A low molecular weight protein was separated from urine samples obtained from a heifer with spontaneous renal disease and from cows with CaNa2EDTA-induced renal dysfunction. The molecular weight and electrophoretic mobility of the separated protein were examined. The low molecular weight protein collected by gel filtration chromatography was further separated into two fractions by ion exchange chromatography using DEAE-cellulose. One of the two fractions, the lowest molecular weight protein showed a single band in SDS-PAGE, and its molecular weight was approximately 12,000. An antiserum against this protein formed a single precipitin line with the urine from cows with experimentally induced renal dysfunction and a heifer with spontaneous renal disease by the double immunodiffusion technique. However, the antiserum did not form any precipitin line with the concentrated urine of healthy cow and human beta 2-microglobulin. In cellulose acetate membrane electrophoresis, this protein migrated in the same position as that of serum gamma-globulin from healthy cow. 相似文献

The cause of the low survival rate of sweet pepper plants after grafting was investigated and compared with findings of eggplant and tomato plants, and the promotive effect of ascorbic acid (AA) was determined. Sweet pepper plants formed less callus at the cut surfaces of the stem compared to eggplant and tomato plants. Foliar application with 100 mg L−1 AA promoted callus formation at the cut surfaces of the scion stems and improved the survival rate after grafting. We suggest that the low survival rate of grafted sweet pepper plants is caused by their low rate of callus formation, and that AA can be used to promote graft-take through the acceleration of callus formation at the cut surfaces of the stems. 相似文献

This is the first report to ascertain potassium location in black heartwood of sugi (Cryptomeria japonica). The objective of this study was to understand the distribution of potassium in sugi black heartwood in connection with
anatomical morphology. Scanning electron microscopy with energy dispersive X-ray analysis (SEM-EDXA) was used to investigate
the distribution of inorganic elements, especially potassium. In black heartwood, potassium was detected in all tissues (tracheids,
ray parenchyma, and axial parenchyma), but its concentrations were different in each xylem tissue. Potassium was particularly
accumulated in the droplets in axial parenchyma cells. In addition, calcium was also detected in all tissues. Phosphorus sometimes
existed in the granules in ray parenchyma cells. From optical microscopy, the axial parenchyma cells were observed to be arranged
in a tangential direction on transverse section and the droplets in these cells were distributed in areas of high ash and
green moisture content. For red heartwood, potassium detected by SEM-EDXA was far lower in all tissues than in the black heartwood,
while the trend for calcium was the converse. Potassium in axial parenchyma cells was more abundant in black heartwood than

Effects of melophlins on colony formation of Chinese hamster V79 cells and IL-8 production in PMA-stimulated HL-60 cells

We have recently isolated four new melophlins P (1), Q (2), R (3), and S (4) together with seven known melophlins A (5), D (6), E (7), G (8), H (9), I (10), and O (11) from two marine sponges of the genus Melophlus collected in Palau. In this study, the influence of these compounds on the colony formation of Chinese hamster V79 cells and the production of IL-8 in PMA-stimulated HL-60 cells were examined. These 11 compounds did not show any effect on IL-8 production. The EC50 values of compounds 2, 3, 4, 5, 7, 9, 10, and 11 against V79 cells were 44.0, 13.3, 16.7, 27.2, 19.8, 8.5, 23.1, and 9.6 μM, respectively. The linear-chain-type compounds (1, 6, and 8) were not active against V79 cells at 50 μM. Although the growth inhibitory activity of these melophlins was not remarkable, some structure-activity relationships of these compounds against V79 and murine leukemia L1210 cells were observed. 相似文献

OBJECTIVE: To map central benzodiazepine receptors (BZRs) in the brain of cats by use of positron-emission tomography (PET) and [11C]flumazenil. ANIMALS: 6 male cats that weighed between 2.0 and 3.6 kg. PROCEDURE: Brain images obtained by PET evaluation of [11C]flumazenil were superimposed on T2-weighted magnetic-resonance imaging (MRI) scans of the same cats. Detailed anatomic regions, such as the cerebral cortex, striatum, thalamus, midbrain, and cerebellum, on the PET images were evident by PET-MRI registration. Regional binding of [11C]flumazenil to BZRs was quantitatively measured by use of a model with 2 tissue compartments and 4 variables. RESULTS: The highest value for distribution volume was observed in the cerebral cortex, and the lowest value was found in the midbrain of cats. CONCLUSIONS AND CLINICAL RELEVANCE: Binding of [11C]flumazenil to BZRs in the brain of cats can be quantitatively measured by use of PET with the aid of PET-MRI registration. It is difficult to diagnose changes in these neuroreceptors within the field of current veterinary science. In the future, PET should prove useful for investigating and diagnosing brain disorders in animals in clinical settings. 相似文献

An efficient method of micropropagation was investigated for Ponerorchis graminifolia. Shoot apexes and florets with inflorescence nodes were used as explants for in vitro cultures. A high efficiency of bud formation was observed on half-strength MS medium containing 4.44 μM N6-benzyladenine (BA) and 0.54 μM α-naphthaleneacetic acid (NAA) using florets from upper positions of inflorescences as explants. However, the explants frequently exuded browning compounds into the media during culture. Dark treatment, a countermeasure, decreased exudation of the browning compounds during floret culture. When shoot apexes from dark treated florets were used as an explant material after the floret culture, the dark-pretreated shoot apexes developed 2.2 buds per explant, compared with 1.2 buds per explant from light pretreated shoot apexes, and showed decreased exudation of browning compounds. These results suggest that the most suitable explants for bud formation in P. graminifolia are top florets and shoot apexes derived from the floret culture with dark treatment. 相似文献

Gelatinolytic enzymes were partially purified from the skeletal muscle of red sea bream Pagrus major and characterized to obtain information on post mortem tenderization of fish muscle. Four gelatinolytic activities, G1 (90 kDa),
G2 (65 kDa), G3 (60 kDa), and G4 (100 kDa), were detected in the Q Sepharose column. G1, the major gelatinolytic enzyme, and
G4 were identified as serine proteinases from results of inhibitor spectrum and substrate specificity. By contrast, G2 and
G3 were found to be metalloproteinases since these were inhibited by ethylenediamine tetraacetic acid and o-phenanthroline, and activated by 4-aminophenylmercuric acetate. The optimum pH and temperature of these enzymes were in the

The daggertooth pike conger, Muraenesox cinereus (Forsskål), has become an important fish resource in the western Seto Inland Sea, Japan, since the 1990s. However, introducing sustainable fisheries resource management for this species is difficult in this region because stock assessments have not been performed, and official fisheries statistics for this stock were discontinued after 2007. This study used existing limited data sets to compile the first report for fisheries resource management for this M. cinereus stock. Yield‐per‐recruit analyses showed that increasing fishing pressure above current levels would provide only a minimal increase in expected catch levels. Hence, the current harvest level is considered to represent the upper limit of fishing pressure. Age composition in a given year could potentially be used to forecast landing abundance for the following 2 years. This study provides a basis for establishing effective fisheries resource management strategies for M. cinereus. 相似文献
