The Eg–Uur River ecosystem in north‐central Mongolia provides an opportunity to study salmonid species in a system that has already experienced significant climate change. These species are currently imperilled in Mongolian waters, with Baikal grayling (Thymallus arcticus baicalensis) listed as near‐threatened and lenok (Brachymystax lenok) listed as vulnerable on the Mongolian red list. Air temperature records demonstrate that in the last 40 years Northern Mongolia's rate of warming has been three times greater than the northern hemisphere average. Despite alarming trends in air temperatures, little is known of the thermal ecology of these species. Due to the threat of climate change to these species, the objective of our study was to quantify metabolic costs for these species from streamside routine metabolic measures and derive bioenergetics models that we used to assess potential climate change response. Streamside measurements of metabolism were remarkably consistent with expectations from measures of other salmonids gathered under more closely controlled laboratory conditions. Metabolism increased exponentially with temperature for both species. The resulting preliminary bioenergetics models suggest these species are already experiencing temperatures near their upper levels for growth during summer and conditions are expected to deteriorate with warming. Even a modest 2 °C increase in water temperatures during ice out would result in a 59% reduction in growth of lenok, and an inability of Baikal grayling to grow (if food levels remained unchanged) or a 14–23% increase in consumption in order to maintain current growth rates.  相似文献

The cell wall envelope of Gram-positive pathogens functions as a scaffold for the attachment of virulence factors and as a sieve that prevents diffusion of molecules. Here the isd genes (iron-regulated surface determinant) of Staphylococcus aureus were found to encode factors responsible for hemoglobin binding and passage of heme-iron to the cytoplasm, where it acts as an essential nutrient. Heme-iron passage required two sortases that tether Isd proteins to unique locations within the cell wall. Thus, Isd appears to act as an import apparatus that uses cell wall-anchored proteins to relay heme-iron across the bacterial envelope.  相似文献

Salmonella Heidelberg and Salmonella Typhimurium are among the most common serotypes responsible for human salmonellosis in Ontario. Introduction of the Ontario Investigation Tools (OIT) in 2014 allowed for standardized case investigation and reporting. This study compared the risk factors and symptomatology for sporadic S. Heidelberg and S. Typhimurium cases reported in Ontario in 2015, following implementation of the OIT. Multilevel logistic regression models were applied to assess associations between serotype and individual‐level demographic characteristics, exposures and symptoms for sporadic confirmed cases of S. Heidelberg and S. Typhimurium in Ontario in 2015. There were 476 sporadic cases of S. Typhimurium (n = 278) and S. Heidelberg (n = 198) reported in Ontario in 2015. There were significant associations between the odds of the isolate from a case being one of these serotypes, and travel, consumption of sprouts (any type), contact with reptiles and development of malaise, fever or bloody diarrhoea. The S. Typhimurium and S. Heidelberg cases differed in both symptom presentation and risk factors for illness. Case–case comparisons of Salmonella serotypes have some advantages over case–control studies in that these are less susceptible to selection and recall bias while allowing for rapid comparison of cases to identify potential high‐risk exposures that are unique to one of the serotypes when compared to the other. Comparing cases of two different Salmonella serotypes can help to highlight risk factors that may be uniquely associated with one serotype, or more strongly associated with one serotype compared to another. This information may be useful for understanding relative source attribution between common serotypes of Salmonella.  相似文献

Few antimicrobials are licensed for use in sheep in Canada, and the range of indications is narrow. Treatment in an "extra-label" manner may be ineffective. In addition, potentially harmful drug residues in food-animal products and antimicrobial resistance in bacteria may be associated with extra-label drug use (ELDU). No data had been documented on drug use, specifically antimicrobial use (AMU), in Ontario sheep, although it was thought that much use was extra-label. This study investigated AMU and ELDU on 49 lamb-producing Ontario sheep farms. Data were prospectively collected over 12 months from the participating farms, and farm-level practices were ascertained with a questionnaire. Treatment-level and farm-level variables were investigated for associations with rates of AMU by means of Poisson rate regression models fit with a generalized estimating equation to control for clustering at the farm level. Antimicrobials with high mean exposure rates included chlortetracycline (in feed), penicillins, and oxytetracycline. The exposure rate in lambs was significantly lower (P < 0.01) with antimicrobial treatment of systemic signs, respiratory disease, or wound or injury than with treatment of other reported diseases or conditions; it was also significantly lower with 3 or more lambing periods per year (α = 0.05). The exposure rate in adult sheep was significantly lower with treatment of 5 of the 6 most prevalent diseases or conditions (α = 0.05) and significantly higher with producer decision to treat and producer experience of 20 y or greater. Rates of using antimicrobials not licensed for use in sheep were high, as was extra-label use of licensed antimicrobials. Diseases reportedly treated most often with antimicrobials (e.g., systemic signs, mastitis) were significantly associated with lower rates of ELDU (α = 0.05). Compared with the rates in adult sheep, the mean rate of use of nonlicensed antimicrobials was similar in the lambs, whereas the mean rate of extra-label use of licensed antimicrobials was lower among the lambs. The results are useful in determining if public health concerns about antimicrobial use in Ontario sheep are warranted and in creating drug use and licensure strategies for the Canadian sheep industry.  相似文献

Humans normally experience the conscious self as localized within their bodily borders. This spatial unity may break down in certain neurological conditions such as out-of-body experiences, leading to a striking disturbance of bodily self-consciousness. On the basis of these clinical data, we designed an experiment that uses conflicting visual-somatosensory input in virtual reality to disrupt the spatial unity between the self and the body. We found that during multisensory conflict, participants felt as if a virtual body seen in front of them was their own body and mislocalized themselves toward the virtual body, to a position outside their bodily borders. Our results indicate that spatial unity and bodily self-consciousness can be studied experimentally and are based on multisensory and cognitive processing of bodily information.  相似文献

The polarized nature of epithelial cells is manifested by the nonrandom partitioning of organelles within the cells, the concentration of intercellular junctions at one pole, and the asymmetric distribution of proteins and lipids within the plasma membrane. These features allow epithelia to fulfill their specific tasks, such as targeted uptake and secretion of molecules and the segregation of different tissue compartments. The accessibility of Drosophila melanogaster and Caenorhabditis elegans to genetic and cell biological analyses, combined with the study of mammalian cells in culture, provides an ideal basis for understanding the mechanisms that control the establishment and maintenance of epithelial cell polarity and tissue integrity. Here, we focus on some of the best-studied junctions and membrane-associated protein complexes and their relation to cell polarity. Comparisons between fly, worm, and vertebrate epithelia reveal marked similarities with respect to the molecules used, and pronounced differences in the organization of the junctions themselves.  相似文献

  1. Freshwater biodiversity is declining at an unprecedented rate. Freshwater conservationists and environmental managers have enough evidence to demonstrate that action must not be delayed but have insufficient evidence to identify those actions that will be most effective in reversing the current trend.
  2. Here, the focus is on identifying essential research topics that, if addressed, will contribute directly to restoring freshwater biodiversity through supporting ‘bending the curve’ actions (i.e. those actions leading to the recovery of freshwater biodiversity, not simply deceleration of the current downward trend).
  3. The global freshwater research and management community was asked to identify unanswered research questions that could address knowledge gaps and barriers associated with ‘bending the curve’ actions. The resulting list was refined into six themes and 25 questions.
  4. Although context-dependent and potentially limited in global reach, six overarching themes were identified: (i) learning from successes and failures; (ii) improving current practices; (iii) balancing resource needs; (iv) rethinking built environments; (v) reforming policy and investments; and (vi) enabling transformative change.
  5. Bold, efficient, science-based actions are necessary to reverse biodiversity loss. We believe that conservation actions will be most effective when supported by sound evidence, and that research and action must complement one another. These questions are intended to guide global freshwater researchers and conservation practitioners, identify key projects and signal research needs to funders and governments. Our questions can act as springboards for multidisciplinary and multisectoral collaborations that will improve the management and restoration of freshwater biodiversity.

  1. The eradication of invasive species is a management strategy implemented to eliminate adverse impacts of invaders on native species communities. After eradications, follow‐up studies are done to confirm eradication of the invasive species and the short‐term recovery of the native species, but long‐term monitoring to confirm full population recovery and stability is often not completed.
  2. In this study, long‐term monitoring of native fishes was carried out over 5 years after the eradication of an invasive fish from the Rondegat River, South Africa. Forty‐six sites distributed along four river sections were sampled for presence/absence using underwater cameras and snorkel surveys. Density data were collected by snorkel surveys.
  3. Using multi‐season occupancy models, the annual probability of colonization and local extinction of the native fishes and annual rate of change in occupancy along the river were estimated. Changes in native fish densities across time and across the control and treatment sections were analysed using Kruskal–Wallis analysis, followed by Dunn's post‐hoc test.
  4. Probability of colonization and local extinction differed for each native fish species and may have been affected by extrinsic factors, such as rainfall, and intrinsic density‐dependent factors, hypothesized from the density data. The occupancy rates of change revealed that the two Near Threatened fish species have reached an occupancy dynamic equilibrium but the Endangered fish has not, suggesting that other conservation efforts may be needed.
  5. Long‐term monitoring of native fishes after an eradication programme has confirmed the successful removal of the invader and the recovery and stability of the community. However, successful eradication was not sufficient for full recovery of all species; additional conservation management strategies are needed to secure the population stability and persistence of endangered fishes. We recommend that eradication programmes, regardless of locality, should employ long‐term monitoring to ensure full recovery of a native fish community.
