Antigenic Detection of Human Strain of Influenza Virus A (H3N2) in Swine Populations at Three Locations in Nigeria and Ghana during the Dry Early Months of 2014 下载免费PDF全文
Since the first detection of human H3N2 influenza virus in Taiwanese pigs in 1970, infection of pigs with wholly human viruses has been known to occur in other parts of the world. These viruses, referred to as human‐like H3N2 viruses, have been known to cause clinical and subclinical infections of swine populations. Due to the paucity and complete unavailability of information on transmission of influenza viruses from other species, especially humans, to swine in Nigeria and Ghana, respectively, this study was designed to investigate the presence and prevalence of a human strain of influenza A (H3N2) in swine populations at three locations in two cities within these two West African countries in January and February, 2014. Using stratified random technique, nasal swab specimens were collected from seventy‐five (75) pigs at two locations in Ibadan, Nigeria and from fifty (50) pigs in Kumasi, Ghana. These specimens were tested directly by a sensitive Quantitative Solid Phase Antigen‐detection Sandwich ELISA using anti‐A/Brisbane/10/2007 haemagglutinin monoclonal antibody. Influenza virus A/Brisbane/10/2007 (H3N2) was detected among pigs at the three study locations, with an aggregate prevalence of 4.0% for the two locations in Ibadan, Nigeria and also 4.0% for Kumasi, Ghana. Transmission of influenza viruses from other species to swine portends serious sinister prospects for genetic reassortment and evolvement of novel viruses. We therefore recommend that further studies should be carried out to investigate the presence of other circulating human and avian influenza viruses in swine populations in West Africa and also determine the extent of genetic reassortment of strains circulating among these pigs. This would provide an early warning system for detection of novel influenza viruses, which could have pandemic potentials. 相似文献

This experiment was conducted to evaluate the effect of administering hormonal treatments (melatonin and progestagen/equine chronic gonadotropin (eCG)) on advancing puberty in Awassi ewe lambs. Fifty-one 6-month-old ewe lambs of similar body weights (around 28 kg) were randomly assigned into four treatment groups; control (CON; n?=?14), melatonin (MEL; n?=?13), melatonin plus progestagen–eCG (MELPP; n?=?11), and progestagen–eCG (PP; n?=?13). Ewe lambs in the PP and MELPP groups were fitted with intravaginal progestagen sponges containing 60 mg medroxyprogesterone acetate for 14 days; 400 IU eCG were administered to each of these ewe lambs on the day of sponge removal. Ewe lambs in the MEL and MELPP groups received subcutaneous melatonin implants (Regulin®, 18 mg melatonin) 36 days before sponge insertion. Hormonal treatment had no effect on ewe lamb body weight change. Estrous behavior was greater (p?0.001) in the PP and MELPP groups than in the CON and MEL ewe lambs. The duration from ram introduction to onset of estrus was shorter (p?0.05) in the CON, PP, and MELPP than in the MEL ewe lambs. The number of ewe lambs showing luteal activity was greater (p?0.001) in the MELPP and PP than in the CON and MEL groups. Lambing rate from mating during the first cycle following ram introduction differed significantly (p?0.01) among treatments being greater in the MELPP and PP than in the CON and MEL groups. Based on these results, it can be concluded that progestagen–eCG appears to be more effective than melatonin in inducing reproductive activity. However, the majority of ewe lambs in the current study either failed to cycle or stopped cycling following the induced estrus. A 5-month-old female Great Dane puppy was treated for hematochezia, tenesmus, and rectal prolapse by resection of a 10-cm-long segment of colon and rectum. Grossly, the colorectal segment had diffuse mucosal and submucosal thickening with multiple polypoid nodules. The histologic diagnosis was colorectal hamartomatous polyps with ganglioneuromatosis. Duplication of PTEN was detected by quantitative multiplex polymerase chain reaction testing. Canine transmissible venereal tumor (CTVT) is a naturally occurring tumor that can be transmitted between dogs via live tumor cell inoculation. It is also a spontaneous self-regression tumor and its behavior is closely related to host immune responses. Since CTVT had been widely used for tumor models in canine cancers, whether this self-regression may overtake the immunity elicited from an exogenous tumor vaccine remains unclear and certainly worthwhile to be investigated. In this study, we used DCs/tumor hybrids as a tumor vaccine to evaluate the CTVT model. We prepared mature allogeneic dendritic cells from bone marrow and then assessed their phenotype (CD80, CD83, CD86, CD1a, CD11c, CD40 and MHC II), antigen uptake and presenting abilities. Fused dendritic cell/CTVT hybrids were then used as a vaccine, administered three times at two-week intervals via subcutaneous injection near the bilateral auxiliary and inguinal lymph nodes. In comparison with unvaccinated dogs (spontaneous regressed group), within a period of 2.5 months, the vaccinations substantially inhibited tumor progression (p<0.05) and accelerated the rate of regression by a mechanism involving amplification of the host tumor-specific adaptive immune responses and NK cytotoxicity (p<0.001). Pathologic examination revealed early massive lymphocyte infiltration resulting in final tumor necrosis. In addition, there are not any detectable effects on routine physical, body temperature or blood chemistry examinations. In conclusion, our data furnishes a reference value showing that CTVT is a model of potential use for the study of immunity elicited by vaccines against tumors, and also enable early-phase evaluation of the dendritic cell/tumor vaccine in terms of raising host immunity. 相似文献

The objectives of this research were to determine the interaction of monensin and haylage supplementation for steers fed 60% dried distillers grains (DDGS) on 1) mineral status, performance, and carcass characteristics, and on 2) ruminal pH, H(2)S, and short-chain fatty acid concentrations. In Exp. 1, Angus-cross steers (n=168; BW=277 ± 67 kg) were blocked by BW and allotted in a 2 × 2 factorial arrangement of treatments to 24 pens. Dietary treatments were 1) 0 mg of monensin/kg of diet + 0% haylage, 2) 33 mg of monensin/kg of diet + 0% haylage, 3) 0 mg of monensin/kg of diet + 10% haylage, and 4) 33 mg of monensin/kg of diet + 10% haylage. The remainder of the diet was 60% DDGS, 10% corn silage, 15% supplement, and corn (either 5 or 15%) on a DM basis. When supplemented with 0 mg of monensin/kg of diet, added haylage increased ADG by 5.7%, whereas when supplemented with 33 mg of monensin/kg of diet, added haylage increased ADG by 13% (P < 0.01). No interactions of monensin and haylage were observed for DMI or G:F (P ≥ 0.36). Haylage inclusion increased (P < 0.01) DMI and decreased (P < 0.01) G:F. No interactions (P > 0.05) on plasma mineral concentrations were observed; however, over time, plasma Cu concentrations decreased (P < 0.01), whereas plasma ceruloplasmin and S concentrations increased (P < 0.01). There were no treatment effects (P ≥ 0.08) on carcass characteristics. Cattle fed the 60% DDGS diets benefitted from increased dietary forage, and the effects of monensin and forage were additive for ADG and final BW. In Exp. 2, ruminally fistulated steers (n=8; BW = 346 ± 34 kg) were used in a replicated 4 × 4 Latin square design and were randomly assigned to the diets used in Exp. 1. Haylage inclusion increased ruminal pH from 1.5 through 12 h postfeeding, and the effects of monensin supplementation were additive (P < 0.05). From 1.5 through 9 h postfeeding, steers fed 33 mg of monensin/kg of diet tended to have reduced (P ≤ 0.10) concentrations of H(2)S when compared with steers fed 0 mg of monensin/kg of diet. Acetate:propionate ratios at 6 h postfeeding were 0.94, 0.93, 1.29, and 1.35 for diets 1 to 4, respectively (P < 0.01); total lactate was decreased regardless of treatment (range: 0.94 to 1.42 μmol/mL). Sulfuric acid in DDGS, not ruminal short-chain fatty acids, may be responsible for the low rumen pH observed and may influence the maximum inclusion of DDGS in cattle diets. Monensin supplementation decreased H(2)S concentration and may decrease the risk of polioencephalomalacia for cattle fed high-DDGS diets. 相似文献

AIM: To test the theory that creating a reservoir of unselected worms by leaving a proportion of lambs in a flock untreated with anthelmintic, i.e. in refugia, will slow the development of anthelmintic resistance in nematode parasite populations. METHODS: Newly weaned Romney lambs (n=180) were infected with two nematode parasite species, Teladorsagia (= Ostertagia) circumcincta and Trichostrongylus colubriformis. For each species, the challenge doses contained a mixture of infective larvae from benzimidazole-resistant and -susceptible isolates calculated to yield, from the combined population, a 95% reduction in faecal nematode egg counts (FEC) following treatment with albendazole. Once the infections were patent, the lambs were divided into nine groups of 20 animals, and each group was allocated to one of three treatments. In Treatments 1, 2 and 3, 100%, 90% and 80% of animals were treated with an anthelmintic, respectively. For treatments 2 and 3, the heaviest animals remained untreated. Following treatment, each group was moved to its own previously prepared low-contamination pasture. Lambs grazed this pasture for 7 weeks before again being treated and moved to new low-contamination pastures (Shift 1 and Shift 2). The parasite populations on pasture resulting from the different treatments were subsequently sampled using tracer lambs, and worm eggs derived from these were used in both egg-hatch assays (EHA) and larval development assays (LDA), to measure albendazole-resistance status. RESULTS: Treating all animals each time the groups were moved to new low-contamination pastures resulted in higher levels of albendazole resistance (p<0.05), measured using EHA and LDA, in subsequent parasite generations than when either 10 or 20% of animals were left untreated. However, higher FEC in the tracer lambs grazed on pastures in Treatments 2 and 3, compared with Treatment 1, indicated an increased level of pasture contamination as a result of leaving some animals untreated. CONCLUSIONS: The results demonstrate that creating a reservoir of unselected parasites slows the development of anthelmintic resistance, and emphasises the risk of treating all animals prior to a shift on to low-contamination pasture. However, higher levels of pasture contamination, resulting from untreated animals, indicate the difficulty in managing both worm control and resistance. 相似文献

A 2-year-old, spayed female domestic shorthair cat was referred with a history of anorexia and depression of 1 week duration. On physical examination, the cat was lethargic and febrile, with splenomegaly, anisocoria and ulcerative stomatitis. A complete blood count (CBC) and a biochemistry profile showed leukocytosis, numerous blast cells in the peripheral blood, thrombocytopenia, hyperglobulinaemia and a positive test for feline leukaemia virus antigen. A diagnosis of acute myelomonocytic leukaemia was made on the basis of the results of bone marrow cytology, histopathology, and immunochemistry (CD3, CD79a, lysozyme, and myeloperoxidase) tests.
