OBJECTIVE: To determine the effect of transportation stress on serum concentrations of oxidative stress biomarkers of calves. ANIMALS: 105 crossbred beef steer calves (mean [+/-SD] body weight, 207 +/- 21.2 kg). PROCEDURE: Calves were assembled at 1 location in Tennessee, and pretransit (day -3) blood samples were collected. Calves were allotted randomly by body weight into 2 groups. Calves were transported 1,930 miles to a feedlot in Texas, and 1 group received tilmicosin phosphate (33 microg/kg, s.c.) upon arrival. Calves were weighed and blood samples collected on the day of arrival (day 1) and on days 15, 22, and 28. Calves were scored daily for signs of bovine respiratory disease (BRD). Serum total antioxidant capacity (TACA) and serum malondialdehyde (MDA) concentrations were determined. RESULTS: Transportation stress significantly decreased mean serum TACA concentrations (from 147 +/- 31.2 U/mL to 133 +/- 20.1 U/mL) and significantly increased serum MDA concentrations (from 10.9 +/- 18.3 microg/mL to 30.2 +/- 50.5 microg/mL). Calves that died had a 43% increase in serum MDA concentration on day 1, compared with calves that lived (42.2 +/- 67.0 microg/mL vs 29.4 +/- 49.4 microg/mL, respectively). Calves that had > or =3 episodes of BRD had 2-fold higher serum MDA concentrations on day 1 than healthy calves. Tilmicosin-treated calves had a 20.8% significantly greater average daily gain and significantly greater serum TACA concentration than nontreated calves on day 28. CONCLUSIONS AND CLINICAL RELEVANCE: Transportation stress increases serum concentrations of oxidative stress biomarkers that are related to episodes of BRD and mortality in calves.  相似文献

The aim of this study was to investigate the prevalence of antibodies against zoonotic agents in employees of the zoological garden of Vienna, Sch?nbrunn, Austria. Sixty out of 120 employees participated in the study. In 97% of them antibodies to at least one zoonotic agent were identified. Only two participants were free of antibodies to the zoonotic agents tested. The following seroprevalences (in brackets) were obtained: Viral zoonotic (and potentially zoonotic) agents: Influenzavirus A/H1N1 (58%), Influenzavirus A/H3N2 (85%), Lymphocytic choriomeningitis virus (13%), Encephalomyocarditis virus (5%), Orthopox- (Cowpox-) virus and Hantavirus type Puumala (3%). Hantavirus type Hantaan and Borna disease virus (all negative). Bacterial zoonotic agents: Bartonella henselae (65 %), Borrelia burgdorferi (10%), Leptospira interrogans serovar copenhageni and serovar icterohaemorrhagiae as well as Chlamydophila psittoci (2% each). Brucella spp., Coxiella bumetii, and Francisella tularensis (all negative). Parasitic zoonotic agents: Toxoplasma gondii (53%), Toxocara spp. (21%), Capillaria hepatica (2%), Fasciola hepatica, Schistosoma mansoni, E. multilocularis, and E. granulosus (all negative). The remarkably high seroprevalence to the causative agent of cat scratch disease, Bartonella henselae, is probably due to the private contact of the employees to cats. Regarding viral zoonotic agents it has to be mentioned that Influenzavirus vaccination and/or human-to-human transmission of especially A/H3N2 Influenzaviruses probably attributed significantly to the very high seroprevalence to both Influenzavirus types A/H1N1 and A/H3N2. When investigating parasitic zoonotic agents, high prevalence rates were found against Toxoplasma gondii and Toxocara spp., however, it was not possible to establish a causal link between seropositivity and the professional activity in the zoo. Interestingly, in the case of antibodies to T. gondii, the typical correlation with age was not found in this study, while in the case of the Toxocara spp. positive subjects a correlation was identified with both age and duration of employment in the zoo. Regarding the later two zoonotic parasites, employees of the zoological garden showed significantly higher seroprevalences than the average Austrian population. Antibodies to Capillaria hepatica, a hepatic-parasite in rodents which is diagnosed in humans rarely, were identified in one employee and another one showed a questionable positive result. Further investigations did not exhibit clinical infestations with the parasite in these two individuals so far.  相似文献

The aim of this study was to investigate the seroprevalences to zoonotic pathogens in hunters, to propose preventive measures and to obtain more information about the occurrence of zoonotic pathogens in local wild animal populations. From 146 male and 3 female hunters originating from the south-eastern Austrian federal states of Styria and Burgenland blood samples were taken and anamnestic data were obtained using a questionnaire. The serological investigations included the following viral, bacterial and parasitic zoonotic agents or zoonoses, respectively (antibody-seroprevalences in brackets): encephalomyocarditis virus (EMCV, 15%), Puumala-Hantavirus (10%), Newcastle Disease virus (NDV, 4%), borreliosis (IgG 42%, IgM 7%), brucellosis (1%), chlamydiosis (3%), ehrlichiosis (IgG 15%, IgM 3%), leptospirosis (10%), tularaemia (3%), Q fever (0%), Echinococcus multilocularis/E. granulosus (5%/11%), toxocariasis (17%). Out of a control group of 50 persons (urban population, no hunters) only one person was found to be seropositive for Toxocara canis and NDV and four for EMCV, all other results were negative in the control group. The high seroprevalences especially to Borrelia burgdorferi s.l., Ehrlichia spp., Leptospira interrogans, E. granulosus, E. multilocularis, encephalomyocarditis virus and Puumala virus demonstrate that hunters are particularly exposed to zoonotic pathogens. It should also be noted that one hunter was seropositive for Brucella abortus and five exhibited antibodies to Francisella tularensis. In these cases, as well as in the cases of the 15 seropositives for Leptospira interrogans, the suspected source of infection may--besides rodents--also include wild boars and brown hares. The infections with NDV and Chlamydophila psittaci may be traced back to contact with certain species of birds (potential risk: aviaries). For Hantaviruses, rodents are considered to be the main source of human infections.  相似文献

OBJECTIVE: To evaluate energy expenditure (EE) in dogs by estimating rate of CO2 production (rCO2). ANIMALS: 15 Beagles. PROCEDURE: Food was withheld for 24 hours, and all dogs received an IV infusion of 13C sodium bicarbonate for 8 hours. Breath samples were collected before infusion and at 30-minute intervals from 4 to 8 hours, and 13C enrichment in breath CO2 was measured, using gas chromatography-isotopic ratio mass spectrometry. Food was withheld from 6 dogs, and rCO2 and O2 consumption were measured, using a conventional indirect calorimeter. The CO2 production and O2 consumption were measured by use of indirect calorimetry in 6 other fed dogs that were injected with 2H2O and H2(18)O. Blood samples were collected before tracer injection, 4 hours later, and on days 4, 7, and 11. Deuterium and 18O enrichments in plasma water were determined. RESULTS: Mean rCO2 measured by indirect calorimetry was 516 +/- 34 and 410 +/- 16 micromol/kg(0.75)/min in 6 fed and 6 food-deprived dogs, respectively. The rCO2 calculated from 13C-bicarbonate dilution was 482 +/- 30 micromol/kg(0.75)/min. Mean rCO2 determined by use of the double-labeled water method was 1,036 +/- 46 mmol/kg(0.75)/d. Mean energy expenditure calculated from rCO2 determined by infusion of 13C bicarbonate, indirect calorimetry in fed and food-deprived dogs, and infusion of double-labeled water was 386 +/- 39, 379 +/- 25, 338 +/- 14, and 552 +/- 25 kJ/kg(0.75)/d, respectively. CONCLUSIONS AND CLINICAL RELEVANCE: Energy expenditure calculated by indirect calorimetry in unfed dogs can be considered representative of basal metabolic rate.  相似文献

To simulate a future ion input reduction scenario in forests, a large scale field experiment was set up in a (1999) 66‒year‒old Norway spruce plantation at Solling, central Germany. Throughfall input of H+, SO42—, and N‒compounds is artificially reduced by means of a permanent roof construction below the canopy and a de‒ionizing equipment since 1991. Here we present long term soil solution records for SO42—, NO3, Al3+ and the pH of the 10 cm mineral soil sampling depth. A significant decrease in ion concentrations since the start of the treatment is observed, but no change of the soil solution pH. Even in the fourth year pH values remained well within the aluminium buffer range (pH < 4.2). Three years after the start of the experiment (July 1994) it was examined whether microbial biomass (Cmic), specific activity (heat production per unit biomass), and the percentage of Cmic in organic C material indicated any changes. Furthermore chemical standard parameters (CEC, base saturation, pH) were analyzed for all soil samples. Results indicate that despite of drastic decreases of soil solution ion concentrations in the upper soil horizons microbial parameters were not affected and that the soil solid phase is not deacidified by the treatment until now.  相似文献

Sole ammonium supply provokes negative effects on dry‐mass formation, leaf growth, and water uptake of ammonium‐sensitive plants. To study the effects of N form on nutrient and water uptake and aquaporin expression, French bean plants were grown in a split‐root system. Five treatments were compared: homogeneous nitrate (NN) and ammonium (AA) supply; spatially separated supply of nitrate and ammonium (NA); and half of the root system supplied with N‐free nutrient solution, the other half with either nitrate (N0) or ammonium (A0). Ten days after onset of treatments, root dry mass (DM) and water‐uptake rate (WUR) were significantly reduced under ammonium compared to nitrate supply. WUR from nitrate‐supplied vessels was 80% higher than that from N‐free nutrient solution, while WUR from N‐free nutrient solution was 130% higher than that from ammonium‐supplied vessels. Potassium uptake was lower under ammonium supply and the ratio of N : K uptake of treatment AA was significantly higher compared to others. High K uptake from N‐free nutrient solution of A0 plants resulted in a ratio of N : K uptake comparable to nitrate‐supplied plants, but shoot growth resembled that to plants under sole ammonium supply. Within 24 h after onset of treatments, expression of aquaporin was lower under ammonium compared to nitrate supply. From these data, it can be concluded that reduced root water transport under ammonium supply is directly related to aquaporin activity.  相似文献

The riparian forests along braided rivers are dynamic, frequently rejuvenated by floods and channel changes, and thus dominated by pioneer to middle stages of succession; they are sites of high biodiversity in some regions. The Lower Eygues River (drainage area 1150 km2 in southeastern France) is such a braided river system with large alluvial forests dominated by Salix alba, Populus alba, and P. nigra. It was identified as a site of ecological interest by the EU under the Natura 2000 program. Such forests elsewhere in Europe have been identified as reference ecosystems. We documented the historical evolution of this alluvial forest from detailed (1:2500 scale) early 19th C parcel maps, early 20th C topographic maps, aerial photography from 1947 to 1996, and field surveys of topography and riparian vegetation in 1997–1998. Our results show that in 1830, the channel was wide, aggraded, and agricultural pressure extended literally to the channel edge. With decline in the rural population and reduced agricultural and grazing pressure in the catchment, erosion rates declined. Reduced sediment supply led to channel narrowing and incision. This channel narrowing, coupled with reduced agricultural pressure along the banks, has allowed riparian forest to colonize former active channel areas, especially within late 19th-century 20th century flood dykes. In recent decades, aggregate mining, and clearing for recreation and agriculture have fragmented the forest. Thus, the alluvial forest of the Lower Eygues is largely an artifact of changing human land-use over the past century, a context that should frame efforts for preservation and restoration.  相似文献

The loss of aldicarb sulphoxide was studied in incubation experiments with soil from four plough layers and two deeper layers. The loss during the 111 days of the experiment could be described by first-order kinetics. The half-lives at 15°C ranged from 20 days in a clay loam to 46 days in a peaty sand. The loss of sulphoxide in deeper layers was considerably slower than in the corresponding top layers of a soil profile. In soil from a silty layer at 70–90 cm depth the half-life was about 53 days. In soil from a sand layer at 90–110 cm depth a loss of only about 15% was measured after 111 days of incubation. First-order rate constants for sulphoxide conversion in a clay loam at 6, 15 and 25°C were found to be 0.009, 0.033, and 0.05 day?1 respectively; in a greenhouse soil these rate constants were 0.0052, 0.019 and 0.04 day?1 respectively. The fractions of aldicarb sulphoxide that were oxidised to sulphone at 15°C in soil from plough layers were computed to range from 0.52 to 0.76.  相似文献
