The ability of PCR to detect infections of Theileria parva, the cause of East Coast Fever, in field-collected tick and bovine samples from Tanzania was evaluated. PCR-detected infection prevalence was high (15/20, 75%) in unfed adult Rhipicephalus appendiculatus ticks that fed as nymphs on an acutely-infected calf, but low (22/836, 2.6%) in unfed adult R. appendiculatus collected from field sites in Tanzania. Tick infection prevalence was comparable to that in previous studies that used salivary gland staining to detect T. parva infection in field-collected host-seeking ticks. Of 282 naturally-exposed zebu calves, seven had PCR-positive buffy coat samples prior to detection of Theileria spp. parasites in stained buffy coat cells or lymph node biopsies. Evidence of Theileria spp. infections was detected in stained smears of lymph node biopsies from 109 calves (38.6%) and buffy coat samples from 81 (28.7%), while buffy coat samples from 66 (23.4%) were PCR-positive for T. parva. Implications of these findings for the sensitivity and specificity of the PCR are discussed. 相似文献

We estimated the population density of dogs by distance sampling and assessed the potential utility of two marking methods for capture-mark-recapture applications following a mass canine rabies-vaccination campaign in Sorsogon Province, the Republic of the Philippines. Thirty villages selected to assess vaccine coverage and for dog surveys were visited 1 to 11 days after the vaccinating team. Measurements of the distance of dogs or groups of dogs from transect lines were obtained in 1088 instances (N = 1278 dogs; mean group SIZE = 1.2). Various functions modelling the probability of detection were fitted to a truncated distribution of distances of dogs from transect lines. A hazard rate model provided the best fit and an overall estimate of dog-population density of 468/km2 (95% confidence interval, 359 to 611). At vaccination, most dogs were marked with either a paint stick or a black plastic collar. Overall, 34.8% of 2167 and 28.5% of 2115 dogs could be accurately identified as wearing a collar or showing a paint mark; 49.1% of the dogs had either mark. Increasing time interval between vaccination-team visit and dog survey and increasing distance from transect line were inversely associated with the probability of observing a paint mark. Probability of observing a collar was positively associated with increasing estimated density of the dog population in a given village and with animals not associated with a house. The data indicate that distance sampling is a relatively simple and adaptable method for estimating dog-population density and is not prone to problems associated with meeting some model assumptions inherent to mark-recapture estimators. 相似文献

Abstract.— In freshwater crayfish aquaculture high densities can result from uncontrolled reproduction. Hence some farmers manually sex yabbies Cherax albidus and stock only mate crayfish in farm dams or ponds. Although male-only yabby populations produce an estimated 70% increase in gross income, hand sexing is time consuming and prone to error. A hybridization experiment using freshwater crayfish from the yabby species complex, collected from geographically isolated regions throughout Australia, revealed a hybrid between Cherax rotundus females and C. albidus males which consistently produces only male progeny. This has been confirmed in nine crosses of the same two species. The reciprocal cross of C. albidus females × C. rotundus males produces both male and female progeny. The consistent production of only male progeny by hybridization has not been recorded previously for a crustacean. 相似文献

Bees are believed to be in decline across many of the world’s ecosystems. Recent studies on British bumblebees proposed alternative theories to explain declines. One study suggested that greater dietary specialization among the rarer bumblebee species makes them more susceptible to decline. A second study disputed this theory and found that declines in British bumblebees were correlated with the size of species’ European ranges, leading to the suggestion that climate and habitat specialization may be better indicators of the risk of decline. Here we use a new and independent dataset based on Irish bumblebees to test the generality of these theories. We found that most of the same bumblebee species are declining across the British Isles, but that, within Ireland, a simple food-plant specialization model is inadequate to explain these declines. Furthermore, we found no evidence of a relationship between declines in Irish bumblebees and the size of species’ European ranges. However, we demonstrate that the late emerging species have declined in Ireland (and in Britain), and that these species show a statistically significant westward shift to the extremity of their range, probably as a result of changing land use. Irish data support the finding that rare and declining bumblebees are later nesting species, associated with open grassy habitats. We suggest that the widespread replacement of hay with silage in the agricultural landscape, which results in earlier and more frequent mowing and a reduction in late summer wildflowers, has played a major role in bumblebee declines. 相似文献

Farmers recorded the clinical signs of cows with clinical mastitis and submitted milk samples for bacteriological examination, so that the clinical signs could be correlated with the bacteriological findings. Odds ratios for the demeanour of the cow, the appearance of the milk, milk yield, udder texture, and the administration of parenteral antibiotics were calculated for mastitis cases classified in terms of their microbiology as either enterobacteriaceae, major Gram-positive pathogens, minor pathogens, 'no growths' or 'all other pathogens'. Animals infected with enterobacteriaceae had the highest odds of being reported as having a reduced milk yield, swollen or hard udders, watery milk and/or being systemically sick. A logistic regression model was used to predict the Gram-staining characteristics of the bacteria causing clinical mastitis. The clinical findings found to be significant predictors in the model were the demeanour of the cow and its milk yield. The regression model was used as a basis for a predictive test. Using a test data set, the sensitivity of the test was 28 per cent, its specificity was 96 per cent, the positive predictive value was 74 per cent and the negative predictive value was 80 per cent. Over-exploitation of whale sharks threatens the future of these wide-ranging pelagic fish. A long-term continuous record (4436 sightings) from a large aggregation (300-500 resident individuals) of whale sharks at Ningaloo Reef, Western Australia shows that mean shark length declined linearly by nearly 2.0 m and relative abundance measured from ecotourism sightings (corrected for variation in search effort and environmental stochasticity) has fallen by approximately 40% over the last decade. This population-level result confirms previous predictions of population decline based on projection models parameterised using mark-recapture estimates of survival. The majority of these changes are driven by reductions in the number of large individuals in the population. Phenomenological time series models support a deterministic (extrinsic) decline in large females, although there was some evidence for density dependence in large males. These reductions have occurred despite the total protection of whale sharks in Australian waters. As this species is highly migratory, the rapid change in population composition over a decade (<1 whale shark generation) supports the hypothesis of unsustainable mortality in other parts of their range (e.g., overfishing), rather than the alternative of long-term abiotic or biotic shifts in the environment. As such, effective conservation of whale sharks will require international protection, and collaborative tagging studies to identify and monitor migratory pathways. 相似文献

The influence of immature municipal solid waste-biosolids composts on emergence and mean days to emergence (MDE) of several weed species was evaluated in a pot trial under greenhouse conditions. The experiment consisted of placing a 7.5 cm deep layer of three-day-old immature compost, a mature and stable compost, an artificial medium or control sand as a mulch on ivyleaf morning glory seeds. Immature three-day-old compost decreased percentage emergence, shoot and root dry weight, and increased MDE of ivyleaf morning glory. In an experiment with eight-week-old immature compost utilizing mulching depths of 2.5, 5, 7.5, 10 cm and the untreated control on seeds of three weed species, common purslane did not emerge under any of the immature compost treatments. The MDE of ivyleaf morning glory and barnyard grass increased linearly as immature compost depths increased. Next, eight economically important weed species were sown in pots with either mature or immature (eight-week-old) compost utilizing mulching depths of 2.5 and 10 cm, in addition to an untreated control. Control pots yielded higher percentages of emergence than compost treatments for all species evaluated. Common purslane, large crabgrass, pig-weed, Florida beggarweed, and dichondra did not emerge through a 10-cm deep layer of mature compost mulch, or 2.5 or 10 cm deep layer of immature mulch. Significant compost maturity/depth interactions were observed for percent emergence on common purslane, ground cherry, large crabgrass, Florida beggarweed, and ivyleaf morning glory. A thinner layer was required to suppress germination using immature eight-week-old compost as compared to mature and stable compost. Immature (three-day or eight-week-old) compost containing acetic acid concentrations of 2474 and 1776 mg.kg?1 respectively reduced percentage emergence of several economically important weed species. These studies suggest that immature composts can be used to control weeds under conditions where spatial separation is maintained between the crop and the compost and phytotoxic fermentation products do not affect the health of the mulched plants and where odors associated with such partially stabilized products do not pose problems. 相似文献

Forest soil organic horizons from old and young plantations in the Alltcailleach Forest, N.E. Scotland were first sampled in 1949/50 and resampled in 1987. Copper, Pb and Zn in the soils were determined simultaneously on the original (stored) and the resampled soils. Overall mean Cu and Pb concentrations increased from 12.7 to 15.3 and 49.5 to 60.8 mg kg?1 between 1949/50 and 1987. Zinc concentrations decreased from 76.4 to 60.7 mg kg?1. Amounts of all 3 heavy metals increased because of increases in the thickness of organic horizons. Mean accumulation rates for Cu, Pb and Zn were 39.1, 186 and 114 g ha?1 yr?1, respectively. The rate of accumulation of Pb was significantly related to the rate of accumulation of organic matter. Copper and Zn concentrations were directly correlated with pH and inversely correlated with C/N ratio, whereas Pb was inversely correlated with pH and directly correlated with % C and C/N ratio. The more acidic soil organic horizons therefore contained higher concentrations of Pb and lower concentrations of Cu and Zn. 相似文献

The broad spectrum, systemic fungicides tebuconazole and propiconazole are used to control rust in boronia (Boronia megastigma Nees). Gas chromatography combined with either a benchtop quadrupole mass spectrometer or a high-resolution mass spectrometer allowed for the monitoring of both pesticides in boronia leaves, flowers, and concrete. Field trials were established at two sites to determine the rate of dissipation of tebuconazole and propiconazole in boronia. At site 1, two application rates of 125 and 250 g active ingredient/hectare (ai/ha) tebuconazole were employed. Treatments were repeated 17 days later. At harvest, 286 days after the final application, tebuconazole was detected at levels of 0.06 +/- 0.05 and 0.5 +/- 0.1 [mg/kg +/- standard error, on a dry matter basis (DMB)] in the leaves collected from plots treated with 125 and 250 g ai/ha of tebuconazole, respectively. The oil produced from the flowers collected at the final harvest had residues of tebuconazole at levels of 0.06 +/- 0.03 and 0.10 +/- 0.08 mg/kg for the 125 and 250 g ai/ha application rates, respectively. Two repeat applications of 125 g ai/ha propiconazole were also used at site 1. Residues of propiconazole were detected at 0.09 +/- 0.03 mg/kg (DMB) 286 days after the final application. At site 2, treatments of 125 g ai/ha of tebuconazole were applied twice. At harvest, 279 days after the final application of tebuconazole, residues were recorded at 0.30 +/- 0.09 mg/kg in the leaves (DMB) while the oil produced had 0.20 +/- 0.07 mg/kg. 相似文献
