Gamma tocopherol (gT) exhibits beneficial cardiovascular effects partly due to its anti-inflammatory activity. Important sources of gT are vegetable oils. However, little is known to what extent gT can be transferred into marine animal species such as Atlantic salmon by feeding. Therefore, in this study we have investigated the transfer of dietary gT into salmon. To this end, fish were fed a diet supplemented with 170 ppm gT for 16 weeks whereby alpha tocopherol levels were adjusted to 190 ppm in this and the control diet. Feeding gT-rich diets resulted in a three-fold increase in gT concentrations in the liver and fillet compared to non-gT-supplemented controls. Tissue alpha tocopherol levels were not decreased indicating no antagonistic interaction between gamma- and alpha tocopherol in salmon. The concentration of total omega 3 fatty acids slightly increased in response to dietary gT. Furthermore, dietary gT significantly decreased malondialdehyde in the fillet, determined as a biomarker of lipid peroxidation. In the liver of gT fed salmon we observed an overall down-regulation of genes involved in lipid homeostasis. Additionally, gT improved the antioxidant capacity by up-regulating Gpx4a gene expression in the pyloric caeca. We suggest that Atlantic salmon may provide a marine functional source capable of enriching gT for human consumption.  相似文献

A screening of endophytic fungi isolated from Mediterranean plants rendered a mycelium-bound lipase from a strain of Rhizopus oryzae that catalyzed the esterification of fatty acids in isooctane. The influence of various factors (water content, temperature, and pH) on ester synthesis was investigated. Catalytic activity was inversely correlated with water content. This enzyme was active over the entire pH range studied, from pH 3 to pH 8, and activity was maximal at pH 4 and pH 7. The enzyme was thermostable, with maximal activity at 60 degrees C.  相似文献

Immunoassays for 2,4,6-trichloroanisol (TCA) have been evaluated. The assays were developed after raising antibodies against three different immunizing haptens (1). Lack of reproducibility has been one of the main problems of these assays. Precision was worse on these assays, reaching lower limits of detection. The high lipophilicity of TCA and its, consequently, low water solubility have been found to be the major cause of this problem. A reliable microplate-based enzyme-linked immunosorbent assay (ELISA) has been set after consideration of the TCA physicochemical features and evaluation of important parameters affecting immunoassay performance. The immunoassay uses As78 (developed against hapten B-KLH) and C9-OVA as the coating antigen. The selectivity is high although the brominated analogue 2,4,6-TBA is also recognized. In buffered media containing 7% ethanol, the resulting assay shows a good accuracy with an IC(50) value of 0.53 microgram L(-)(1) and a limit of detection of 0.044 microgram L(-)(1). Red and white wine samples caused important interferences in the immunoassay demonstrating the necessity of a cleanup procedure prior to the ELISA.  相似文献

Cynodon dactylon (L.) Pers. is a perennial rhizomatous grass (Poaceae), grown for cattle nutrition on the riverbanks of Río Tinto (Southwest Iberian Peninsula, Spain), a highly acidic area with high concentrations of iron (Fe) and other metals. This study focuses on the absorption, distribution, and accumulation of Fe in the root, rhizome, and leaves of C. dactylon under controlled conditions. Plants collected from Río Tinto were grown in a Hoagland solution containing 500 mg kg–1 of ferrous Fe. Samples were collected up to 2 months after exposure and analyzed for total Fe concentration using inductively coupled plasma–mass spectrometry (ICP‐MS) and for Fe distribution and bioformations by scanning electron microscopy (SEM) with an energy‐dispersive x‐ray analyzer (EDX). The results show high concentrations of Fe in all plant organs, with fast Fe translocation from roots to leaves. Iron bioformations composed mainly of Fe, S, and K were detected in all plant organs and were especially apparent in roots and leaves. These results differ from those reported for another species of Poaceae, Imperata cylindrica, which grows under the same environmental conditions, suggesting the existence of different resistance strategies between species of the same family.  相似文献

The low field region (5.8-9.0 ppm) corresponding to aromatic protons and the region 1.8-3.0 ppm of the (1)H NMR spectra were used for characterization and chemometric differentiation of 52 apple juices obtained from six cider apple varieties. The data set consisted of 14 integrated areas corresponding to resonances from acids and phenolic compounds. Multivariate procedures based on hierarchical cluster and discriminant analysis were performed on selected signals of the spectra to determine whether it was possible to distinguish the different juices. Cluster analysis was able to satisfactorily classify the six apple varieties. Discriminant analysis, by means of stepwise procedure for variables selection and leave-one-out for cross-validation, was applied to 40 samples from the year 2001, obtaining recognition and prediction abilities of 100%. The most discriminant variables corresponded to poliphenols, (-)-epicatechin, phloridzin-phloretin, and p-coumaric, chlorogenic, and malic acids. The classification model was applied to 12 samples from apples harvested in the years 2002 and 2003, and the prediction ability was 91.7%.  相似文献

The Fabaceae (legume family) is one of the largest families of plants with a worldwide distribution and a major role in agriculture and in agroforestry. A hard seed coat impermeable to water is a typical feature of several species. Physical dormancy delays and reduces germination so that mechanical, physical and chemical scarification methods have been classically used to break seed dormancy of many species. We evaluate the effectiveness of a methodology to scarify seeds of several woody Fabaceae of ecological and economical importance, including Robinia pseudoacacia and Acacia dealbata and the shrubs Cytisus scoparius, C. multiflorus and Ulex europaeus. We describe the optimized use of a handheld rotary tool (HRT), and compare its effectiveness with other scarification methods reported to break dormancy such as boiling or dry heating. Total germination and/or speed of germination were enhanced after the application of the HRT, with germination percentages significantly higher than those achieved by other methods of scarification. Based on a thorough literature review, a mode of action for the HRT is suggested which could operate by breaking the physical and physiological dormancy of treated seeds through the combined action of coat abrasion and moderate temperatures. Considering these results, we recommend the application of this rapid, effective, low-cost and highly reproducible HRT method to break seed dormancy and enhance germination of these species and others with similar dormancy constraints.  相似文献

Wholegrain consumption is associated with several health benefits, in contrast to the consumption of refined grains. This can partly be related to the antioxidant compounds in the outer parts of the grain kernel. The bioaccessibility of these antioxidant compounds from the wholegrain matrix during gastrointestinal digestion is crucial for their absorption and bioavailability. In the current study, the bioaccessible compounds from aleurone, bran and flour were obtained from a dynamic in vitro model of the upper gastrointestinal tract. They were collected at 1 h time intervals to assess their antioxidant capacity (TEAC assay) and also their anti-inflammatory effect (TNF-α reduction in U937 macrophages stimulated with LPS). The bioaccessible compounds from aleurone had the highest antioxidant capacity and provided a prolonged anti-inflammatory effect, shown by the TNF-α reduction of a relatively late time-interval (3–4 h after start of digestion). The contribution of ferulic acid to those effects was minor due to its low bioaccessibility. Aleurone seems a promising wheat fraction for cereal products with a healthy added value.  相似文献

Metacaspases are distant relatives of animal caspases found in protozoa, fungi, and plants. Limited experimental data exist defining their function(s), despite their discovery by homology modeling a decade ago. We demonstrated that two type I metacaspases, AtMC1 and AtMC2, antagonistically control programmed cell death in Arabidopsis. AtMC1 is a positive regulator of cell death and requires conserved caspase-like putative catalytic residues for its function. AtMC2 negatively regulates cell death. This function is independent of the putative catalytic residues. Manipulation of the Arabidopsis type I metacaspase regulatory module can nearly eliminate the hypersensitive cell death response (HR) activated by plant intracellular immune receptors. This does not lead to enhanced pathogen proliferation, decoupling HR from restriction of pathogen growth.  相似文献
