ABSTRACT: The clonal triploid ginbuna crucian carp Carassius auratus langsdorfii , a naturally occurring gynogenetic fish, is a useful model for studying T-cell-mediated immunity. CD4, a T-cell receptor (TCR) coreceptor, is a membrane-bound glycoprotein found on helper T-cells, and assists in the binding of major histocompatibility complexes. In the present study, full-length cDNAs encoding the CD4 molecule from the S3n strain of ginbuna crucian carp were cloned and characterized. 5'-rapid amplification of cDNA ends (RACE) and 3'-RACE yielded two distinct cDNA clones of CD4 homolog from the ginbuna, and these sequences share 95% identity at the amino acid level. These ginbuna CD4 molecules consisted of a signal peptide, immunoglobulin superfamily (IgSf) like domains, a transmembrane domain, and a cytoplasmic domain similar to other known CD4. A tyrosine protein kinase p56lck binding motif is conserved in the cytoplasmic tail of ginbuna CD4. Phylogenetic tree analysis indicated that ginbuna CD4 sequences are closely related to CD4L-1 from other fish species. Expression of ginbuna CD4 mRNA was detected in the gill, thymus, head kidney, trunk kidney and peripheral blood leukocytes, indicating that its expression pattern is similar to that of ginbuna TCRβ mRNA. We improved the dry ash procedure for detecting titanium dioxide (TiO2) in cattle feces containing chromium dioxide (Cr2O3). First, the effect of amount of sodium sulfate (Na2SO4) on the recovery of TiO2 from cattle feces that contained Cr2O3 was evaluated. Average recovery of TiO2 at the 2.5 g Na2SO4 level was significantly higher (P < 0.0001) than that at 0.75 g Na2SO4. Second, the effect of Cr2O3 concentration on the recovery of TiO2 of cattle feces by using two levels of Na2SO4 addition was examined. The recovery of TiO2 decreased with the increase in the amount of Cr2O3 at the 0.75 g Na2SO4 level but was consistently high at 2.5 g Na2SO4. Third, the recovery of Cr2O3 from cattle feces was checked. The recoveries of TiO2 and Cr2O3 were high enough at the 2.5 g Na2SO4 level. Fourth, the improved dry ash procedure (5 mL of concentrated H2SO4 and 2.5 g of Na2SO4 were used for sample digestion) was compared to the wet ash procedure. Average recovery of TiO2 by the improved dry ash procedure was significantly higher (P = 0.0077) than that by the wet ash procedure. A series of studies were carried out to determine the effects of increasing environmental temperature (20 to 28 to 33 °C) at a constant (60%) relative humidity on the physiological responses, energy and nitrogen balance of prepubertal Holstein heifers (n = 4, initial body weight = 198 ± 3 kg). The relatively high environmental temperatures of 28 and 33 °C increased rectal temperature by 0.2 and 1.2 °C and increased respiration by 23 and 58 breaths per min, respectively. At 28 °C, dry matter intake was similar to that at 20 °C, whereas at 33 °C dry matter intake was lower (P < 0.10) by about 9%. However, the dry matter digestibility was higher (P < 0.05) at 33 °C than at 20 °C, hence the thermal treatments did not affect daily weight gain. At 33 °C blood urea nitrogen increased whereas hematocrit and plasma glucose concentrations decreased. At 33 °C energy and nitrogen intake were lower (P < 0.10), while energy and nitrogen digestibility were higher at 33 °C than at 20 °C. Urinary nitrogen was elevated at 33 °C resulting in lower nitrogen retention. Although, energy retention as fat did not differ (P > 0.10) among the treatments, energy retention as protein was lower (P < 0.05) at 33 °C than at 20 °C or 28 °C. We investigated the impact of land-use changes on the soil biomass at several soil sites in Indonesia under different types of land-use (primary forest, secondary forest, coffee plantation, traditional orchard, and deforested area), located within a small geographical area with similar parent material and climatic conditions. Various parameters of soil microbial biomass (biomass C, biomass N, content of anthrone-reactive carbohydrate carbon, and soil ergosterol content) were examined. Our results suggested that the removal of the natural plant cover did not cause any appreciable decrease in the amount of microbial biomass; on the contrary it led to a short-time increase in the amount of microbial biomass which may be due to the availability of readily decomposable dead roots and higher sensitivity to the decomposition of residual litter in recently deforested soils. However, the amount of microbial biomass tended to decrease in proportion to the duration of the land history in coffee plantation soils. This may be ascribed to the effect of the loss of available substrates associated with soil erosion in the long term. Lower ergosterol contents in recently deforested areas reflected a reduction in the amount of fungal biomass which may be due to the destruction of the hyphal network by the slash and burn practice. On the other hand, the higher soil ergosterol content at the sites under bush regrowth indicated that microbial biomass was able to recover rapidly with the occurrence of a new plant cover. 相似文献

SEM observations on larval shell morphology of Manila clam Ruditapes philippinarum and their utility

Manila clam Ruditapes philippinarum is common and abundant on Japanese tidal flats, forming a commercially important clam fishery. However, annual catches of Manila clam have decreased drastically since 1975?C1985. To study larval recruitment processes of Manila clam, we carried out scanning electron microscopy (SEM) observations on larval and juvenile shells of clams reared at 20 and 24?°C. There was no significant difference in final shell length of trochophores between 20 and 24?°C. However, the larval duration was much longer and the shell length of settled size of pediveligers was much larger for clams reared at 20?°C than those reared at 24?°C. These findings suggest that larval duration and growth, as well as settlement size, may vary markedly depending on temperature (and probably on season). The larval shell morphology of Manila clam can provide essential information about larval recruitment processes. 相似文献

The effects of intraperitoneal implantation of Taenia taeniaeformis larvae and inoculation of in vitro larval products on gastric mucosa of SCID mice were investigated in this study. Mice surgically implanted with T. taeniaeformis larvae developed slight and moderate gastric hyperplasia. When in vitro cultured T. taeniaeformis larval excretory-secretory (TtLES) products containing 1 mg of protein were injected daily into mice, they caused gastropathy after 5-7 days. Mice injected daily with 0.5 mg of TtLES products also showed slight gastric hyperplasia after day 14 and 28. The gastropathy was characterized by reduction of both parietal and zymogenic cell number and increased number of alcian blue-periodic acid Schiff (AB-PAS)-positive cells and by two-fold extension of proliferative zone of gastric units. Larval implantation demonstrated a more potent effect in inducing gastropathy than did in vitro larval culture products. Significant decrease in number of parietal cells with concomitant increase of proliferative zone and AB-PAS-positive cell number indicated their important roles in inducing the hyperplastic lesion. Similarities with other gastropathies indicated that there is a common fundamental regulatory mechanism involved, and that the host response may not be specific to parasites. Present study validated the induction of gastric mucosal hyperplasia by larval ES products of T. taeniaeformis. This proved the hypothesis of previous studies suggesting the role of larvae-derived products in inducing gastric mucosal hyperplasia in T. taeniaeformis-infected rats. 相似文献
