An experiment was conducted to determine the effects of supplemental dietary l-carnitine, chromium picolinate (Cr-Pic) and their combination on growth performance and serum total protein, cholesterol, triglyceride and glucose of rainbow trout (Oncorhynchus mykiss). A total of 276 rainbow trout were randomly allocated to four groups. Fish (average initial body weight = 151 ± 1.69 g) were fed a basal diet without supplemental l-carnitine and chromium picolinate in the control group. The basal diet was supplemented with either 500 mg/kg l-carnitine, 1.6 mg/kg chromium picolinate or 500 mg/kg l-carnitine plus 1.6 mg/kg chromium picolinate for experimental groups 1 (C), 2 (Cr-Pic) and 3 (C + Cr-Pic), respectively. Fish were fed twice a day to apparent satiation for 58 days. Weight gain, growth rate, feed consumption and feed conversion ratio (feed/gain) were calculated for the whole period. At the end of the study, six fish were chosen randomly from each tank to represent the experiment and frozen at −20°C for subsequent dorsal muscle analysis. The results showed no significant differences in final body weight, weight gain or feed conversion ratio among groups. There were also no significant differences in serum total protein, cholesterol, triglyceride or glucose concentrations among groups. Significantly higher lipid concentration of dorsal muscle was observed in experimental groups 1 and 3. In conclusion, supplemental dietary l-carnitine, chromium picolinate and their combination have no beneficial effects on improving growth performance and feed conversion ratio in rainbow trout. However, dietary l-carnitine slightly increased lipid concentration in dorsal muscle of rainbow trout.  相似文献

In their natural environment, microbes organize into communities held together by an extracellular matrix composed of polysaccharides and proteins. We developed an in vivo labeling strategy to allow the extracellular matrix of developing biofilms to be visualized with conventional and superresolution light microscopy. Vibrio cholerae biofilms displayed three distinct levels of spatial organization: cells, clusters of cells, and collections of clusters. Multiresolution imaging of living V. cholerae biofilms revealed the complementary architectural roles of the four essential matrix constituents: RbmA provided cell-cell adhesion; Bap1 allowed the developing biofilm to adhere to surfaces; and heterogeneous mixtures of Vibrio polysaccharide, RbmC, and Bap1 formed dynamic, flexible, and ordered envelopes that encased the cell clusters.  相似文献

Race-specific resistance against powdery mildews is well documented in small grains but, in other crops such as grapevine, controlled analysis of host-pathogen interactions on resistant plants is uncommon. In the current study, we attempted to confirm powdery mildew resistance phenotypes through vineyard, greenhouse, and in vitro inoculations for test cross-mapping populations for two resistance sources: (i) a complex hybrid breeding line, 'Bloodworth 81-107-11', of at least Vitis rotundifolia, V. vinifera, V. berlandieri, V. rupestris, V. labrusca, and V. aestivalis background; and (ii) Vitis hybrid 'Tamiami' of V. aestivalis and V. vinifera origin. Statistical analysis of vineyard resistance data suggested the segregation of two and three race-specific resistance genes from the two sources, respectively. However, in each population, some resistant progeny were susceptible in greenhouse or in vitro screens, which suggested the presence of Erysiphe necator isolates virulent on progeny segregating for one or more resistance genes. Controlled inoculation of resistant and susceptible progeny with a diverse set of E. necator isolates clearly demonstrated the presence of fungal races differentially interacting with race-specific resistance genes, providing proof of race specificity in the grape powdery mildew pathosystem. Consistent with known race-specific resistance mechanisms, both resistance sources were characterized by programmed cell death of host epidermal cells under appressoria, which arrested or slowed hyphal growth; this response was also accompanied by collapse of conidia, germ tubes, appressoria, and secondary hyphae. The observation of prevalent isolates virulent on progeny with multiple race-specific resistance genes before resistance gene deployment has implications for grape breeding strategies. We suggest that grape breeders should characterize the mechanisms of resistance and pyramid multiple resistance genes with different mechanisms for improved durability.  相似文献

Efforts to breed for the ability to tuberize under long days or for the length of tuber dormancy should benefit not only from information on the location of genes associated with these traits, but also from an understanding of the hormones the genes control. Understanding the role of the respective genes is a logical step in developing the best breeding strategy, both for conventional breeding and for gene transfer using molecular techniques. Polygene mapping affords a way to achieve such understanding and is also a tool to study the physiology of potato tuberization and potato tuber dormancy. The addition of DNA markers to a plant population facilitates the mapping of polygenes that control quantitatively inherited traits segregating in the population. A quantitative trait locus denotes a region of chromosome that is linked to the marker gene and which has a significant effect on the quantitative trait under study. Our approach has been to find the quantitative trait loci associated with tuberization and tuber dormancy in a segregating diploid population, and then to map the population for quantitative trait loci associated with levels of hormones implicated in the control of these two traits. We are using a population derived from a hybrid between haploidSolanum tuberosum andS. berthaultii that was backcrossed to a different haploidS. tuberosum. We have found ten quantitative trait loci for the ability to tuberize under long days and eight quantitative trait loci for tuber dormancy. In the same population we have found one or more quantitative trait loci for polyamines, abscisic acid, tuberonic acid, tuberonic acid glucoside, zeatin riboside, and gibberellin A1. Some of the hormone quantitative trait loci have coincided with quantitative trait loci for tuberization or dormancy. Implications of such commonality are discussed, along with the usefulness and limitations of the methods.  相似文献

Of the various client groups with interests in irrigation system performance, farmers are probably the least often considered. This is, at least in part, attributable to the profound difficulty encountered in attempting to obtain reliable measurements of performance at farm level in most systems. Yet irrigation systems should be seen as a service to farmers, whose perspective on the utility of the supply is an important component of performance monitoring. Direct measurement at field level is generally not feasible and an alternative approach is proposed utilising fuzzy set theory. The procedure can be seen as a rapid appraisal technique which is also applicable to performance assessment in informal farmer-managed systems. The usefulness of the approach lies in its ability to gather data in imprecise linguistic terms from individual farmers and to aggregate them rationally and objectively.  相似文献

The antimicrobial activities of chloroform, methanol and aqueous extracts of Plicosepalus acaciae leaves and stem are reported. Samples of tubers collected from commercial potato seed lots produced across North America were assayed forVerticillium dahliae. V. dahliae was successfully isolated from 65 of 224 seed lots tested, a successful isolation rate of nearly 30%. Vegetative compatibility of the isolates was assessed through complementation tests using nitrate non-utilizing mutants. AllV. dahliae isolates belonged to vegetative compatibility group (VCG) 4. Of the 162 isolates recovered, 64% belonged to VCG 4A, 33% to VCG 4B, and 3% to VCG 4AB. All 39 of the isolates tested in the greenhouse on potato cv. Superior were pathogenic to potato. Disease symptoms developed earlier, were more severe, and plants died earlier when inoculated with VCG 4A compared to 4B isolates. As a group, AUSPC values were significantly higher (p=0.05) for VCG 4A than for 4B isolates. These data suggest that (1) commercial certified seed tubers from diverse locations are commonly infected withV. dahliae and thus may serve as primary sources of the pathogen; (2) potato isolates ofV. dahliae in North America belong to VCG 4A and 4B and these strains are widely distributed via seed tubers; and (3) VCG 4A and 4B are distinct pathotypes ofV. dahliae that vary in their aggressiveness to potato.  相似文献
