Pink rot of potato, most commonly caused by Phytophthora erythroseptica , is a major field and post-harvest problem in southern Idaho, USA, particularly since 1998 when isolates resistant to the phenylamide fungicide metalaxyl-M (mefenoxam) were detected. Isolates of P. erythroseptica were collected from infected tubers in 2001 and 2002 from six Idaho counties and tested for resistance to metalaxyl-M on amended agar. Metalaxyl-M resistant (MR) and metalaxyl-M-sensitive (MS) isolates were identified in six counties; 160 isolates were highly resistant, seven moderately resistant and 57 sensitive to metalaxyl-M with mean EC50 values of 182, 23 and 0·5 mg L−1 ai metalaxyl-M, respectively. Mycelial growth rates and oospore production in agar were assessed for 20 MS and 20 MR isolates at 10, 15, 20, 25 and 30°C. Growth rates of MR isolates were between 2·5 and 3·1 times greater ( P < 0·05) than those of MS isolates at 10, 15, 20 and 25°C, and oospore production was between 6·8 and 20·5 times greater ( P < 0·0001) for MR than for MS isolates at the same temperatures. Colony growth in V8 broth at 18°C was greater for MR than MS isolates ( P < 0·0032). However, zoospore production at 18°C was greater for MS than for MR isolates ( P < 0·0109), and zoospore production m m −1 of colony circumference was also greater for MS than for MR isolates, 14 191 and 9959, respectively ( P = 0·0109). Sexual reproduction of MR isolates in nature may be greater than MS isolates, but MS isolates may be more asexually fit based on the fitness parameters studied. 相似文献

An important feature of maintaining the agricultural stability in millennia-old mountain oases of northern Oman is the temporary abandonment of terraces. To analyse the effects of a fallow period on soil microbial performance, i.e. microbial activity and microbial biomass, samples of eight terrace soils abandoned for different periods were collected in situ, assigned to four fallow age classes and incubated for 30 days in the laboratory after rewetting. The younger fallow age classes of 1 and 5 years were based on the records of the farmers recollections, the two older fallow age classes of 10–20 and 25–60 years according to the increase in the d -to- l
ratio of valine and leucine enantiomers. The increase in these two ratios was in agreement with that of the d
-to- l ratio of lysine. The strongest relationship was observed between the increase in the d -to- l
ratio of lysine and the decrease in soil microbial biomass C. However, the most stringent coherence between the increase in fallow age and soil properties was revealed by the decreases in cumulative respiration and net N mineralisation rates with decreasing availability of substrate to soil microorganisms. During the 30-day incubation following rewetting, relative changes in microbial activity (respiration and net N mineralisation) and microbial biomass (C and N) indices were similar in the eight terrace soils on a fallow age-class-specific level, indicating that the same basic processes occurred in all of the sandy terrace soils investigated. 相似文献

Influence of the VA-Mycorrhiza on P uptake and recovery potential of corn (Zea mays L.) under water stress conditions In a seven week pot experiment the influence of the VA-Mycorrhiza on P uptake and water stress tolerance of corn was investigated. During the eight day water stress period the VAM-plants took up less phosphorus than the control plants. However, during the successive five day recovery period under a normal water regime the VAM-plants showed an increased root growth and an enhanced phosphorus uptake compared with the control plants. The free prolin concentration was lower in leaves of VAM-plants than in those of nonmycorrhizal plants at the end of the water stress period, pointing to a different water stress tolerance. 相似文献

The degenerative wear and pathologic damage of the joints are reasons for total endoprotheses in man as well as in dogs. The main problem is the aseptic loosening of the protheses. By usig the finite-element-method, the total endoprothesis is designed with new features, with the purpose of preventing loosening and being better adapted to load transmission. In order to simulate the femur of the dog for the numerical analysis, a material law is developed. By taking into account the anisotropy and the local density of the cancellous bone in the femoral head, the young's modules are experimentally determined. Recognition and clearance of a bacterial infection are a fundamental properties of innate immunity. Here, we describe an effector B cell population that protects against microbial sepsis. Innate response activator (IRA) B cells are phenotypically and functionally distinct, develop and diverge from B1a B cells, depend on pattern-recognition receptors, and produce granulocyte-macrophage colony-stimulating factor. Specific deletion of IRA B cell activity impairs bacterial clearance, elicits a cytokine storm, and precipitates septic shock. These observations enrich our understanding of innate immunity, position IRA B cells as gatekeepers of bacterial infection, and identify new treatment avenues for infectious diseases. 相似文献

Despite the fact that docking of dogs has been banned in Germany for years, many animals are still docked, mainly owing to overcome traditional thinking. This is possible by some exceptions made by the law: the import of dogs that have been docked abroad, and medical indications for docking. In order to substantiate the necessity of such medical indications, the patient material of the Small Animal Clinic of the Veterinary School Hanover was analysed and compared with a number of veterinary certificates for docked dogs that were to be shown at exhibitions, stating a medical indication. It shows that the latter predominantly cover dogs belonging to breeds that are traditionally docked, the Doberman in the first line. Contrasted with this, dogs from our own patient material showed an even breed distribution, with no Doberman at all within a five year period. Moreover, the fact that certain breeds were predominantly docked by the same veterinarian as well as the medical reasons stated were suspicious. Consequently, doubts should be raised in cases of accumulations of tail amputations in dogs belonging to a traditionally docked breed, when the reasons given are the same in several animals, and when the same veterinarian performs the operation in several dogs belonging to the same breeder. Because of the scientifically proven pain that is caused to the animal both at the time of the procedure and later, as well as the possibility of long lasting chronic health impairment owing to docking, it is necessary to consequently fight this malpractice. The complete genome or the genome region containing the two fiber genes of two reference strains and one field isolate representing both serotypes of Fowl adenovirus C were sequenced. Two fiber genes were revealed in the genomes of all three isolates. Fiber-1 and fiber-2 genes of several Fowl adenovirus C isolates were sequenced as well. Both serotypes 4 and 10 have two fiber genes. The genome region containing the fiber gene was also sequenced for the reference strain of Fowl adenovirus B. Just one fiber gene was revealed in this strain. Predicted amino acid sequences were compared to already published fiber sequences of different adenovirus isolates and one amino acid substitution within fiber-2 was detected in all of the Fowl adenovirus C isolates that were isolated from chickens with hepatitis-hydropericardium syndrome in comparison to apathogenic isolates. Phylogenetic analyses provided insights about the evolution of fiber genes in avian adenoviruses and their genetic relationships. 相似文献

Soil texture is one of the main factors controlling soil organic carbon (SOC) storage. Accurate soil‐texture analysis is costly and time‐consuming. Therefore, the clay content is frequently not determined within the scope of regional and plot‐scale studies with high sample numbers. Yet it is well known that the clay content strongly affects soil water content. The objective of our study was to evaluate if the clay content can be estimated by a simple and fast measure like the water content of air‐dried soil. The soil samples used for this study originated from four different European regions (Hainich‐Dün, Germany; Schwäbische Alb, Germany; Hesse, France; Bugac, Hungary) and were collected from topsoils and subsoils in forests, grasslands, and croplands. Clay content, water content of air‐dried soil, and SOC content were measured. Clay content was determined either by the Pipette method or by the Sedigraph method. The water content of air‐dried soil samples ranged from 2.8 g kg–1 to 63.3 g kg–1 and the corresponding clay contents from 60.0 g kg–1 to 815.7 g kg–1. A significant linear relationship was found between clay content and water content. The scaled mean absolute error (SMAE) of the clay estimation from the water content of air‐dried soil was 20% for the dataset using the Pipette method and 28% for the Sedigraph method. The estimation of the clay content was more accurate in fine‐textured than in coarse‐textured soils. In this study, organic‐C content played a subordinate role next to the clay content in explaining the variance of the water content. The water retention of coarse‐textured soils was more sensitive to the amount of organic C than that of fine‐textured soils. The results indicate that in our study the water content of air‐dried soil samples was a good quantitative proxy of clay contents, especially useful for fine‐textured soils. 相似文献

Aim of this study is to clinically, microbiologically and histopathologically characterize inflammatory bowel disease (IBD) in the cat. Nine cats with chronic persistent or intermitent vomitus and diarrhea were examined between 1998 and 2001. All cats had a thuorough diagnostic workup performed. Full thickness biopsies from stomach, duodenum, jejunum, ileum and colon were surgically obtained for histopathological examination. Duodenal juice was obtained by direct aspiration for microbiologic qualitative and semiquantitative examination. Seven cats euthanized for other medical reasons were used as controls. Six cats had a lymphoplasmacytic IBD and three an eosinophilic IBD. Four cats with IBD had additional diseases diagnosed. Three cats with IBD had elevated bacterial counts. Retrospectively no correlation could be found between clinical symptoms and histopathological results. Serum TLI was not able to differentiate chronic pancreatitis and IBD. Serum folic acid and cobalamin did not correlate with the distribution of lesions in the gastrointestinal tract. Finally, no correlation was found between bacterial counts in the proximal duodenum and IBD. 相似文献

Various Allium hybrids, obtained by the crossbreeding of Allium cepa (onion) as the mother plant and six taxonomically distant wild species obtained by embryo rescue, were investigated with special respect to their individual profiles of cysteine sulfoxides as well as enzymically and nonenzymically formed aroma substances. Alliinase (EC 4.4.1.4) catalyzes the conversion of odorless (+)-S-alk(en)yl-L-cysteine sulfoxides into volatile thiosulfinates. These thiosulfinates were converted to a variety of sulfides by steam distillation. SPME-gas chromatography (GC) and high-performance liquid chromatography (HPLC) used for the analysis of aroma components and their precursors permitted a high sample throughput, so that numerous gene bank accessions and Allium breeding materials were analyzed within a comparatively short time. Cysteine sulfoxides as well as alliinase activity were found in all investigated samples at different levels, but (+)-S-methyl-L-cysteine sulfoxide (methiin) was the most abundant sulfoxide present. (+)-S-(trans-1-Propenyl)-L-cysteine sulfoxide (isoalliin) is typical for onion and was found in all investigated hybrids. The pattern of the other cysteine sulfoxides depended strongly on the parent plants used. The profile of aroma components corresponded with the related pattern of aroma precursors (cysteine sulfoxides). Successful hybridization was proven by randomly amplified polymorphic DNA analysis. Together with these established marker techniques, HPLC and SPME-GC analysis provide support to breeding projects designed to improve the health and aroma properties of Allium hybrids. 相似文献
