealed 58 aroma peaks with flavor dilution (FD) factors ranging from 10 to 3000; from these, 41 compounds were identified and 7 compounds were tentatively identified (FD factor > or = 300). Among these HHSMP and UHT milk components, methyl 2-methyl-3-furyl disulfide and bis(2-methyl-3-furyl) disulfide, which appeared to be generated during the processing of each product, were identified. When the results of the AEDA of both samples were compared, it was considered that the characteristic aroma of HHSMP was not explained by a single compound but instead formed from a mixture of several types of compounds contained in common with the UHT milk. The contribution of these compounds to the aroma of HHSMP was confirmed by an aroma simulation experiment. 相似文献

We examined the response of the tea plant (Camellia sinensis L.) to aluminum (Al) exposure under sterile conditions, focusing specifically on the secretion of low molecular mass organic compounds from roots. After germination in agar medium, tea seedlings together with medium were placed on agar containing 0.4?mM Al with 0.2% hematoxyline (hematoxylin-Al medium). The purple color of the hematoxylin-Al medium was observed to fade gradually, until none of the color remained 6 days later. The tea seedlings were then treated with simple calcium solution (0.2?mM, at pH 4.2) containing AlCl3, which ranged in concentration from 0 to 0.8?mM, for 24?hrs. The amount of oxalate secreted into the medium increased as the external Al concentration increased, while the concentrations of malate and citrate in the medium remained unchanged. Oxalate secretion started within 30?min after Al exposure and increased linearly thereafter. The findings demonstrated that oxalate was a key compound in the Al-tolerance mechanism employed by the tea plant, which detoxifies Al3+ externally in the rhizosphere. In addition to oxalate, caffeine was also secreted by tea roots in response to Al exposure. It is possible that caffeine excretion from the roots of tea plants may stimulate root growth through the inhibition of callose deposition in root tips. 相似文献

Feline immunodeficiency virus (FIV) infection is characterized by chronic overactivation of immune and inflammatory system, resulting in anergic state and dysfunction of immune cells. Lactoferrin (LF), a glycoprotein present in exocrine secretions and neutrophils, plays an important role in host defense system. Our previous study showed that oral administration of bovine LF (bLF) suppressed oral inflammation, improved the clinical symptoms and decreased serum gamma-globulin as a marker of inflammation in FIV-infected cats with intractable stomatitis. The anti-inflammatory effect was partly involved in regulation of neutrophil function by bLF. In this study, to clarify the relationship between anti-inflammatory effects of bLF and peripheral blood mononuclear cells (PBMC), we examined the effect of bLF on proliferation, cell cycle progression and cytokine expression in mitogen-activated PBMC. MTT [3-(4,5-dimethylthiazol-2-yl)- 2,5-diphenyl tetrazolium bromide] assay showed that bLF inhibited the concanavalin A (ConA)-induced cell proliferation in FIV-infected cats with the asymptomatic carrier and AIDS-related complex (ARC) phase. Bovine LF restored ConA-induced cell cycle progression and resulted in suppression of the induced apoptosis in feline PBMC. Real-time RT-PCR showed that bLF suppressed ConA-induced expression of interferon-gamma and interleukin-2 in cells of the ARC group regardless of the time of its addition to the medium. These results suggest the hypothesis that therapy with bLF may have the potential to improve and protect functions of overactivated lymphocytes by modulating the cell proliferation, cell cycle and cytokines expression in cats in terminal stage of FIV infection. 相似文献

In this study, the proteins contained in royal jelly (RJ) produced by Africanized honeybees and European honeybees (Apis mellifera) haven been analyzed in detail and compared using two-dimensional gel electrophoresis, and the N-terminal amino acid sequence of each spot has been determined. Most spots were assigned to major royal jelly proteins (MRJPs). Remarkable differences were found in the heterogeneity of the MRJPs, in particular MRJP3, in terms of molecular weights and isoelectric points between the two species of RJ. Furthermore, during the determination of the N-terminal amino acid sequence of each spot, for the first time, MRJP4 protein has been identified, the existence of which had been only implied by cloning of its cDNA sequence. The presence of heterogeneous bands of glucose oxidase was also identified. Thus, the results suggest that two-dimensional gel electrophoresis provides a suitable method for the qualitative analysis of the proteins contained in RJ derived from different honeybee species. 相似文献

The nature of soil is modified differently depending upon the artificial condition such as its utilization or management. It is therefore expected that the microbiological characteristic of soil is changed also. Greaves1) and Williams2) reported that the reclamation of virgin soil brought about a change of bacterial count. Suzuki et al3) observed that the kind of fungi differed between a virgin and a cultivated soil and that the vegetative mycelium was numerous in the former than in the latter. On the other hand, according to W aksman and Starkey4), the bacterial count differed depending upon the fertility of soil. Singh5) reported also that the number of fungi and actinomycetes was higher in a fertile than in an infertile soil. Lochhead6), and Lochhead and Chasen studied the bacterial flora of a fertile (long-continued application of manure) and an infertile (no fertilizer for many years) soil and found that a certain difference could be observed when morphological, physiological and nutritional classification are tried. 相似文献

The central districts of Japan are mountainous areas rising to 3,000 metres which are predominantly covered with forest vegetation. A comprehensive and systematic classification of the soils in this region has not yet been made, but the following soil types have been reported to occur: - 1. 1. Abstract It has been well documented by short-term artificial experiments that the CH4 uptake is inhibited by N input, especially NH4 p+-N input. To investigate the effect of the natural N input by throughfall and other factors on the CH4 uptake in forest soils, we measured the CH4 uptake rates for 6 months during the snow-free period of the year and N input by throughfall throughout the year at 10 sites in Hokkaido, Japan, from 1997 to 2002. Water filled pore space (WFPS) and pH values in the soils varied widely among the sites (38-93% and 3.9-6.2, respectively). The rates of NH4 p+-N and NH3 p--N inputs ranged from 1.3 to 6.9 kg N hap-1 yearp-1 and from 0.8 to 2.9 kg N hap-1 yearp-1, respectively. The NH4 p+-N input was generally higher than the NH3 p--N input. Total N input by throughfall amounted to 2.3-9.4 kg N hap-1 yearp-1. The highest CH4 uptake rate occurred within the period from July to September (41-215 μg CH4 mp-2 hp-1) each year at most sites. CH4 uptake rate was relatively low (~50 μg CH4 M-2 hp-1) at northern sites, while a high CH4 uptake rate was observed throughout the year 100 (? CH4 mp-2 hp-1) at southern sites. The mean CH4 uptake rates were significantly different among the sites. Cumulative CH4 uptake ranged from 1.4 to 6.6 kg CH4 hap-1 [184 d]p-1 with a mean values of 3.22 ± 1.36 kg CH4 hap-1 [184 d]p-1. Cumulative CH4 uptake increased with increasing temperature and decreased with an increase in precipitation (Rain), NH4 p+-N input (TFNH4) WFPS, soil total C (TC), and total N (TN). There was a quadratic relationship between the CH4 uptake and NH3 p--N input (TFNO3), soil pH, and C / N ratio in soil. A regression equation was obtained as follows to predict the CH4 uptake in forest soils: Cumulative CH4 uptake = 0.47 / Rain + 0.38 / TFNH4 + 0.34 / TC - 0.30 / TFN03 (R p2 = 0.74, p = 0.0001). This equation indicates that atmospheric N input into forest soils is one of the main factors that control cumulative CH4 uptake with precipitation, total carbon content in soil in Hokkaido, Japan. 相似文献

Recent studies have shown that the incorporation of ammonium nitrogen into amino acids in the leaves is strictly dependent on light (1-4). It is speculated that the effect of light on ammonium assimilation may be through the synthesis of the precursors of amino acids, or by the supply of the energy required for amination and amidation with organic acids. In the Vicia faba chloroplasts Givan et al. (1) exhibited that the synthesis of glutamic acid from a-ketoglutarate was linked with the generation of reduced pyridin nucleotide by photosynthetic electron transport. Mitchell and Stocking (2) suggested the direct coupling of glutamine formation with photophosphorylation in the pea chloroplasts. On the other hand. the processes of nitrate assimilation are more indebted to light than those of ammonium assimilation, because the former ones involve the reduction of nitrate to ammonium which is believed to be light-dependent (5). Canvin and Atkins (6). and Atkins and Canvin (7) reported that the incorporation of 15N-labeled ammonium and nitrate into amino acid fractiom was depressed by the dark treatment and by photosystem inhibitors; 3-(3′,4′-dichlrophenyl)-1-1-dimethylurea (DCMU) and carbonyl-cyanide-m-chlorophenyl-hydrazone(CCCP). 相似文献
