Hormonal asynchronies during oestrus, related to the presence of suprabasal plasma-progesterone (P4) concentrations and a delayed ovulation, interfere with the fertility of repeat-breeder heifers (RBH). Since tubal dysfunction can occur in connection with hormonal asynchronies and constrained availability of fertile spermatozoa at the time of ovulation, the present study tested the hypothesis that frequent sperm deposition from onset of oestrus to ovulation may improve pregnancy rates in RBH. Five RBH and five virgin heifers (VH; controls) were repeatedly artificially inseminated (AI) at 6 h intervals from onset of oestrus to spontaneous ovulation. Hormone analyses revealed suprabasal P4 concentrations and a delay in the occurrence of the luteinising hormone (LH) surge, but a normal cortisol profile in RBH. Compared with controls, RBH presented longer interval from onset of oestrus to ovulation, and therefore, received more AIs. Pregnancy rates in RBH reached control levels (60%; NS), indicating that the hypothesis might be correct. Pregnancy rates in VH were below the expected range, presumably attributed to a deleterious influence of the frequent handling. The study suggests that pregnancy rates can be improved in RBH by frequent AI in relation to spontaneous ovulation. However, this practice of repeated manipulations, while seeming not to show adverse effects, lacks practicality for routine use.  相似文献

An established time-resolved immunofluorometric assay designed for measurement of C-reactive protein (CRP) in canine blood was evaluated and validated for use in canine saliva. C-reactive protein was measured in saliva specimens from 5 healthy dogs before and after the injection of casein and in 37 dogs with different disease conditions. The analytical and functional limits of detection were 0.000053 microg/ml and 0.0091 microg/ml, respectively, and intra- and interassay coefficients of variation ranged between 6.7-9.9% and 8.5-16.5%, respectively. A recovery experiment showed no significant disagreement between detected values and expected ones, and saliva CRP concentration was measured in a linear and proportional manner. A positive correlation was found between CRP levels obtained in saliva and serum samples in the experimental (R2 = 0.76) and clinical studies (R2 = 0.70). The assay was able to detect significant differences between salivary CRP levels in healthy dogs and dogs with inflammatory processes. These results suggest that saliva can be used for CRP measurement in dogs. The use of saliva presents the advantage of an easier and less stressful sampling method for the animals, which might be performed outside of hospital environments.  相似文献

Chondrocytes dedifferentiate to a fibroblast‐like phenotype on plastic surfaces. Dedifferentiation is reversible if these cells are then cultured embedded in gels as alginate, agarose or collagen. Chondrocytes cultured in suspension on a non‐adherent surface are also known to form aggregates of differentiated cells. The knowledge of chondrocyte behavior in culture is relevant for tissue engineering purposes. In this report we describe a simple method to culture differentiated or redifferentiated rabbit auricular chondrocytes on plastic surfaces with a stable phenotype. When chondrocyte aggregates formed in suspension are next seeded on plastic surfaces, most of them attach to the plastic as round or polygonal cells, and this morphological differentiation, confirmed by the presence of type II collagen, is stable for long culture periods. We also report that the addition of aggregates to monolayer cultures of dedifferentiated chondrocytes results in their redifferentiation, as is shown by their morphological changes and the synthesis of type II collagen. Therefore, this simple method can be useful for the study of chondrocyte behavior on plastic surfaces and for redifferentiating previously proliferated chondrocytes in tissue engineering techniques. Furthermore, these results demonstrate that, in addition to culture conditions such as cell isolation method or cell‐density, chondrocyte behavior on plastic depends on the presence or absence of aggregates resulting from the dissociation process.  相似文献

Spread of Verticillium wilt into newly established olive orchards in Andalucía, southern Spain, has caused concern in the olive industry in the region. This spread may result from use of Verticillium dahliae-infected planting material, which can extend distribution of the highly virulent, defoliating (D) pathotype of V. dahliae to new areas. In this study, a molecular diagnostic method for the early in planta detection of D V. dahliae was developed, aimed especially at nursery-produced olive plants. For this purpose, new primers for nested PCR were designed by sequencing a 992-bp RAPD marker of the D pathotype. The use of the specific primers and different nested-PCR protocols allowed the detection of V. dahliae pathotype D DNA in infected root and stem tissues of young olive plants. Detection of the pathogen was effective from the very earliest moments following inoculation of olive plants with a V. dahliae pathotype D conidia suspension as well as in inoculated, though symptomless, plants.  相似文献

The aim was to identify biological and physical factors responsible for reducing maize yield in Cameroon. Two surveys were conducted in 137 fields in two agroecological zones in 1995–1997. In the Humid Forest (HF), Bipolaris maydis, Stenocarpella macrospora, Puccinia polysora, Rhizoctonia solani and soil fertility were factors that reduced maize production in 1995 and 1996. In the Western Highlands (WHL), Cercospora zeae-maydis, and the interaction between soil fertility and maize variety were the most important constraints to maize production in 1996. In 1997, C. zeae-maydis, S. macrospora, physiological spot and stem borer damage (Busseola fusca) were negatively related to ear weight. The combination of these biological factors (diseases and insects), and the physical parameter of soil fertility were responsible for reducing maize yield in these selected benchmarks of Cameroon. Maximum potential yield reductions were estimated at 68% due to B. maydis and 46% due to S. macrospora, respectively, in the HF in 1995. In 1996, maximum potential yield reductions in the HF were estimated at 34%, 41% and 30% due to S. macrospora, P. polysora and R. solani, respectively. In the WHL, C. zeae-maydis had the potential to cause a yield reduction of 79% in 1996. In the WHL in 1997, the interaction between C. zeae-maydis and B. fusca, stem diseases and the physiological spot caused potential reductions of 52%, 34% and 39%, respectively.  相似文献

A mathematical model has been developed for the risk assessment of the spread of genes conferring herbicide resistance in plant populations. The model combines an age-and-stage-structured population dynamic model, a population genetic model and a model of spatial spread. This is achieved by embedding a local matrix population model into a cellular automaton model with raster cells as spatial units. The dynamics of each cell is determined by both its local dynamics and the interaction with neighbouring cells. The model is applied to the evaluation of management strategies to delay or even to prevent long-term evolution of resistance in an annual grass weed. The results show that the appearance and spread of resistant genes is a highly non-linear process exhibiting threshold phenomena, which occur for a wide range of parameters. The properties of the seed survival curve constitute the `genetic memory' of the system and thus determine its long-term dynamics. It is possible to delay the evolution of resistance by suspension of treatment, reduction in herbicide application rate and introducing fallow periods. Spatial spread from an infested plot is inhibited by leaving untreated strips between adjacent fields.  相似文献

Biological control of the western flower thrips (WFT)Frankliniella occidentalis, using the entomopathogenicMetarhizium anisopliae-7 (M. a-7) strain was studied in three consecutive seasons under greenhouse conditions. Cucumber plants infested with WFT were sprayed with spore suspension of the fungusM. a-7 (0.5 g m-2), or the soil was treated with dry powder of the fungus (0.5 g m-2); the control was without fungus application. In the 1997 spring experiment, when the cucumber plants were initially infested with only three or four insects per leaf, the spore suspension spray caused a significant reduction in growth of the thrips population compared with the other treatments and the control. However, in the 1997 summer experiment, when the plants were initially heavily infested with WFT (10–15 insects per leaf), the spray treatment caused only a modest reduction in WFT population growth, and only after 4 weeks of treatment was the reduction significant. In the 1999 experiment, with a low initial WFT population of three or four insects per leaf, the spray treatment was effective in reducing the population growth to a lower level than in the other treatments or control. Cocoa is a key or source of income and poverty reduction in the humid forest of Southern Cameroon. Cameroon like other African countries went through a major economic crisis in the early 1980s with a decline in international commodity prices and significant changes in macroeconomic policies. Structural adjustment reforms following the economic crisis led to removal of fertilizers and pesticides subsidies, cocoa price liberalization and the overall withdraw of Government interventions from the cocoa sub-sector. Cocoa input price increases have been compounded by the devaluation of the CFA Franc, which doubled the prices of the imported pesticides which were considered key to the control of cocoa pests. This overall economic shock led to changes in cocoa producer's production decisions as a response to minimize cost. Among the changes the use of alternatives to imported chemicals for cocoa pest control. Farmers responded to the high prices of pesticides by developing, from local botanical knowledge and pest management strategies, which include plant extracts and plant extracts mixed with pesticides at different proportions. This is a major decision given the importance of imported chemical in the cost of production of cocoa. Valuable indigenous knowledge from farmers could be used also as an effective support system for communicating and diffusing modern knowledge and technologies to farmers. The paper describes the farmer knowledge-based alternatives to chemical pesticides for pest control in cocoa fields as a response to high pest control costs. Pest management specialists are urged to take advantage of this shift in practice and assess their effectiveness for further use. Two sets of questions are posed: (1) were the conventional insecticides, with all their problems, really necessary? and (2) are the materials derived from locally grown plants effective pest management agents or are they, in some way, placebos?  相似文献
