The primary objectives of this study were to determine the seroprevalence of West Nile virus (WNV) infection of horses in Saskatchewan in 2003 and to identify risk factors for the infection. Blood samples were collected in August and October from 212 horses in 20 herds in 5 geographic zones. After accounting for within-herd clustering, the proportion of horses that had been infected with WNV, as determined by IgG and IgM antibody response, was 55.7% (95% confidence interval, 44.9% to 65.8%). The proportion of antibody-positive horses differed among herds (0% to 100%) and across ecoregions (20% to 76%). Horses in southern ecoregions were more likely to have either IgM antibodies or IgG concentrations suggesting infection than were horses in northern ecoregions. The use of mosquito-control measures was associated with decreased risk. After accounting for ecoregion, there was no difference between recipients of an inactivated WNV vaccine and nonrecipients in the occurrence of antibodies reflecting natural infection.  相似文献

Landscape diagnosis provides a bridge between scientific knowledge and socio-economic issues that is needed to meet the demands of sophisticated landscape planning and management. The diagnostic assessment of landscape functions (capacities, goods and services supported by the landscape) at different spatio-temporal scales is a valuable tool that can solve the transformation problem. A variety of landscape classification systems – including biophysical and landscape units – can be applied as a spatial reference system. Examples are described from the multitude of approaches to assess landscape functions that can be employed in landscape diagnosis. The theoretical and methodological aspects of the approach are illustrated using examples both from Germany and the Czech Republic. The examples focus on landscape functions such as groundwater recharge, regulation of water balance, and resistance to wind erosion. In addition, the rarity of and threats to landscape types, landscape aesthetic values, and the landscape character and landscape persistence are discussed.  相似文献

Our objective was to identify temperature-related risk factors associated with the colonization of broiler-chicken flocks with Campylobacter spp. in Iceland, with an underlying assumption that at minimum ambient temperatures, flies (Musca domestica) play a role in the epidemiology and seasonality of Campylobacter. At slaughter, pooled caecal samples were obtained from 792 flocks that hatched between March 15 and September 15 each year from 2001 to 2004, and cultured for Campylobacter. Daily temperature data (average, maximum and minimum temperatures) were obtained from the Icelandic Meteorological Office. We determined the closest weather station to each farm using ArcGIS 9. We then used logistic-regression models (with a random-effects term for farm) to examine associations between flock positivity and temperature-related predictors. The overall prevalence of Campylobacter was 27.4%, and the month with the highest prevalence (54.4%) was August. The final model included cumulative degree-days (CDD) above an average temperature of 4.4 degrees C and the presence of 1 or more days below a maximum temperature of 8.9 degrees C (threshold below which fly activity was expected to be substantially reduced and egg laying was not expected to occur) during the period 2-4 weeks (i.e. days 8-28) before slaughter. Below 79 CDD, the risk of flock Campylobacter colonization was generally low. Between 79 and 139 CDD, the risk increased gradually, but was lower for flocks raised during periods with 1 or more days <8.9 degrees C than flocks raised without days <8.9 degrees C. The risk increased sharply under conditions of high CDD (>139) and when the maximum temperature remained >8.9 degrees C during the period 2-4 weeks before slaughter.  相似文献

The parasite Echinococcus multilocularis is currently of great concern in Europe because of its spreading behavior, which has public health implications. This study investigates the spatiotemporal distribution of Echinococcus multilocularis in Red Foxes. The infection status of 8459 foxes was sampled from 43 regions in the north German province of Lower Saxony over three investigation periods 1991--1994, 1994--1997 and 2003--2005. Linear empirical Bayesian smoothing within the binomial model was used to produce smoothed choropleth maps. Geostatistical kriging was applied to generate prevalence risk maps. Further geostatistical modeling of the prevalence difference between study periods facilitated spatiotemporal trend investigations. The spatial scan statistic was used for cluster detection analysis. The average prevalence risk for Lower Saxony increased from about 12 to 20% during 1991--2005. Specifically the increases from first to second and to third study periods were estimated by 3.3% (CI 95%: 0.6%-5.9%) and 8.5% (CI 95%: 5.2%-11.8%), respectively. Infections in foxes were clustering and a location stable disease cluster was detected in the south of the province. This study is the first showing evidence for steady emergence of Echinococcus multilocularis in Red Foxes. First cases of human Alveolar Echinococcosis were recorded recently.  相似文献

Rapid advances in microscopy have boosted research on cell biology. However sample preparation enabling excellent reproducible tissue preservation and cell labeling for in depth microscopic analysis of inner cell layers, tissues and organs still represents a major challenge for immunolocalization studies. Here we describe a protocol for whole-mount immunolocalization of proteins which is applicable to a wide range of plant species. The protocol is improved and robust for optimal sample fixation, tissue clearing and multi-protein staining procedures and can be used in combination with simultaneous detection of specific sequences of nucleic acids. In addition, cell wall and nucleus labelling can be implemented in the protocol, thereby allowing a detailed analysis of morphology and gene expression patterns with single-cell resolution. Besides enabling accurate, high resolution and reproducible protein detection in expression and localization studies, the procedure takes a single working day to complete without the need for robotic equipment.  相似文献

Melanized focal changes in skeletal muscle of farmed Atlantic salmon (Salmo salar) are a major quality problem. The aetiology is unknown, but infection with Piscine orthoreovirus (PRV) has been associated with the condition. Here, we addressed the pathogenesis of red and melanized focal changes and their association with PRV. First, a population of farmed fish (PRV‐negative prior to sea transfer) was sequentially investigated throughout the seawater period. The fish were autopsied and tested for PRV infection. Muscular changes were described by macroscopy and histology, and a classification system was established. Second, in an experimental infection trial, PRV was injected intramuscularly to induce changes. The farmed fish was gradually infected with PRV. Red focal changes occurred throughout the observation period with a low prevalence regardless of PRV status. Melanized changes were highly diverse and their prevalence increased during the trial. Changes of low macroscopic grade and histological category were more prevalent in PRV‐negative fish. Diffuse granulomatous melanized changes only occurred after PRV infection. No muscular changes were observed in the experimentally challenged fish. Our studies do not indicate that PRV infection causes red focal changes, but seems important in the development of granulomatous melanized changes.  相似文献

The intensive irrigated rice-wheat systems in the northwest Indo-Gangetic Plains of South Asia are built on a long tradition of canal irrigation and the more recent advent of tubewells. Findings from farm surveys are used to examine water management and water productivity in the rice-wheat belt of India's Haryana State and Pakistan's Punjab province. Attributes of the irrigation sources help explain the widespread interest in groundwater use and the relative demise of canal water use. In each area groundwater now is the main irrigation source, used either solely or in conjunction with surface water. The ownership of tubewells is near universal among the surveyed farms, whereas conjunctive water use is more widespread during the monsoon season, among better endowed farmers and in the Pakistan Punjab. In Pakistan Punjab farmers primarily rely on diesel powered tubewells whereas Haryana farmers mainly use relatively cheaper electric power. Water productivity indicators for rice are markedly lower than those for wheat—largely reflecting significantly higher water inputs in paddy cultivation—but also vary between the study areas and by the prevailing water use, reflecting the limited incentives for farmers to use water wisely. A combination of technological, land use and market based approaches is likely to be most effective in achieving sustainable water management in these intensive cereal systems.  相似文献
