An international effort (sponsored by the Australian Centre for International Agricultural Research) is being made to develop oral vaccines that will protect village chickens against Newcastle disease. The vaccines being used are derivatives of the avirulent Australian V4 strain that have been selected for enhanced heat resistance. The present study, undertaken in Sri Lanka, used local processed (parboiled) rice as a vehicle for the vaccine. Chickens receiving two doses of vaccine on cooked, parboiled rice were completely protected against contact challenge with the virulent SL 88/1 Sri Lankan strain of Newcastle disease virus Chickens kept in contact with these vaccinated chickens were similarly protected. Lower levels of protection were achieved with vaccine given on uncooked parboiled rice. V4 vaccine administered intranasally also gave complete protection. Serums from vaccinated chickens that survived challenge were tested for haemagglutination-inhibition antibodies, using both vaccine virus and challenge virus as antigens. In contrast to mammalian systems, avian species lack a resident or harvestable macrophage population in the abdominal exudate. Peritoneal macrophages in the chicken can be elicited if an inflammatory agent such as sephadex is injected. This study examines the kinetics of different macrophage populations, derived by different methods of isolation and from different hosts, with respect to the elicited oxidative burst upon infection with host-adapted Salmonella serotypes.
The nature of the oxidative burst elicited by murine and avian-derived and cell line macrophages was determined after stimulation with phorbol myristate (PMA), zymosan A, and Salmonella serotypes. Both murine and chicken peritoneal macrophages, chicken blood monocytes and corresponding cell lines, J774A.1 and HD-11, were unable to produce a detectable chemiluminescent (CL) response after interaction with Salmonella using the luminescent probe luminol. However, both PMA and zymosan A induced a CL response in all cell types, with PMA eliciting a higher and earlier peak response (pkH) than zymosan A. Lucigenin-enhanced CL in both murine and chicken macrophages was achieved with PMA, zymosan A and Salmonella serotypes. In this case, zymosan A induced higher responses than PMA. In the peritoneal macrophages of both hosts, there were no significant differences in the oxidative burst induced by the different Salmonella serotypes. However, the J774A.1 (murine) cells demonstrated significant differences, with S. enterica serotype Choleraesuis (S.choleraesuis and S. gallinarum producing the highest response. In the HD-11 (chicken) cells, S. choleraesuis and S. dublin elicited the higher CL. With both cell lines, S. abortusovis failed to induce an appreciable CL response.
In these experiments it was demonstrated that oxidative burst was not detectable in monocytes/macrophage populations using luminol, which suggests a link to the lack of a myeloperoxidase system in these cells. Lucigenin-enhanced CL appeared independent from the myeloperoxidase system, indicating production of another oxidative species compared with luminol. No discernable effect of host specificity with regard to Salmonella serotype and respective host was seen in host-derived or cell line macrophages, and cell line macrophages displayed altered functional characteristics with regard to oxidative burst in comparison with their primary counterparts. 相似文献
Summary Grain filling rate in wheat (Triticum aestivum L. emend. Thell.) positively influences grain yield under a wide range of conditions. The effective utilization of this trait in breeding depends on an understanding of its genetic control. A study was, therefore, conducted to determine the genetic basis of grain filling rate in six crosses of wheat. Higher order genic interactions and/or linkage were important in the genetic regulation of grain filling rate (GFR) in the majority of crosses. Additive ([d]) and dominance ([h]) gene effects were important in the control of GFR in main ears (ME) and whole plant ears (WPE). Additive and additive × additive epistatic effects were the most important in the genetic control of GFR in last ears (LE). Location effects on genetic effects for GFR were significant (P < 0.05) in all ear types of some crosses except in ME. Genotype × environment interaction effects were important (P < 0.001) in LE and WPE.It was concluded that the inheritance of GFR is complex and is dependent on ear type. Breeding procedures that facilitate the exploitation of non-additive and additive gene effects were recommended for the genetic improvement of grain filling rate of wheat. 相似文献
A new type of lake and shore assemblage has been found in the Late Triassic age rocks of North Carolina and Virginia (Dan River group). It includes abundant aquatic reptiles, fishes, at least seven orders of insects, crustaceans, and a diverse flora. Cyclic changes in the fauna and flora correlate with sedimentary cycles, which together reflect the repetitive development and extinction of large meromictic lakes. 相似文献
The discovery of the first definitively correlated earliest Jurassic (200 million years before present) tetrapod assemblage (Fundy basin, Newark Supergroup, Nova Scotia) allows reevaluation of the duration of the Triassic-Jurassic tetrapod extinction event. Present are tritheledont and mammal-like reptiles, prosauropod, theropod, and ornithischian dinosaurs, protosuchian and sphenosuchian crocodylomorphs, sphenodontids, and hybodont, semionotid, and palaeonisciform fishes. All of the families are known from Late Triassic and Jurassic strata from elsewhere; however, pollen and spore, radiometric, and geochemical correlation indicate an early Hettangian age for these assemblages. Because all "typical Triassic" forms are absent from these assemblages, most Triassic-Jurassic tetrapod extinctions occurred before this time and without the introduction of new families. As was previously suggested by studies of marine invertebrates, this pattern is consistent with a global extinction event at the Triassic-Jurassic boundary. The Manicouagan impact structure of Quebec provides dates broadly compatible with the Triassic-Jurassic boundary and, following the impact theory of mass extinctions, may be implicated in the cause. 相似文献
The repeated association during the late Neoproterozoic Era of large carbon-isotopic excursions, continental glaciation, and stratigraphically anomalous carbonate precipitation provides a framework for interpreting the reprise of these conditions on the Late Permian Earth. A paleoceanographic model that was developed to explain these stratigraphically linked phenomena suggests that the overturn of anoxic deep oceans during the Late Permian introduced high concentrations of carbon dioxide into surficial environments. The predicted physiological and climatic consequences for marine and terrestrial organisms are in good accord with the observed timing and selectivity of Late Permian mass extinction. 相似文献
The minimum temperature of growth of the mesophilic bacterium Pseudomonas aeruginosa has been significantly lowered from approximately 11 degrees to 0 degrees C. This shift in the minimum temperature of growth was accompanied by a corresponding decrease in the maximum temperature of growth. Transfer of this genetic characteristic by a transducing phage grown on a psychrophile or by ultraviolet mutagenesis was used to accomplish these shifts in range of growth temperature. 相似文献
An improved procedure was employed for linking ferritin to antibodies against prolyl hydroxylase, the enzyme that synthesizes the hydroxyproline in collagen. By electron microscopy, the enzyme was then found to be localized in cisternae of the rough endoplasmic reticulum of embryonic tendon cells; this indicates that hydroxylation of proline occurs while newly synthesized polypeptides are fed into the cisternae. 相似文献
